Neuroleptics of the 8-methylthio-10-piperazino-10,11-dihydrodibenzo[b,f]thiepin series: New compounds and new procedures

Heating of 8-methylthiodibenzo[b,f]thiepin-10(11H)-one with mono-p-toluenesulfonates of 1-methylpiperazine and piperazine in vacuo led in good yields to the enamines IX-XI, the first of which was reduced with zinc and acetic acid to the base I (methiothepin). The ester III(oxyprothepindecanoate) was obtained by reaction of the amino alcohol II (oxyprothepin) with decanoic acid in the presence of N,N'-carbonyldiimidazole and by substitution of 10-chloro-8-methylthio-10,11-dihydrobenzo[b,f]thiepin with 1-(3-decanoyloxypropyl)piperazine. The substitution reaction of the same chloro compound with piperazine gave two stereoisomeric 1,4-disubstituted piperazines V. Reaction of the amino alcohol II with octyl isocyanate afforded the carbamate VI, an isoster of oxyprothepin decanoate (III). This substance showed in the test of antiapomorphine activity in dogs the properties of a long-acting antiemetic. Two new potential metabolites (XII, XIII) of compounds I-III were synthesized and new pharmacological data are given for two further potential metabolites (XIV, XV) of oxyprothepin (II).

Diazotization of 4-fluoroanthranilic acid (V) and the following reaction with sodium disulfide gave the dithio diacid VII which was reduced with lithium aluminium hydride to 4-fluoro-2-mercaprobenzyl alcohol (XI). Its reaction with 2-chloro-5-iodothiophene afforded the alcohol XIII which was transformed via the chloride XIV and the nitrile XV to [2-(5-chloro-2-thienylthio)-4-fluorophenyl]acetic acid (XVI). Cyclization with phosphorus pentoxide in toluene resulted in 2-chloro-8-fluorothieno[2,3-b]-1-benzothiepin-4(5H)-one (XVIII) which was converted via the alcohol XIX to the chloro derivative XX. The substitution reaction with 1-methylpiperazine led to the title compound IV which is a long-acting and very potent tranquillizer but did not reveal, in the animal tests performed, the properties of a neuroleptic agent.

The resolution of racemic clorothepin (Ia) was repeated and the water-soluble methanesulfonates of (S)(+)-clorothepin and (R)(-)-clorothepin were prepared which were used in recent studies of the stereoselectivity of action of this neuroleptic agent. Alkylation of the secondary amine VIa with 2-chloroethyl decanoate resulted in noroxyclothepin decanoate IVa whose basically catalyzed ethanolysis afforded smoothly the amino alcohol IIa. Reactions of amines VIa and VIb with 1,2-butene oxide gave the amino alcohols VIIab. Alkylation of the amine VIa with 5-bromopentan-2-one and the following reduction of the amino ketone IXa formed gave the amino alcohol VIIIa. Amino alcohols IIa and IIIb were transformed by treatment with thionyl chloride to the chloroalkylamines Xa and XIb which were used for the synthesis of mandelates XIIa, XIIIb and benzilates XIVa, XVb derived from noroxyclothepin IIa and oxyprothepin IIIb. A substitution reaction of 2,11-dichloro-10,11-dihydrodibenzo[b,f]thiepin with 1,4-diazabicyclo[4,3,0]nonane led to the clorothepin analogue XVI. From 2-chlorodibenzo[b,f]thiepin-10(11H)-one XVII via the 11-bromo derivative XVIII the amino ketone XIX was prepared. While its reduction with sodium borohydride gave the cis-amino alcohol XXI, the reduction with diborane gave the trans-amino alcohol XXII. The pharmacological properties of the new piperazine derivatives are described; some of them showed a high degree of neuroleptic activity of various profile.

The acid VI, obtained from 2,5-difluorothiophenol (IV) and (2-iodophenyl)acetic acid, afforded by cyclization with polyphosphoric acid 6,9-difluorodibenzo[b,f]thiepin-10(11H)-one (VII) in a satisfactory yield. Two further steps led to the chloro derivative X giving by a substitution reaction with 1-methylpiperazine the title compound III. This substance exhibits some 10% incoordinating activity of the unsubstituted compound I and an indication of cataleptic activity, in contrast to the inactive analogous dichloro compound II. The bulky atom of chlorine in the vicinity of the methylpiperazine residue interferes evidently with the CNS activity; the influence of the atom of fluorine is much less pronounced in this line.

In this work we describe the synthesis of some novel fused imidazo [2, 1-b] [1, 3] thiazole derivatives. The reaction of 1, 2-diaminoethane 1 with carbon disulphide in H2O/ETOH as solvent furnishes 4, 5-dihydro-1H-imidazol-2-thiol 2 under reflux condition. the reaction of 4,5-dihydro-1H-imidazol-2-thiol on treatment with ethylchloro acetate and aromatic aldehyde in presence of anhydrous sodium acetate and acetic acid as solvent to give (Z)-2-(arylidene)-5,6-dihydroimidazo [2,1-b] [1,3] thiazol-3(2H)-one 3a-j. Compounds 3a-j was condensed with hydroxylamine to give 3-(aryl)-2, 3, 6, 7-tetrahydroimidazo [2, 1-b] [1,3] thiazolo [5, 4-d] isoxazole 4a-j. The structures of the new compounds were established by elemental analyses, IR,1H NMR and13C NMR data.

By reacting calix[4]-1,3-diethoxylaminoethyl derivative (2) with phenyl isothiocyanate, novel dendritic calix[4]arene derivative (3) with two different branched chains was synthesized in a yield of 78%. By reacting compound 2 with 1,6-diisocyanatohexane or N,N′-bis(2-chloracetamide)ethylene in a “1 + 1” intermolecular addition mode, novel lariat calix[4]-1,3-aza-crowns (4 and 5, respectively) with two branched ethoxyl chains were prepared in reasonable yields. The composition, structures, and conformations of all new compounds were confirmed by elemental analyses, IR, ESI-MS, 1H NMR, and so forth. The liquid–liquid extraction experiments showed that all new hosts possessed good extraction abilities towards soft and hard metal cations. The liquid membrane transport experiments suggested that they had good transport abilities for K+ and Ag+. The experiments of phase transfer catalysis indicated that they possessed excellent catalytic properties of aromatic nucleophilic substitution reaction and benzyl nucleophilic substitution. The optimum yields of products in catalytic reaction were as high as approximately 100%.

Two improved routes to synthesize 1-benzyl-1,4,7,10-tetraazacyclododecane (6) and 1,4,7,10- tetraazacyclododecane-1-acetic acid ethyl ester (11) are described as well as the synthesis of 1-{2-[4-(maleimido-N-propylacetamidobutyl)amino]-2-oxoethyl}-1,4,7,10-tetraazacyclododecane- 4,7,10-triacetic acid (17) and its Y, Ho, Tm, and Lu complexes. The 1H and 13C NMR spectra of the new compounds as well as the single crystal X-ray structure analyses of the intermediates 4-benzyl-1,7-bis(p-toluenesulfonyl)diethylenetriamine (3) and 1,4,7-tris(p-toluenesulfonyl)diethylenetriamine (7) are reported and discussed. The rare earth complexes of 17 have been characterized by 1H NMR spectroscopy and MALDI-TOF mass spectrometry.

A number of new complexes have been synthesized by reaction of novel ligands acetic acid(4-methyl-benzylidene)hydrazide (L1) and acetic acid(naphthalen-1-ylmethylene)hydrazide (L2) with copper(II) nitrate. These new compounds were characterized by elemental analysis, TG, DTA, IR spectroscopy, UV spectral techniques. The changes observed between the FT-IR and UV-Vis spectra of the ligands and of the complexes allowed us to establish the coordination mode of the metal in complexes. The results suggest that the Schiff bases L1and L2coordinate as univalent anions with their bidentate N,O donors derived from the carbonyl and azomethine nitrogen. Also the probing of thermal analysis complexes can detect which complex has excellent thermal stability.
