EFFECTS OF INGESTED ETHANOL ON ADULT DROSOPHILA MELANOGASTER (DIPTERA: DROSOPHILIDAE)

AbstractAn apparatus consisting of a plastic box and a feeding circuit permits the calculation of μl volumes of ethanol ingested by Drosophila melanogaster Meigen. The net amount of ethanol taken (expressed in 100% ethanol) has been examined in various experimental conditions: sucrose solutions, ethanol–sucrose solutions at various sucrose and alcohol concentrations, ethanol solutions - cube of sucrose and ethanol solutions alone. The data obtained on ingestion by Drosophila show that the consumption of sucrose solutions was inversely proportional to the concentrations of sucrose. The amount of ethanol ingested during 14 days was directly related to the concentration of ethanol in the diets although in general the total volume of solution consumed did not vary significantly in most cases. The analyses of observations of 14 days showed that decreased mortality was obtained with the rise in the concentration of sucrose in sucrose-water solutions in 0.5–3% range and the mortality increased with the rise in the concentration of ethanol in the diets, yet ethanol-sucrose was better than cube of sugar alone.

The effects of various experimental conditions on locomotor activity of adult Drosophila melanogaster flies of both sexes were measured in the locometer. This apparatus measures linear progression of flies and is based on the fractionating principle of mazes used for behavioural studies. Groups of approximately 100 flies are tested simultaneously but every individual fly is assigned a locomotor activity score. This score is positively correlated with open-field behaviour as measured in the Elens-Wattiaux-chamber. Strong lasting effects of ether anaesthesia were demonstrated. Other anaesthetics (c.q. CO 2 and N 2 ) did not show such profound effects. The effects of culturing conditions, density, age and sex were measured. The locometer proved to be useful for the investigations of the effects of different stimuli on the locomotory behaviour of fruitflies.

Reaction of cerium ammonium nitrate and tetrafluoroterephthalic acid in water afforded two new metal-organic frameworks with UiO-66 [F4_UiO-66(Ce)] and MIL-140 [F4_MIL-140A(Ce)] topologies. The two compounds can be obtained in the same experimental conditions, just by varying the amount of acetic acid used as crystallization modulator in the synthesis. Both F4_UiO-66(Ce) and F4_MIL-140A(Ce) feature pores with size < 8 Å, which classifies them as ultramicroporous. Combination of X-ray photoelectron spectroscopy and magnetic susceptibility measurements revealed that both compounds contain a small amount of Ce(III), which is preferentially accumulated near the surface of the crystallites. The CO<sub>2</sub> sorption properties of F4_UiO-66(Ce) and F4_MIL-140A(Ce) were investigated, finding that they perform better than their Zr-based analogues. F4_MIL-140A(Ce) displays an unusual S-shaped isotherm with steep uptake increase at pressure < 0.2 bar at 298 K. This makes F4_MIL-140A(Ce) exceptionally selective for CO<sub>2</sub> over N<sub>2</sub>: the calculated selectivity, according to the ideal adsorbed solution theory for a 0.15:0.85 mixture at 1 bar and 293 K, is higher than 1900, amongst the highest ever reported for metal-organic frameworks. The calculated isosteric heat of CO<sub>2 </sub>adsorption is in the range of 38-40 kJ mol<sup>-1</sup>, indicating a strong physisorptive character.

A series of solid-phase syntheses of the protected precursor II of DDAVP was carried out. Experimental conditions were developed under which practically pure II can reproducibly be obtained in yields better than 60%. The protected precursors of DDAVP obtained by liquid- and solid-phase synthesis and DDAVP samples obtained from these precursors were undistinguishable by conventional analytical or pharmacological assays.

The laminopathy Hutchinson–Gilford progeria syndrome (HGPS) is caused by the mutant lamin A protein progerin and leads to premature aging of affected children. Despite numerous cell biological and biochemical insights into the basis for the cellular abnormalities seen in HGPS, the mechanism linking progerin to the organismal phenotype is not fully understood. To begin to address the mechanism behind HGPS using Drosophila melanogaster, we have ectopically expressed progerin and lamin A. We found that ectopic progerin and lamin A phenocopy several effects of laminopathies in developing and adult Drosophila, but that progerin causes a stronger phenotype than wild-type lamin A.

The effects of intracerebroventricular application of melanin-concentrating hormone (MCH) on licking for sucrose, quinine hydrochloride (QHCl), and water solutions were evaluated in two experiments. In experiment 1, rats received 90-min access to sucrose and water solutions after MCH or vehicle microinjection to the third ventricle (3V). MCH increased intake largely through increases in the rate of licking early in the meal and in the mean duration of lick bursts, suggesting an effect on gustatory evaluation. Therefore, in experiment 2, brief access tests were used with a series of sucrose and QHCl concentrations to behaviorally isolate the effects of intracerebroventricular MCH on gustatory evaluation. MCH uniformly increased licking for all sucrose solutions, water, and weak concentrations of QHCl; however, it had no effect on licking for the strongest concentrations of QHCl, which were generally avoided under control conditions. Thus MCH did not produce nonspecific increases in oromotor activity, nor did it change the perceived intensity of the tastants. We conclude that MCH enhanced the gain of responses to normally accepted stimuli at a phase of processing after initial gustatory detection and after the decision to accept or reject the taste stimulus. A comparison of 3V NPY and MCH effects on licking microstructure indicated that these two peptides increased intake via dichotomous behavioral processes; although NPY suppressed measures associated with inhibitory feedback from the gut, MCH appeared instead to enhance measures associated with hedonic taste evaluation.

ABSTRACT Polymorphism for both alleles of a gene ref(2)P, which is a usual trait of French natural populations of Drosophila melanogaster, can be reproduced in experimental conditions. ref(2)P is a gene for resistance to the hereditary, noncontagious Rhabdovirus α, responsible for CO2 sensitivity in Drosophila melanogaster. The equilibrium frequencies observed in cages are the same as in the wild, whether α virus is present or not. The rapid rate of return to these equilibrium frequencies indicates that strong forces, which remain to be determined, are responsible for the maintenance of this polymorphism.
