Objective. We studied prosexual effects ofEremomastax speciosaaqueous extract in male adult rats.Materials and Methods. 100 and 500 mg/kg of extract were administered orally (days 0, 1, 4, 7, 14, and 28 (posttreatment)). The sexual behavior of rats receiving a single dose (500 mg/kg) was also evaluated after pretreatment with Lω-NAME (10 mg/kg), haloperidol (1 mg/kg), or atropine (5 mg/kg). Controls received distilled water or testosterone enanthate (20 mg/kg/day/3 days(s.c.)before the test).Results. The extract (days 1–14) had no significant effect on mount, intromission, and ejaculation frequencies but on day 28 (14 days after treatment), it increased frequency of mounts and intromissions at 500 mg/kg. Mount, intromission, and ejaculation latencies reduced and postejaculatory intervals decreased but the effect did not persist 2 weeks after treatment. Extract prosex effects were greatly reduced by atropine and completely abolished by haloperidol, while Lω-NAME increased mount latency and potentiated extract effect on intromission and ejaculation latencies.Conclusion. In summary,E. speciosaextract can have positive effects on male sexual motivation and performance when administered for two weeks at the dose of 500 mg/kg. The effects (dopaminergic and/or cholinergic dependent) tend to appear during the posttreatment period.

Intestinal sucrase activity of the rat varies with the age, but not the sex, of the animal. Sucrase activity of rats 23 days of age was approximately two-thirds that of adults.Sucrase activity of adult rats was significantly decreased by several days of fasting. The decrease was rapid during the first 2 to 4 days of the fast, but became negligible thereafter.Diets containing large (70%) amounts of sucrose, galactose, melizitose, or α-methyl-D-glucoside produced highly significant increases in intestinal sucrase levels (compared with a carbohydrate-free, high-casein control diet) when fed ad libitum for 24 hours to adult male rats previously fasted for 3 days. Similar diets containing fructose, fructose plus glucose in equimolar amounts, or maltose significantly increased sucrase activity, but diets containing glucose, mannose, xylose, or lactose were not stimulatory. A 70% raffinose diet significantly decreased sucrase activity. Normal male rats which were fed the 70% sucrose diet for 4 weeks had sucrase activities similar to those of controls fed Purina fox checkers, but animals fed the carbohydrate-free, high-casein diet for 1 day or longer had sucrase activities significantly lower than those of controls. The significance of these observations in regard to enzyme "adaptation" is discussed.

Creatine plays a crucial role in developing the brain, so much that its genetic deficiency results in mental dysfunction and cognitive impairments. Moreover, creatine supplementation is currently under investigation as a preventive measure to protect the fetus against oxidative stress during difficult pregnancies. Although creatine use is considered safe, posing minimal risk to clinical health, we found an alteration in morpho-functional maturation of neurons when male rats were exposed to creatine loads during brain development. In particular, increased excitability and enhanced long-term potentiation (LTP) were observed in the hippocampal pyramidal neurons of weaning pups. Since these effects were observed a long time after creatine treatment had been terminated, long-lasting modifications persisting into adulthood were hypothesized. Such modifications were investigated in the present study using morphological, electrophysiological, and calcium imaging techniques applied to hippocampal Cornu Ammonis 1 (CA1) neurons of adult rats born from dams supplemented with creatine. When compared to age-matched controls, the treated adult offspring were found to retain enhanced neuron excitability and an improved LTP, the best-documented neuronal substrate for memory formation. While translating data from rats to humans does have limitations, our findings suggest that prenatal creatine supplementation could have positive effects on adult cognitive abilities.

1 The aim of this study was to ascertain the reproductive effects of PCB 77 on adult male rats and to determine its concentration in the liver and testis. Adult male rats (n=15/group) were treated subcutaneously with a singledoseof18 mg/kgbw(PC18)orwith60 mg/kg bw (PC60). The substance was dissolved in a 10 ml volume of peanut oil/kg. Control rats received the same volume of the vehicle. The reproductive effects as well as the concentration of PCB 77 in the liver and testis were investigated 1, 4 and 8 weeks after treatment. 2 In both groups, the daily sperm production (DSP; 6106) remained permanently reduced in the PC18 as well as in the PC60 groups throughout the entire investigation period (DSP week 8: control: 31+7; PC18: 22+5; PC60: 20+7). The sperm number (6106) per cauda epididymis was affected only at the 1st and 4th week after treatment (control week 1: 211+67; PC18 week 1: 135+62; PC60 week 1: 142+49). Moreover, a significant increase in the percentage of abnormal sperm was observed 4 weeks following treatment in the PC18 and PC60 groups and 8 weeks after treatment in the PC60 group. Abnormal tails were the most frequent changes observed. 3 The relative testicular and prostata weights (g) were slightly increased in the PC60 group at the 1st and 4th week following treatment (testis weight: control/I: 0.46+0.02; PC60/I: 0.51+0.03). 4 The serum testosterone concentrations and effects on testis morphology were not reported. 5 The maximum concentration of PCB 77 was detected in the liver and testis 1 week after treatment. The concentration declined 4 weeks after treatment in both organs, but still a significant amount of PCB 77 was detectable in the liver as well as in the testis 8 weeks after treatment. 6 The results demonstrate that PCB 77 affects sperm variables when applied to adult rats and that the elimination of PCB 77 in the testis parallels that of the liver.

Gray (Asteraceae) andTurnera diffusaWilld (Turneraceae) are employed in traditional medicine as aphrodisiacs; however, there is no scientific evidence supporting the prosexual properties ofC. mexicana. The aim of this study was to determine whether an aqueous extract ofC. mexicana(Cm) stimulates rat male sexual behavior in the sexual exhaustion paradigm. Sexually exhausted (SExh) male rats were treated with Cm (80, 160, and 320 mg/kg), an aqueous extract ofT. diffusa(Td), or yohimbine. The sexual exhaustion state in the control group was characterized by a low percentage of males exhibiting mounts, intromissions, and ejaculations and no males demonstrating mating behavior after ejaculation. Cm (320 mg/kg), Td, or yohimbine significantly increased the proportion of SExh rats that ejaculated and resumed copulation after ejaculation. In males that exhibited reversal of sexual exhaustion, Cm (320 mg/kg) improved sexual performance by reducing the number of intromissions and shrinking ejaculation latency. The effects of treatments on sexual behavior were not related with alterations in general locomotion. In conclusion, the prosexual effects of Cm, as well as those of Td, are established at a central level, which supports the traditional use ofC. mexicanafor stimulating sexual activity.

(1) Background: Hormone-dependent events that occur throughout spermatogenesis during postnatal testis maturation are significant for adult male fertility. Any disturbances in the T/DHT ratio in male progeny born from females fertilized by finasteride-treated male rats (F0:Fin) can result in the impairment of testicular physiology. The goal of this work was to profile the testicular transcriptome in the male filial generation (F1:Fin) from paternal F0:Fin rats. (2) Methods: The subject material for the study were testis from immature and mature male rats born from females fertilized by finasteride-treated rats. Testicular tissues from the offspring were used in microarray analyses. (3) Results: The top 10 genes having the highest and lowest fold change values were mainly those that encoded odoriferous (Olfr: 31, 331, 365, 633, 774, 814, 890, 935, 1109, 1112, 1173, 1251, 1259, 1253, 1383) and vomeronasal (Vmn1r: 50, 103, 210, 211; Vmn2r: 3, 23, 99) receptors and RIKEN cDNA 5430402E10, also known as odorant-binding protein. (4) Conclusions: Finasteride treatment of male adult rats may cause changes in the testicular transcriptome of their male offspring, leading to a defective function of spermatozoa in response to odorant-like signals, which are recently more and more often noticed as significant players in male fertility.

Aim: To study the long-term effects of exposure of pregnant Wistar rats to low dose of bisphenol A (BPA) by measuring to the level of steroid hormones and sexual behavior of adult male offspring of the first generation. Material and research methods: BPA as part of the Dorfman gel was gavaged during the last week of pregnancy, when androgen-dependent sexual brain differentiation occurs, in a daily dose of 25 mcg/kg b.w. (threshold teratogenic dose). Male sexual behavior was evaluated by proceptive reactions, the duration of latent and refractory periods, the number of mounts, intromissions and ejaculations in the presence of a receptive female. Female sexual behavior was assessed by lordosis reactions of orchidectomized and activated by the introduction of estradiol and progesterone males in the presence of a normal male. A neuromorphological analysis of the sex-dimorphic area of the brain, the medial preoptic nucleus of the hypothalamus, was performed by histological examination and karyometry of neurons. Results: Prenatally administered BPA caused a very slight increase in the anogenital distance in newborn animals and did not affect the terms of puberty. The levels of testosterone and corticosterone in the blood plasma of males of 6 months of age did not differ from the control indices. At 10 months of age, all experimental males showed sharply weakened sexual motivation for mating with females, and in 4 from 5 animals, copulative components of sexual behavior were absent. There was no ejaculations in the 5th male as well, while numbers of the mounts without intromissions and ones with intromissions significantly reduced. In the BPA group, all descendants showed active female behavior in the presence of a normal male, which manifested in lordosis reactions and a high lordosis index. According to the histological study of medial preoptic nucleus, the activity of neurocytes in the male offspring of BPA-exposed females was significantly reduced, and their nuclei volume distribution was some different from the control. Conclusions: The data obtained indicate epigenetic disorders of the sexual brain differentiation program due to the prenatal exposure to BPA in dose that does not cause significant teratogenic effects. This should be taken into account when evaluating the potential hazard of BPA for reproductive health. Single neonatal treatment with vitamin A (retinol) dramatically reduced the sexual activity of adult male rats. In females there was a significant decrease in the Meyerson index and a non significant decrease in the lordosis quotient. The effect of three perinatal treatments (at the first, third and fifth day) with all-trans retinoic acid was much weaker, causing only a significant increase in the time of the first ejaculation in males and non- significant decrease in the lordosis quotient of females. The experiments call attention to the false imprinting provoking effect of materials acting on members of the steroid receptor superfamily with possible human health aspect.
