This study puts forward an efficient method for protein detection in virtue of the tremendous fluorescence enhancement property of 6-aza-2-thio-thymine protected gold nanoclusters (ATT-AuNCs). In-depth studies of the protein-induced photoluminescence enhancement mechanism illustrate the mechanism of the interaction between ATT-AuNCs and protein. This new-established probe enables feasible and sensitive quantification of the concentrations of total protein in real samples, such as human serum, human plasma, milk, and cell extracts. The results of this proposed method are in good agreement with those determined by the classical bicinchoninic acid method (BCA method).

We have developed a very simple colorimetric sensor array by using only unmodified gold nanoparticles and NaCl salt for discrimination of multiple proteins. The inexpensive and convenient sensor array and the ubiquitous smartphone are coupled to achieve an immediate point-of-care diagnosis without additional devices.

Mutation and aggregation of superoxide dismutase (SOD) are reported as one of the causes of amyotrophic lateral sclerosis (ALS). To detect SOD1 protein from the motor neuron, surface plasmon resonance (SPR) analysis was adopted due to its advantages in the in-situ biomolecular recognition by surface analysis without labeling. For the patterning of protein antigens at Au surface for use in SPR imaging experiments, microfluidic devices were fabricated with polydimethylsiloxane (PDMS) by replica molding method. They were designed for the solution to flow by capillary force only without using any additional pumping equipments or flow controllers. Performance of microfluidic devices was verified by the simple microfluidic experiments, and multiple protein-patterned sensor surfaces were constructed by using these microfluidic devices.
