Blood culture is currently the most commonly used method for diagnosing sepsis and bloodstream infections. However, the long turn-around-time to achieve microbe identification remains a major concern for clinical microbiology laboratories. Gram staining for preliminary identification remains the gold standard. We developed a new rapid strategy using a tabletop scanning electron microscope (SEM) and compared its performance with Gram staining for the detection of micro-organisms and preliminary identification directly from blood cultures. We first optimised the sample preparation for twelve samples simultaneously, saving time on imaging. In this work, SEM proved its ability to identify bacteria and yeasts in morphotypes up to the genus level in some cases. We blindly tested 1075 blood cultures and compared our results to the Gram staining preliminary identification, with MALDI-TOF/MS as a reference. This method presents major advantages such as a fast microbe identification, within an hour of the blood culture being detected positive, low preparation costs, and data traceability. This SEM identification strategy can be developed into an automated assay from the sample preparation, micrograph acquisition, and identification process. This strategy could revolutionise urgent microbiological diagnosis of infectious diseases.

A preparation of nanometer silver sol by micro arc discharge has been study here through the reduction of Ag3PO4. Sodium citrate and polyvinylpyrrolidone were added respectively into the electrolyte as stabilizer. The results show that, the Ag3PO4 concentration, stabilizer type and concentration have great impacts on the formation of the nanometer silver sol. By means of UV-VIS extinction spectrophotometer, scanning electron microscope and energy dispersive spectrometer, it is found that the solid powder extracted from the electrolyte solution after discharge is the aggregation of the silver formed in the solution and their original size maybe less than 20nm. Nanometer silver with smaller size and narrower size distribution can be obtained with sodium citrate as stabilizer than with polyvinylpyrrolidone. But the latter has higher conversion rate. From this experiment, we found that micro arc discharge can be a rapid, stable preparation method of nanometer silver sol.

Introduced various preparation method of one dimensional nanowire and the merit and shortcoming of various methods, adopt two anodic oxidation method carry on oxadation to the aluminum sheet and Fabricate thickness about 15 ums, 50 nms nanohole diameter AAO template .In the H2PtCl6·6H2O+H3BO4 solutions carry on electrochemistry deposition to the AAO template.The deposited templates were characterized by scanning electron microscope (SEM ) , approval that growth speed of nanowires are the 1 um/h.After had electrochemistry deposited for 12h, the nanowire can pierce through AAO template.The length of nanowire is 15 ums, diameter is 50 nms. Nanowires are independent, they meet demand as conducting nanowires.

This modification to the technique of epi-illumination light microscopy makes use of a new system of lenses that replaces expensive and not readily available dipping cone objectives. The newer objectives offer at least comparable resolution and depth of field, along with simple preparation procedures. An epi-illumination system is a good intermediate between the stereo microscope and a scanning electron microscope, offering magnification at high power that can aid in evaluation of potential scanning electron microscope specimens, as well as the time- and material-saving feature of being able to eliminate unsuitable scanning electron microscope specimens. The microphotograph, size distribution and chemical composition of second phase particles in Q195 steel were characterizated using SEM and EDS. In order to observe the nanometer grade second phase particles, SEM with high resolution should be used. Whether can be second phase particles discovered lies on the preparation method of samples. This method is easier for sample preparation, lower cost, more efficient than TEM method.

Extremely fine, detailed morphological characteristics of micro-organisms, hitherto unavailable, can now be determined with the Scanning Electron Microscope. Morphological characteristics related to evolutionary sequences, specific, generic, and familial relationships will permit more natural faunal classifications and enable more accurate ecological and biostratigraphic correlations.

The pore spaces in sandstones are the result of the original depositional fabric and the degree of post-depositional alteration that the rock has experienced. The largest pore volumes are present in coarse-grained, well-sorted materials with high sphericity. The chief mechanisms which alter the shape and size of the pores are precipitation of cementing agents and the dissolution of soluble components. Each process may operate alone or in combination with the other, or there may be several generations of cementation and solution.The scanning electron microscope has ‘been used in this study to reveal the morphology of the pore spaces in a variety of moderate porosity, orthoquartzites.

The ease of operation of the scanning electron microscope has insured its wide application in medicine and industry. The micrographs are pictorial representations of surface topography obtained directly from the specimen. The need to replicate is eliminated. The great depth of field and the high resolving power provide far more information than light microscopy.
