Liver cancer has a high incidence and a poor prognosis, which seriously affects human health. Doxorubicin is one of the chemotherapeutics used in the treatment of tumours, but its severe adverse reactions, especially cardiac toxicity, have limited its clinical application. The nanometre drug delivery system enables drug-loaded nanoparticles to be specifically concentrated in tumour tissues, increasing cell uptake and improving curative effect. Therefore, in this paper, folic acid-modified mesoporous silica nanoparticles (MSN-NH2-PEG-FA) were synthesized by modifying the folic acid on the surface of a drug carrier by using the characteristics of the expression of folic acid receptors, and using it as a drug. The carrier was loaded with antitumor drug doxorubicin hydrochloride (DOX), and a nanometre drug delivery system (MSN-NH2-PEG-FA/DOX) was constructed. At the same time, the near-infrared dye Cy5 was used to mark the mother nucleus to construct fluorescent nanoparticles (MSN-NH2-PEG-FA/DOX-Cy5) for cell and tumour imaging, so as to obtain the abdominal image of liver cancer patients, thereby realizing diagnosis and treatment. The research results show that the carrier can specifically gather in the liver area, reduce the distribution in the heart, reduce the toxic and side effects of drugs, and prolong the survival time of patients. The results of this study provide new ideas for the treatment of liver cancer, and provide a new theoretical basis and experimental basis for the study of inorganic nanomaterials as targeted drug delivery systems.

Potential Strategy

Uniform Microbubbles (MBs) were routinely used as ultrasound contrast agents, but the MBs served for drug carrier showed well-marked advantage. In this study, we prepared a novel docetaxel–loaded microbubbles by carboxymethyl chitosan with W/O/W emulsion technique. Then, the surface morphology was characterized by scanning electron microscope (SEM). Ultrasound-triggered drug release experiments were performed with adjustable intensity and time. In the antitumor experiment in vitro, Ultrasound-assisted drug release from MBs exhibited good treatment of tumor. The results proved the combination of ultrasound and drug-loaded carboxymethyl chitosan microbubbles could be used as a potential strategy for anti-tumor therapy.

Herein, poly (n-(4-aminophenyl) methacrylamide)) carbon nano-onions (PAPMA-CNOs = f-CNOs) and γ-cyclodextrin/DOX-complex (CD) reinforced gelatin methacryloyl (GelMA)/f-CNOs/CD supramolecular hydrogel interfaces were fabricated using the photo-crosslinking technique. The physicochemical properties, morphology, biodegradation, and swelling properties of hydrogels were investigated. The composite hydrogels demonstrated enriched drug release under the acidic conditions (pH 4.5 = 99%, and pH 6.0 = 82%) over 18 days. Owing to the f-CNOs inclusion, GelMA/f-CNOs/CD supramolecular hydrogels presented augmented tensile strength (σult = 356.1 ± 3.4 MPa), toughness (K = 51.5 ± 0.24 Jg−1), and Young’s modulus (E = 41.8 ± 1.4 GPa). The strengthening of GelMA/f-CNOs/CD hydrogel systems indicates its good dispersion and the degree of polymer enveloping of f-CNOs within GelMA matrixes. Furthermore, the obtained hydrogels showed improved cell viability with human fibroblast cells. Nevertheless, the primed supramolecular hydrogels would pave the way for the controlled delivery systems for future drug delivery.

Magnetic targeted drugs delivery system (MTDDS) is a new targeted drug system, which can greatly reduce the dosage and improve the therapeutic efficiency of medicine. Currently superparamagnetic ferric oxide plays important function as targeted drug in the treatment of tumors, but cytotoxicity was still regarded as side effect in the process of drug. In this paper, we take advantage of drug carrier (ferric oxide) toxicity controlling cancer cell growth in cancer treatment, increasing targeted drug efficiency. We applied the modified chemical precipitation method to prepare polylactic acid (PLA) coated high-purity superparamagnetic Fe3O4 nanoparticles for targeted drug, characterized PLA/Fe3O4 microspheres physical and chemical properties, and then investigated cytotoxicity influence of PLA/Fe3O4 nanomagnetic microspheres as carrier for normal liver cells (7701) and liver cancer cells (HePG2) in different concentration; results of MTT and hemolysis and micronucleus test showed that carrier restrained the growth of HePG2 in special concentration, meanwhile the proliferation rate of liver cells was not affected. The study demonstrates that compared with liver cell, liver cancer cells (HepG2) are easy to be disturbed by PLA/Fe3O4 nanomagnetic microsphere, which have higher sensitivity and absorption ability. We hope to take advantage of the susceptible property of cancer cells for carriers to improve targeted drug function.
