An efficient and reproducible regeneration protocol via direct organogenesis has been established using nodal explants of Coscinium fenestratum. Nodal explants were inoculated on MS medium supplemented with BAP and TDZ at 2.0 and 1.0mg/L respectively were produced high frequency of shoots (91.01%). The elongated shoots subcultured on half strength MS medium supplemented with IBA alone at 0.6mg/L produced high frequency of roots (97.42%). Rooted plantlets were acclimatized and established in garden soil, sand and vermicompost in the ratio 1:1:1 with good survival rate under greenhouse conditions. This protocol can be efficiently used for mass multiplication and conservation of this critically endangered medicinal liana in its native habitat.

Shoot tips and nodal explants from in vitro growing seedlings of Ficus glomerata Roxb. (Moraceae). showed best shoot induction (88%) on MS medium supplemented with 0.5 mg/l BAP, where maximum number of shoots were produced per culture. In vitro raised shoots rooted well on half strength of MS medium with 2.0 mg/l IBA + 0.1 mg/l NAA. The survival rate of regenerated plantlets was 82%. Shoot multiplication of M. brunoniana Wight & Arn. was achieved from the nodal explants of mature plants using MS with different concentrations and combina-tions of growth regulators. Maximum explant response and highest number of shoots per explant was obtained on MS medium fortified with 1.0 mg/l BAP. The highest degree of shoot proliferation was found to be 90%. The combination of BAP and Kn was also found to be effective for regeneration. The regenerated shoots were successfully rooted on MS supplemented with 0.5 mg/l NAA, after sequential hardening; survival rate was 90%.  Key words: Marsdenia brunoniana, Medinal plant, Micropropagation, Conservation D. An efficient protocol was developed for shoot proliferation and plant regeneration of Phlogacanthus thyrsiflorus Nees. (Acanthaceae) - a rare medicinal shrub of Bangladesh, through in vitro culture using shoot tip and nodal explants. Best shoot induction was observed on MS with 1.0 mg/l BAP + 0.5 mg/l NAA, in which 84.2% of nodal explants responded to produce maximum number (12.4 ± 0.66) of shoots per culture. In vitro raised shoots rooted on half-strength MS with 0.5 mg/l IBA + 0.5 mg/l NAA. For acclimation and transplantation, the plantlets in the rooting culture tubes were kept in normal room temperature for 7 days before transplanting in pots where plantlets were reared for three weeks. Breeding work carried out during the period 1971–85 by the Coffee Research Institute, Ruiru, Kenya resulted in the release of a new improved hybrid Coffea arabica named Ruiru 11. The cultivar combines resistance to coffee berry disease (CBD) and leaf rust, with high yield and good cup quality attributes. The propagation by F1 hybrid seeds production, cuttings, and tip grafting do not produce enough planting materials. There was a need to explore alternative methods and tissue culture offers potential options. The objective of the study was to evaluate the effect of explant sources and cytokinins on induction and regeneration of somatic embryos. Eight different explants were cultured on half-strength Murashige and Skoog (MS) medium supplemented with 10 µm benzylaminopurine (BAP). The effect of kinetin, N6-(2-isopentyl) adenine (2iP) evaluated at (0, 0.5, 5, or 25 µm) or thidiazuron (TDZ) (0, 0.5, 1.0, or 5 µm) added in separate experiments was also evaluated. The percentage of embryogenic cultures and the numbers of embryos per explant were determined after 3 months’ culture. The explant type had a significant effect (P > 0.05) on the induction of somatic embryos. Explants from in vitro-germinated seedlings produced the highest embryogenic cultures (90%) and the highest mean number of embryos (19.36) per explant. Cytokinins strongly enhanced induction and regeneration of somatic embryos. TDZ at 1 µm produced the highest embryogenic cultures (100%) and the highest mean number of embryos (24.2). The embryos were germinated on half-strength MS medium without any hormones. A high (98%) survival rate of the regenerated plantlets was recorded over all the treatments in the greenhouse. This is the first report on induction of high-frequency direct somatic embryos from coffee juvenile tissues. This is of great significance in tissue culture and indeed molecular biology manipulations because it allows regeneration of coffee from several explants.

An efficient plant regeneration protocol was developed for Justicia gendarussa Burm.f. (Acanthaceae), an important medicinal shrub. Nodal segments grown on Murashige and Skoog (MS) medium containing 1.0 mg/l Benzyl adenine (BAP) with 10 % coconut milk showed better growth response and produced 10.5 ± 0.6 shoots per explant with an average length of 4.4 ± 0.3 cm after 35 days. Rooting of shoots was achieved on growth regulator free half strength MS medium produced 5.3 ± 0.25 cm roots with an average height of 4.8 ± 0.2 cm after 30 days. The rooted plantlets were transferred for hardening, 80 % of plants were successfully established in the field. Key words: Justicia gendarussa; Nodal segments; Regeneration; Coconut milk. In vitro clonal propagation of Clerodendrum serratum L., a rare medicinal plant has been reported by using LM medium supplemented with different growth hormones. The maximum number of shoots with maximum length were obtained from stem derived callus on LM media fortified with 1.5 mg/l BAP and 0.3 mg/l NAA. Nodal explants showed direct organogenesis on LM media containing BAP (0.5 mg/l) alone. The regenerated shoots were successfully rooted with maximum frequency (100%) on half strength LM media supplemented with 0.5 mg/l NAA. The well rooted microshoots were successfully transferred to hardening and survival rate was 88%. A protocol for multiple shoot induction and plant regeneration from nodal explants of Ocimum citriodorum has been developed. Nodal explants inoculated on Murashige and Skoog (MS) medium supplemented with 1.0 mg/l Benzyl adenine (BAP) and 0.025 mg/l indole -3- acetic acid (IAA) showed better growth response (80%) and produced 15.2 ± 1.28 shoots per explant with an average length of 6.17 ± 0.29 cm after 35 days. Roots were induced after transfer to half strength MS medium supplemented with 0.5 mg/l Indole -3- butyric acid (IBA) produced 6.0 ± 1.0 roots with an average height of 4.9 ± 0.26 cm after 30 days. Plantlets with well developed root and shoot systems were successfully acclimated (80 %) and established in earthen pots containing mixture of soil, vermiculite and farm yard manure (1:1:1). Abstract The aim of this study is to develop and improve methods of in vitro propagation of representatives of Dactylorhiza: D.baltica , D. fuchsii. For the study, we used protocorms obtained by the asymbiotic germination of seed during 90 days. It has been established that half-strength of Murashige and Skoog (1962) medium (½ MS) supplemented with 1-2 mg/l 6-Benzylaminopurine(6-BAP), potato puree (20g/l), and charcoal (1g/l) effectively influenced the development of protocorms, and seedlings formation in the studied species. The result of the study showed that the survival rate of protocorms was high in all experimental culture media, but in D. fuchsii it was better at a concentration 2mg/l of 6-BAP (95.4%), while in D. baltica it was high at 1mg/l (87.0%). The highest percentage of multiple protocorms (68%) and the formation of new secondary protocorms in D. fuchsii (5,5±0,3 units) were observed on a culture medium containing 2 mg/l 6-BAP. The highest percent of rooting of D. fuchsii protosoms (78%) and length of roots (0.9cm) observed in ½ MS medium without growth regulators. During the development of D. baltica protosoms, the culture medium of ½ MS containing 1 mg/l 6-BAP had the best effect on the number of roots (1.8±0.1root/protosom), while the medium supplemented with 2mg/l of 6-BAP contributed to the formation of a larger number of new secondary protocorms (3,2±0,1protocorm/unit). During the subsequent cultivation of protosoms of D. baltica on a culture medium containing 1 mg/l it was observed an increase in the height of shoots (4,8±0,3 см), and the length of roots (2,2±0,1 см), wherein the number of newly formed protocorms was higher by 30% on the medium supplemented with 2 mg/l 6-BAP. In vitro regenerative potential of protocorm like bodies (PLBs) of Dendrobium barbatulum were assessed. The formation of secondary PLBs from the primary PLBs that formed from the callus, it is derived from a few larger injured protocorm like bodies. The injured larger PLBs were cultured on half strength MS supplemented with different concentrations of BAP (0.32, 1.62, 3.23 and 16.15 μM), Zn (0.46, 2.28, 4.56 and 22.80 μM) and Kn (0.47, 2.33, 4.65 and 23.25 μM). The highest percentage of secondary PLBs (66.33%) were obtained on half strength MS medium supplemented with BAP 3.23 μM, after 5 weeks of culture. Then these clumps of PLBs were subcultured on half strength MS fortified with coconut water (CW), cane juice (CJ), peptone (P) and casein hydrolysate (CH) for regeneration. Among the complex organic additives 20% CW and 2.0 g/l P to half strength MS resulted in the development of plantlet 78.31 and 62.60%, respectively. Well developed plantlets were successfully acclimatized in the community pots having brick pieces, charcoal, decaying litter and coconut husk (1 : 1 : 1 : 1) gave maximum survival rate of 87.03%. <p>Bauhinia purpurea L. is a leguminous plant moderate sized tree with multipurpose value. It is distributed in sub-Himalayan tracts. It has been cultivated in the plain region up to the elevation of 1350 m. Mature seeds of Bauhinia purpurea L. were cultured on half strength Murashige and Skoog (1962) (MS) medium. Nodal explants obtained from germinated seedlings were cultured on MS medium containing 0.5 M BAP produced multiple shoots which were used for experimental purposes. Nodal explants obtained from cultured were subcultured on different concentrations of N-Benzyl -9-(2-tetrahydropyranyl) (BPA) and Indole-3acetic acid (IAA). The best proliferation of nodes and shoots were observed on the MS medium supplemented with 0.5 M BPA and 0.1 M IAA. After 8 weeks of culture, the propagated plants were acclimatized and transferred to the sand box containing 1:1 soil and sand. Well rooted plants were then established in the field. All the data collected were worked out statistically with SPSS, a system of analytical procedure.</p>
