Evaluation of Species Invasiveness: A Case Study with Acacia dealbata Link. on the Slopes of Cabeça (Seia-Portugal)

One of the main causes of biodiversity loss in the world is the uncontrolled expansion of invasive plants. According to the edaphoclimatic conditions of each region, plants acquire different invasion behaviors. Thus, to better understand the expansion of invasive plants with radial growth, it is proposed to use two equations, the Annual Linear Increment (ALI) and the Annual Invasiveness Rate (AIR). These equations are applied using spatiotemporal data obtained from the analysis of orthophotomaps referring populations of Acacia dealbata Link. in areas located in Serra da Estrela, Portugal. As a result, the area occupied by this species in the parish of Cabeça was evaluated and a 20-year projection was carried out. The data produced by these equations contributed to improving the knowledge about the invasion behavior of exotic species in a rigorous and detailed way according to local ecological conditions. This study may serve as the basis for the application of other similar situations concerning invasive species in other territories, to improve the efficiency of future projections for these species. Local technical and scientific knowledge will contribute to improving spatial and management planning, enabling a better adequacy and effectiveness of the control measures to be adopted.

Tumor-derived exosomal components: the multifaceted roles and mechanisms in breast cancer metastasis

Breast cancer (BC) is the most frequently invasive malignancy and the leading cause of tumor-related mortality among women worldwide. Cancer metastasis is a complex, multistage process, which eventually causes tumor cells to colonize and grow at the metastatic site. Distant organ metastases are the major obstacles to the management of advanced BC patients. Notably, exosomes are defined as specialized membrane-enclosed extracellular vesicles with specific biomarkers, which are found in a wide variety of body fluids. Recent studies have demonstrated that exosomes are essential mediators in shaping the tumor microenvironment and BC metastasis. The transferred tumor-derived exosomes modify the capability of invasive behavior and organ-specific metastasis in recipient cells. BC exosomal components, mainly including noncoding RNAs (ncRNAs), proteins, lipids, are the most investigated components in BC metastasis. In this review, we have emphasized the multifaceted roles and mechanisms of tumor-derived exosomes in BC metastasis based on these important components. The underlying mechanisms mainly include the invasion behavior change, tumor vascularization, the disruption of the vascular barrier, and the colonization of the targeted organ. Understanding the significance of tumor-derived exosomal components in BC metastasis is critical for yielding novel routes of BC intervention.

Modeling Invasion of Campylobacter jejuni into Human Small Intestinal Epithelial-Like Cells by Bayesian Inference

ABSTRACT Current approaches used for dose-response modeling of low-dose exposures of pathogens rely on assumptions and extrapolations. These models are important for quantitative microbial risk assessment of food. A mechanistic framework has been advocated as an alternative approach for evaluating dose-response relationships. The objectives of this study were to investigate the invasion behavior of Campylobacter jejuni, which could arise as a foodborne illness even if there are low counts of pathogens, into Caco-2 cells as a model of intestinal cells and to develop a mathematical model for invading cell counts to reveal a part of the infection dose-response mechanism. Monolayer-cultured Caco-2 cells and various concentrations of C. jejuni in culture were cocultured for up to 12 h. The numbers of C. jejuni bacteria invading Caco-2 cells were determined after coculture for different time periods. There appeared to be a maximum limit to the invading bacterial counts, which showed an asymptotic exponential increase. The invading bacterial counts were higher with higher exposure concentrations (maximum, 5.0 log CFU/cm2) than with lower exposure concentrations (minimum, 0.6 log CFU/cm2). In contrast, the ratio of invading bacteria (number of invading bacteria divided by the total number of bacteria exposed) showed a similar trend regardless of the exposure concentration. Invasion of C. jejuni into intestinal cells was successfully demonstrated and described by the developed differential equation model with Bayesian inference. The model accuracy showed that the 99% prediction band covered more than 97% of the observed values. These findings provide important information on mechanistic pathogen dose-response relationships and an alternative approach for dose-response modeling. IMPORTANCE One of the infection processes of C. jejuni, the invasion behavior of the bacteria in intestinal epithelial cells, was revealed, and a mathematical model for prediction of the cell-invading pathogen counts was developed for the purpose of providing part of a dose-response model for C. jejuni based on the infection mechanism. The developed predictive model showed a high accuracy of more than 97% and successfully described the C. jejuni invading counts. The bacterial invasion predictive model of this study will be essential for the development of a dose-response model for C. jejuni based on the infection mechanism.

Effect of contact stress on the tribology behaviors of PTFE/316L seal pairs under various abrasive-contained conditions

This study focuses on the tribological characteristics of polytetrafluoroethylene rubbing against 316L stainless steel in the presence of various Al2O3 abrasive particles. The ranges of the contact stresses and abrasive sizes were 2.50–5.01 MPa and 2.0–230 µm, respectively. The evolution of the coefficient of friction, abrasive invasion behavior and damage characteristics on worn surfaces, and the transformation of damage mechanisms under different contact stresses are summarized. Results demonstrate that contact stress exerts a considerable effect on abrasive invasion behavior, the coefficient of friction trends become complicated at low contact stress. With decreasing particle sizes under a contact stress of 2.50 MPa, the wear failure changes from two-body wear to three-body wear, or two wear behaviors exist simultaneously. As contact stress increases, the wear mechanism is gradually simplified. The particle size threshold that causes the change of the wear mechanism is also gradually reduced. The wear rate of polytetrafluoroethylene has no linear relationship with abrasive size, whereas wear increases with contact stress. Under 2.50 MPa conditions, the particles easily invade the tribo-interface in the friction process, thus increasing the wear rate of the metallic counterpart. In addition, with increases contact stress, the polytetrafluoroethylene material transfers to the counterface, and local adhesion occurs. When the contact stress is increased to 5.01 MPa, polytetrafluoroethylene even shows an obvious creep.
