Bone health depends on the bone mineral density and mechanical strength, characterised quantitatively and inferred through qualitative parameters such as the trabecular and cortical micro-architecture, and other parameters describing the bone cells. Among these cells, the osteocyte has been recognised as the orchestrator of bone remodelling, playing a key role in directing osteoblastic and osteoclastic activities. Conventional optical and electron microscopies have greatly improved our understanding of the cell physiology mechanisms involved in different osteoarticular pathophysiological contexts, especially osteoporosis. More recently, imaging methods exploiting synchrotron radiation, such as X-ray tomography, ptychography, and deep ultraviolet and Fourier transform infrared spectroscopies, have revealed new biochemical, chemical and 3D morphological information about the osteocyte lacuna, the surrounding matrix and the lacuna–canalicular network at spatial length scales spanning microns to tens of nanometres. Here, we review recent results in osteocyte lacuna and lacuna–canalicular network characterisation by synchrotron radiation imaging in human and animal models, contributing to new insights in different physiologic and pathophysiological situations.

Ag/TiO2 thin films were prepared using the sol-gel spin coating method. The microstructural growth behaviors of the prepared Ag/TiO2 thin films were elucidated using real-time synchrotron radiation imaging, its structure was determined using grazing incidence X-ray diffraction (GIXRD), its morphology was imaged using the field emission scanning electron microscopy (FESEM), and its surface topography was examined using the atomic force microscope (AFM) in contact mode. The cubical shape was detected and identified as Ag, while the anatase, TiO2 thin film resembled a porous ring-like structure. It was found that each ring that coalesced and formed channels occurred at a low annealing temperature of 280 °C. The energy dispersive X-ray (EDX) result revealed a small amount of Ag presence in the Ag/TiO2 thin films. From the in-situ synchrotron radiation imaging, it was observed that as the annealing time increased, the growth of Ag/TiO2 also increased in terms of area and the number of junctions. The growth rate of Ag/TiO2 at 600 s was 47.26 µm2/s, and after 1200 s it decreased to 11.50 µm2/s and 11.55 µm2/s at 1800 s. Prolonged annealing will further decrease the growth rate to 5.94 µm2/s, 4.12 µm2/s and 4.86 µm2/s at 2400 s, 3000 s and 3600 s, respectively.

nanoCT with synchrotron radiation has a high spatial resolution up to nanoscale which provides an approach to the 3D inter-structure of sample of tiny size in the research of biology, materials and so on. However, due to the high resolution it provide, the field of view is limited to only about 15~60μm. In order to improve the scale of field of view, we discussed a new scan mode called half-scan which is using the symmetry of parallel-beam x-ray and newly applied in the synchrotron radiation imaging field. In this paper, we deduced the data processing method which is consisted of data preprocessing and filter back projection (FBP) algorithm. Numerical simulation and analysis demonstrate that the field of view can be enlarged up to 100% with this method.

To evaluate the anti-osteoporotic effect of antler blood, the relationship between the change of element content and the bone mineral density (BMD) increase in femur was investigated. Female Wistar rats were randomly divided into three groups: sham-operated group (SHAM,n=5), ovariectomized group (OVX,n=5) and ovariectomized group with antler blood treatment (n=5). The femoral BMD was analyzed by dual-energy X-ray absorptiometry (DXA) and the element relative content was determined by synchrotron radiation X-ray fluorescence (SRXRF) microprobe. The results showed that the femoral BMD in ovariectomized rats was significantly lower than that of sham-operated rats (p<0.05) but reversed by antler blood treatment (p<0.05). A further study demonstrated that the relative contents of phosphorus (P), calcium (Ca), zinc (Zn) and strontium (Sr) were obviously lower in ovariectomized rats compared to sham-operated rats but only the relative contents of P, Ca and Zn were normalized by antler blood treatment (p<0.05). Our experiments revealed that loss of element Ca, P, Zn and Sr was closely related to the BMD reduction in ovariectomized rats and the anti-osteoporotic effect of antler blood was mediated by increasing the contents of P, Ca and Zn.

Since the early works on GP zones and the model independently proposed by Preston and Guinier on the first steps of precipitation in supersaturated solid solution of aluminium containing a few percent of copper, many works have been performed to understand the structure of different stages in the sequence of precipitation.The scheme which is generally admitted can be drawn from a work by Phillips.In their original model Guinier and Preston analysed a GP zone as composed of a single (100) copperrich plane surrounded by aluminum atomic planes with a slightly shorter distance from the original plane than in the solid solution.From X-ray measurements it has also been shown that GP1 zones were not only copper monolayer zones. They could be up to a few atomic planes thick. Different models were proposed by Guinier, Gerold, Toman. Using synchrotron radiation, proposals have been recently made.
