Four factorial experiments were conducted in which were examined and compared factors which affect revival of ram spermatozoa after pellet.freezing and other methods of freezing. Revival of spermatozoa after pellet.freezing was the best in egg yolk-lactose (333 mM), followed by egg yolk-raffinose (333 mM), egg yolk-glucose (355 mM), and egg yolk-fructose (355 mM) diluents. Within the glycerol range of 0-7%, lactose gave the best results with 3%, raffinose and glucose with 5%, and fructose with 7% (vJv) glycerol.

Six factorial experiments were conducted to examine the effects of concentrations of 2-amino-2-hydroxymethylpropane-l,3-diol (tris), sugars, erythritol, catalase, ethylenediaminetetra-acetic acid (EDTA), glycerol and egg yolk in the freezing diluent on the survival of boar spermatozoa after freeze-thawing.

From the results of four factorial experiments on the deep-freezing of bull spermatozoa: (1) Revival rates of semen treated with lecithin followed by cooling to 5�C with dilution just before freezing did not differ significantly from samples diluted at 30�C soon after collection of the ejaculate. Aging the spermatozoa at 5�C for 6 hr before freezing was beneficial and a slow freezing rate of O� 5 to 1 degC fall per minute to -15�C followed by 3 degC fall below this temperature gave better results than faster rates. Time of storage at 5�C and freezing rate interacted, as fast freezing was much better tolerated by spermatozoa which had been aged at 5�C for 6 or 18 hr.

Purpose: Conducting a honey diluent test for creeples of sperm of a drone honey bee.Materials and methods. The material for the research served a sperm of the milled drone drums of the "Prioksky" type of the Midway breed of bees. The selection of sperm was carried out in June-July 2020 g by the method of artificial stimulation of the turning of the endofalosha in half-armed drones aged 25-30 days. The rock type "Prioksky" of the middle Russian breed of bees. Before freezing, the sperm was stored in glass capillaries in the cooled state at 3 ° C for 2 months. The following composition of the diluent was tested - 10% honey, lactose, sucrose, egg yolk and dimethyl sulfoxide.Results. Studies have shown the viability of sperm at 64.0 ± 1.8% (41.5-83.7), and a total mobility of 2.2 ± 0.6% (0-11.5). To evaluate the fertilizing ability of sperm, carried out artificial insemination of 10 bee modules. In 4 seeded bees dykens, the presence of sperm in a seed-hearter with a concentration of sperm from 0.22-4.4 million / μl is revealed. In paired eggs of three other seeded matters, the presence of sperm and the complete absence of spermatozoa in the seed-receptionist are recorded.Conclusion. Tests of the honey diluent for deep freezing sperm of the drone honey bees in liquid nitrogen confirmed its cryophylactic properties.

The addition of 6% (vJv) egg yolk to a synthetic diluent [247 mM glucose, 49 mM NaCI, 5 mM KCI, 5 mM phosphate buffer, and 7�5% (vJv) glycerol] improved the survival of ram spermatozoa after deep�freezing. Judging by the activity of spermatozoa after thawing, a single addition of glycerol to the diluted semen at 5�C was as effective as multiple additions giving a graded increase of glycerol concentra-tion over a period of 20 min. Reciprocal replacement of the sodium chloride of the diluent by an increase in the concentration of the phosphate buffer showed that motility of the spermatozoa after thawing was depressed when the level of phosphate exceeded 10 mM.

ABSTRAKAir kelapa muda varietas viridisdapat dijadikan pengencer aletrnatif semen cair bagi program IB di daerah minim sarana semen beku. Tujuan penelitian ini untuk menguji pengaruh penggunaan air kelapa muda viridissebagai bahan pengencer terhadap kualitas semen cair kambing Boer setelah didinginkan. Dilaksanakanselama 3 bulan di Laboratorium Fakultas Peternakan UBUnit SumberSekar,Malang. Metodenya yaitu eksperimen. Semen dari  3 pejantan Boer umur 3-5 tahun, dikoleksi seminggu sekali dengan VB. Air kelapa mudaviridis umur 5-7 bulan serta tris aminomethane sebagai kontrol. Didesain menggunakan Rancangan Acak Kelompok (RAK) dengan 2 perlakuan yaitu P0 (tris aminomethane + 10% KT) dan  P1 (air kelapa muda viridis + 10% KT) masing-masing diulang 10 kali. Data dianalisis dengan analisis Ragam (Anova) dengan software Genstat 18. Variabelnya yaitu motilitas individu, viabilitas dan abnormalitas. Hasil penelitian yaitu motilitas individu pada P1bertahan sampai 4 hari (40,5± 24,3%), viabilitas terbaik sampai hari ke-5 (42±24,6%), abnormalitas terendah di hari ke-7(1,31± 0,6). Kesimpulannya, Pengencer air kelapa muda viridis dapat mempertahankan kualitas semen cair kambing Boer selama 4 hari untuk motilitas dan 5 hari untuk viabilitas.Kata Kunci:pengencer, air kelapa, varietas viridisABSTRACTYoung viridis coconut water could be used as an alternative to liquid semen diluent for artificial insemination program in the area with limited facility for frozen semen production. This study evaluated the use of young coconut water as a diluent on liquid semen quality of Boer goat after cold storage. This study was carried out for 3 months at Sumber Sekar Laboratory, Faculty of Animal Husbandry, University of Brawijaya, Malang. The semen was collected from 3 Boer bucks aged at 3 to 5 years old. The semen collection was done once a week with the aid of artificial vagina. The diluents used were young Viridis coconut (5 to 7 months old) and tris aminomethane. The method used was an experiment in a randomized block design with 2 treatments and 10 replicates. The treatments used were T0: tris aminomethane + 10% egg yolk (control) and T1:  young Viridis coconut water + 10% egg yolk. Data were analyzed by analysis of variance using Genstat 18 software. The variables measured were sperm individual motility, viability, and abnormality. The results showed that the sperm individual motility in T1 survived up to 4 days (40.5± 24.3%), the best viability at 5 days (42.0±24.6%),  while the lowest abnormality at 7 days (1.31±0.6). It could be concluded that: 1. Tris aminomethane diluent has higher quality with the storage length up to 9 days, 2. Young Viridis coconut water diluent could preserve liquid semen quality of Boer goat up to 4 days for sperm motility and 5 days for sperm viability.Keywords: diluents, coconut water, viridis variety
