Yeast diversity in the pia and small-intestinal epithelium of Pon Yang Kham fattening cattle in Thailand was studied using a culture-dependent method. A total of 701 yeasts were isolated from the pia of the duodenum, jejunum, and ileum of the small intestine, while 425 isolates were obtained from the epithelium of all three parts of the small intestine. Yeast identification was performed and ascomycetous yeasts were found at levels of 96.9% and 86.8% in the pia and small intestine, respectively, whereas basidiomycetous yeasts were found at levels of 2.3% and 12.7%. Candida parapsilosis was the species with the highest occurrence in the duodenal and jejunal pia, with an 83.3% and 77.8% frequency of occurrence (FO), respectively. Both C. parapsilosis and C. tropicalis were species with the highest occurrence in the ileum, with a 61.1% FO. Moreover, C. parapsilosis was the species with the highest occurrence in the epithelium of the duodenum, jejunum, and ileum, with FOs of 88.2%, 87.5%, and 87.2%, respectively. Principal coordinate analysis revealed no marked differences in yeast communities from either the pia or epithelium of all three parts of the small intestine. An estimation of the expected richness of the species showed that the observed species richness was lower than the predicted richness.

1. The object was toinvestigate the importance of urea a source of ammonia in the large intestine of herbivores. Urea was present in small intestinal contents of slaughtered horses in concentrations similar to those in blood but, in the small intestine of salughtered sheep, the urea was less than in blooc.2. There was little ammonia in small intestinal contents of slaughtered horses but consider-able ammonia was present in small intestinal contents of slaughtered sheep. The ammonia in small intestinal contents of the slaughtered sheep was probably formed from urea, as ileal con-tents taken from a sheep with an ileal cannula contained considerable urea and little ammonia.3. The ammonia concentration in caecal contents of sheep was related to the concentration of urea in blood except when ileal contents were prevented from entering the caecum.4. Ileal digesta of sheep contained more free amino nitrogen than did caecal digesta.5. Ammonia was absorbed more rapidly than water from the caecum of sheep. The rate of absorption was related to the concentration of ammonia in the caecum.

1. Semi-synthetic diets containing 200 g protein/kg were meal-fed for 1.5 h to groups of rats. The contents of the whole small intestinal tract were collected and the amount of soluble calcium was determined.2. In the rats given 200 g casein/kg diet, formation of a fraction containing macrophosphopeptide in the small intestine was confirmed by gelfiltration of the intestinal contents on Sephadex G-25. However, this macrophosphopeptide fraction was not found when casein alone was fed.3. In the intestinal contents at 2.5 h after ingestion, the amounts of both soluble Ca and phosphorus were significantly higher in rats fed the casein diet than in those fed diets containing egg albumin, isolated soyabean protein or an amino acid mixture. However, the amount of insoluble Ca was least in rats fed the casein diet.

1. Maltase sucrase, palatinase (the enzyme that hydrolyses palatinose, i.e. 6-o-α-D-gluco-pyranosyl-D-fructose) and lactase activities were measured in the small and large intestines of germ-free and conventional chicks given either a diet of purified ingredients or a practical chick mash.2. With the purified diet there were no differences in body-weight or small intestinal disaccharidase activities between germ-free and conventional chicks. With the chick mash the germ-free birds were heavier and had higher total amounts of maltase, sucrase and palatinase activities in the small intestine than did their conventional controls. When disaccharidase activities were expressed in terms of body-weight there were no differences between birds in the two environments. Enzyme activities were consistently higher in the birds given chick mash.3. Inclusion of milled fibre in the purified diet did not increase the weight or disaccharidase activities of the small intestine in either environment.4. Lactase was virtually absent from the small intestine of birds in both environments and from the large intestine of germ-free birds. There was appreciable lactase activity in the large intestinal contents of conventional chicks, and it was increased by inclusion of lactose in the diet.5. When lactose was the sole source of carbohydrate the birds grew poorly but mortality rate was less among conventional compared with germ-free chicks.6. It was concluded that the presence of micro-organisms has no direct effect on disaccharidase production in the small intestine of the chick. Microbial lactase is present in the large intestine, and at least some of the products of its action can be utilized by the bird.

Unipolar recordings of small intestinal potentials were obtained from chronic dog preparations with exteriorized segments of bowel, from anesthetized dogs, cats and rabbits at laparotomy and from unanesthetized human subjects. Two dominant types of electrical activity were observed and correlated with function. The first type consisted of brief, nonpropagated, spike-like negative potentials which occurred only immediately preceding and during contraction and varied in frequency of occurrence proportionate to intensity of contraction. It is concluded that they represent the action potentials of contracting muscle fibers. The second type of electrical activity consisted of a recurring complex of slow potential changes, the configuration of which varied with the region of intestine and type of preparation. The complex was most stable and most clearly defined in transplanted segments of dog jejunum. The form of the complex was not altered by anesthetics or by sympathomimetic or parasympathomimetic drugs, and was the same during inactivity as during contractions. Initiation of contractions occurred only during the relatively positive phase of the slow complex. It is concluded that the slow complex is a cyclic alteration of the resting potential of the smooth muscle cells of the intestine which originates more or less synchronously in adjacent cells and is associated with a mechanism which renders the muscle alternately relatively excitable and absolutely refractory.

Although high-fat diet (HFD)-related dysbiosis is involved in the development of steatohepatitis, its pathophysiology especially in the small intestine remains unclear. We comprehensively investigated not only the liver pathology but also the microbiome profile, mucosal integrity and luminal environment in the small intestine of mice with HFD-induced obesity. C57BL/6J mice were fed either a normal diet or an HFD, and their small-intestinal contents were subjected to microbial 16S rDNA analysis. Intestinal mucosal permeability was evaluated by FITC-dextran assay. The levels of bile acids in the small-intestinal contents were measured by liquid chromatography/mass spectrometry. The expression of tight junction molecules, antimicrobial peptides, lipopolysaccharide and macrophage marker F4/80 in the small intestine and/or liver was examined by real-time RT-PCR and immunohistochemistry. The abundance of Lactobacillus was markedly increased and that of Clostridium was drastically decreased in the small intestine of mice fed the HFD. The level of conjugated taurocholic acid was significantly increased and those of deconjugated cholic acid/secondary bile acids were conversely decreased in the small-intestinal contents. The expression of occludin, antimicrobial Reg IIIβ/γ and IL-22 was significantly decreased in the small intestine of HFD-fed mice, and the intestinal permeability was significantly accelerated. Infiltration of lipopolysaccharide was significantly increased in not only the small-intestinal mucosa but also the liver of HFD-fed mice, and fat drops were apparently accumulated in the liver. Pathophysiological alteration of the luminal environment in the small intestine resulting from a HFD is closely associated with minimal inflammation involving the gut-liver axis through disturbance of small-intestinal mucosal integrity.

The high contribution of postruminal starch digestion (>50%) to total tract starch digestion on certain energy dense diets (Mills et al. 1999) demands that limitations to small intestinal starch digestion are identified. Therefore, a dynamic mechanistic model of the small intestine was constructed and evaluated against published experimental data for abomasal carbohydrate infusions in the dairy cow. The mechanistic structure of the model allowed the current biological knowledge to be integrated into a system capable of identifying restrictions to dietary energy recovery from postruminal starch delivery.

The effect of an intestinal microflora consisting of selected microbial species on myoelectric activity of small intestine was studied using germ-free rat models, with recording before and after specific intestinal colonization, in the unanesthetized state. Intestinal transit, neuropeptides in blood (RIA), and neuromessengers in the intestinal wall were determined. Clostridium tabificum vp 04 promoted regular spike burst activity, shown by a reduction of the migrating myoelectric complex (MMC) period from 30.5 ± 3.9 min in the germ-free state to 21.2 ± 0.14 min ( P < 0.01). Lactobacillus acidophilus A10 and Bifidobacterium bifidum B11 reduced the MMC period from 27.9 ± 4.5 to 21.5 ± 2.1 min ( P < 0.02) and accelerated small intestinal transit ( P < 0.05). Micrococcus luteus showed an inhibitory effect, with an MMC period of 35.9 ± 9.3 min compared with 27.7 ± 6.3 min in germ-free rats ( P < 0.01). Inhibition was indicated also for Escherichia coli X7gnotobiotic rats. No consistent changes in slow wave frequency were observed. The concentration of neuropeptide Y in blood decreased after introduction of conventional intestinal microflora, suggesting reduced inhibitory control. Intestinal bacteria promote or suppress the initiation and aboral migration of the MMC depending on the species involved. Bacteria with primitive fermenting metabolism (anaerobes) emerge as important promoters of regular spike burst activity in small intestine.

Objectives The aim of the study was to evaluate ultrasonographic changes in the small intestine of cats with clinical signs of gastrointestinal disease and low or low–normal serum cobalamin concentrations. Methods Records for client-owned cats presenting to the small animal hospital with signs of gastrointestinal disease and in which serum cobalamin concentrations were measured from 2000–2013 were reviewed. Inclusion criteria were cobalamin concentrations <500 ng/l, abdominal ultrasound within 1 month of cobalamin testing and definitive diagnosis. Results Of 751 serum cobalamin measurements, hypocobalaminemia or low–normal cobalamin was identified in 270 cats, abdominal ultrasound was performed in 207 of those cats and a diagnosis was available for 75 of them. Small intestinal ultrasound changes were detected in 49/75 (65%) cats. Abnormalities included thickening, loss of wall layer definition, echogenicity alterations and discrete masses. Serum cobalamin concentrations <500 ng/l were observed with diagnoses of inflammatory disease, neoplasia, infectious disease and normal histopathology. Cobalamin concentration was significantly lower in cats with lymphoma or inflammatory bowel disease compared with other gastrointestinal neoplasia ( P = 0.031). No difference was found between cobalamin concentration and the presence of ultrasound abnormalities, specific ultrasound changes or albumin concentration. Conclusions and relevance One-third of symptomatic cats with hypocobalaminemia or low–normal cobalamin concentrations may have an ultrasonographically normal small intestine. For the majority of cats in this study, histopathologic abnormalities were observed in the small intestine, regardless of ultrasound changes. These findings suggest gastrointestinal disease should not be excluded based on low–normal cobalamin concentrations, even with a concurrent normal ultrasound examination. Additional studies are needed in cats with low–normal serum cobalamin concentrations, as a definitive diagnosis was not pursued consistently in those cats. However, data from this study suggest that careful monitoring, histopathologic evaluation and future cobalamin supplementation may be warranted.
