Two Friesian calves were fitted with re-entrant cannulae in the duodenum and four Friesian calves were fitted with re-entrant cannulae in the duodenum and terminal ileum within 1 week of birth. At 35 days of age the calves were weaned from milk and offered a diet containing 72% barley, 3.5% urea and 15 % wheat chaff. Total collections of digesta flowing from the duodenum and ileum were made for 12 hr at weekly intervals for 6 weeks after weaning. Of the dry matter ingested, 26% was apparently digested in the stomach 1 week after weaning, 40% by 2 weeks after weaning and increasing to 51% by 6 weeks after weaning. The flow of nitrogen to the duodenum was greater than the nitrogen intake in the first week after weaning, and it gradually decreased to 83% of the nitrogen intake by 6 weeks after weaning. During this period the contribution of microbial nitrogen to the nitrogen flow increased from 28 to 71%. The apparent absorption of amino acids from the small intestine increased with increasing feed intake, but the uptake of methionine was less than the methionine requirement for growth. _________________ * Part I, Aust. SUMMARY1. Three pigs prepared with re-entrant cannulae in the proximal duodenum and terminal ileum were used to study flow rate of nitrogen, and digestion and absorption of dry matter, nitrogen, gross energy and starch. The pigs were given a semi-purified diet, a hard wheat diet and a soft wheat diet. These were approximately isonitrogenous.2. Nitrogen content of insoluble dry matter of duodenal digesta was much higher on the semi-purified diet than on the wheat-based diets.3. Apparent digestibilities of dry matter, energy and nitrogen in the small intestine were in the order semi-purified diet > hard wheat > soft wheat although differences were not statistically significant. Similar results were obtained for total digestibility of these components.4. Absorption of most amino acids from the small intestine was higher for casein in the semi-purified diet than for wheat protein. Significantly more lysine, arginine, isoleucine and tyrosine were absorbed from hard than from soft wheat.5. With all diets there was almost complete digestion of starch in the small intestine.6. There was little digestion of nutrients in the large intestine, and thus digestibilities calculated from faecal analyses showed similar trends to those calculated from analyses of digesta from the terminal ileum. Most values compared favourably with those reported elsewhere for digestibilities of amino acids in wheat.7. It was concluded that casein was superior to the wheat proteins but that protein of hard wheat was of better quality than that of soft wheat when evaluated by ileal recovery of animo acids, due largely to a greater absorption of lysine.

Eleven pigs were fed on one of six diets containing wheat alone or supplemented with either meat meal, fish meal, peanut meal, soybean meal or sunflower meal. Digesta were collected from the re-entrant cannulas at the mid-point and the terminal ileum of the small intestine at 2, 4, 6, 9 and 12 h after feeding. Measurements were made of flow rates of digesta, dry matter, nitrogen and amino acids at both locations in the small intestine. Peak values were usually observed for all parameters, at 2 h and from 4 to 6 h after feeding, at the mid-point and the terminal ileum respectively. Flow rates and the amounts of components in digests were often characteristic of the protein source in the diet and the location of the cannula. High concentrations of some free amino acids were observed in digesta at the mid-point, and lower concentrations at the terminal ileum in the small intestine; the significance of endogenous secretions in the small intestine is discussed.

Four castrated pigs, each fitted with a re-entrant cannula in the terminal ileum, were used to study digestion in the small intestine. A nitrogen-free diet was used for the estimation of metabolic nitrogen and amino acids. Faba beans, as the sole source of dietary protein, were used raw or after autoclaving for 30 or 60 min. The four diets were fed to the pigs in a 4 × 4 latin square experiment. The pigs were fed each diet for 6 days prior to a 24-hr collection of total ileal contents. Autoclaving of faba beans had no significant effect on digestibility of dry matter, gross energy, nitrogen and individual amino acids except arginine, which was significantly increased. The intestinal uptake of arginine was the highest and of cystine the lowest in all faba bean diets. It was concluded that autoclaving faba beans had no beneficial effect on the digestion of nutrients in the small intestine of the pig.

This study used an experimental model, described in a companion paper, to examine the effects of feed intake on protein turnover in the small intestine of lambs. Ten male castrate lambs (∼ 10 months old) were offered, via continuous feeders, either 400 (n = 5) or 1200 (n = 5) g/day lucerne chaff, and mean experimental liveweights were 28 and 33 kg respectively. All lambs were prepared with catheters in the cranial mesenteric vein (CMV), femoral artery (FA), jugular vein and abomasum, and a blood flow probe around the CMV. Cr-EDTA (0·139 mg Cr/ml, ∼ 0·2 ml/min) was infused abomasally for 24 h and L-[2,6-3H]phenylalanine (Phe) (420±9·35 μCi into the abomasum) and L-[U-14C]phenylalanine (49·6±3·59 μCi into the jugular vein) were also infused during the last 8 h. Blood from the CMV and FA was sampled during the isotope infusions. At the end of infusions, lambs were killed and tissue (n = 4) and digesta (n = 2) samples removed from the small intestine (SI) of each animal. Transfers of labelled and unlabelled Phe were measured between SI tissue, its lumen and blood, enabling both fractional and absolute rates of protein synthesis and gain to be estimated.Total SI mass increased significantly with feed intake (P < 0·05), although not on a liveweight basis. Fractional rates of protein gain in the SI tended to increase (P = 0·12) with feed intake; these rates were −16·2 (±13·7) and 23·3 (±15·2) % per day in lambs offered 400 and 1200 g/day respectively. Mean protein synthesis and fractional synthesis rates (FSR), calculated from the mean retention of 14C and 3H in SI tissue, were both positively affected by feed intake (0·01 < P < 0·05). The choice of free Phe pool for estimating precursor specific radioactivity (SRA) for protein synthesis had a major effect on FSR. Assuming that tissue free Phe SRA represented precursor SRA, mean FSR were 81 (±15) and 145 (±24) % per day in lambs offered 400 and 1200 g/day respectively. Corresponding estimates for free Phe SRA in the FA and CMV were 28 (±2·9) and 42 (±3·5) % per day on 400 g/day, and 61 (±2·9) and 94 (±6·0) on 1200 g/day. The correct value for protein synthesis was therefore in doubt, although indirect evidence suggested that blood SRA (either FA or CMV) may be closest to true precursor SRA. This evidence included (i) comparison with flooding dose estimates of FSR, (ii) comparison of 3H[ratio ]14C Phe SRA in free Phe pools with this ratio in SI protein, and (iii) the proportion of SI energy use associated with protein synthesis.Using the experimental model, the proportion of small intestinal protein synthesis exported was estimated as 0·13–0·27 (depending on the choice of precursor) and was unaffected by feed intake. The contribution of the small intestine to whole body protein synthesis tended to be higher in lambs offered 1200 g/day (0·21) than in those offered 400 g/day (0·13). The data obtained in this study suggested a role for the small intestine in modulating amino acid supply with changes in feed intake. At high intake (1200 g/day), the small intestine increases in mass and CMV uptake of amino acids is less than absorption from the lumen, while at low intake (400 g/day), this organ loses mass and CMV uptake of amino acids exceeds that absorbed. The implications of these findings are discussed.

Six Friesian steers (300 kg liveweight) were prepared with rumen and abomasal cannulae, and fed three artificially dried grasses at three levels of organic matter (OM) intake and at two stages of maturity. The grasses were paspalum (Paspalum dilatatum), oats (Avena sativa) and kikuyu (Pennisetum clandestinum). The apparent digestion of organic matter (OM) in the whole tract decreased with increasing maturity for all grasses. The proportion of OM digested in the rumen was greater for paspalum and kikuyu than oats, and increased with advancing maturity for paspalum and kikuyu. Microbial nitrogen flow from the rumen was 26 and 25 g/kg of OM apparently digested in the rumen for paspalum, 32 and 36 for oats and 25 and 14 for the early and late maturity kikuyu. The true degradation of grass protein in the rumen was between 0.70 and 0.80 for all three grasses, at all three feed intakes and for both stages of maturity of the grasses. Total rumen pool size was less for oats than paspalum or kikuyu. The retention time of particles and fluid in the rumen increased with decreasing feed intake for oats.

1. In two experiments, sheep prepared with a rumen cannula and with re-entrant cannulas in the duodenum and ileum were continuously fed on diets of dried grass, dried grass plus formalin-treated casein, or dried grass plus untreated casein. Paper impregnated with chromic oxide was given once daily via the rumen fistula.2. In ten 24 h collections of digesta entering the duodenum and eleven 24 h collections of digesta reaching the ileum of sheep given dried grass, there were highly significant correlations between the 24 h flows of Cr marker and the corresponding flows of dry matter, organic matter, nitrogen, gross energy, hemicellulose and cellulose (P < 0.01) at both sites.3. Daily amounts of non-ammonia N and of individual amino acids entering and leaving the small intestine and of total N excreted in faeces and urine are given.4. Net retention of supplementary N was 36% when the supplement was administered as formalin-treated casein, but only 17% when it was administered as untreated casein.5. Formalin treatment of casein significantly increased the daily amounts of non-ammonia N entering the small intestine (P < 0.01) and the amounts of non-ammonia N apparently absorbed therein (P < 0.05).6. Apparent absorption of amino acids from the small intestine was significantly greater (P < 0.05) with treated casein than with untreated casein. There were relative increases in the small amounts of several free amino acids measured, including taurine, in the ileal digesta of sheep receiving the treated casein supplement.

1. Casein was labelled with pairs of radioactive amino acids, lysine, tyrosine and leucine, one with I4C and the other with 3H, by jugular infusion into lactating goats followed by isolation of the double-labelled casein from the milk. Total milk protein was similarly labelled by jugular infusion of [35S]cystine. U-14C-labelled fraction- 1 leaf protein was isolated from lucerne (Medicago sativa) grown in an atmosphere of 14C022. The proteins were treated withdifferent levels(333 and667 mmol/kgprotein) offormaldehyde, glutaraldehyde and glyoxal.3. Absorption from the small intestine was measured in sheep with fistulas in the abomasum and terminal ileum, using Cr-EDTA as the digesta flow marker, by introducing radioactive casein into the abomasum.4. Lysine, tyrosine and cystine became increasingly unavailable for absorption from the small intestine of sheep with increasing levels of aldehyde. At the lower level (333 mmol/kg) the proportions of the amino acids that were unavailable were 0.192, 0.051 and 0.123 respectively. At the higher level of formaldehyde (667 mmol/kg) the corresponding values were 0.335, 0.201 and 0.432 respectively. Leucine was not made unavailable with formaldehyde.5. The proportions of lysine, tyrosine and leucine that were unavailable were higher, on a molar basis, after treatment of the proteins with the dialdehydes glutaraldehyde and glyoxal than after treatment with formaldehyde. However, the extent of protein protection provided by the dialdehydes in the rumen, measured using an in vitro procedure, was lower.

1. Six 45 kg pigs with re-entrant ileal cannulas were used in two 3 × 3 Latin-square design experiments to study the site of absorption of protein and amino acids. Semi-purified diets containing soya-bean meal (SBM), rapeseed meal (RSM) or no protein source (protein-free) were offered at the rate of 1 kg dry matter/d.2. Flow-rates of ileal contents for 24 h collection periods, corrected for recovery of marker, were 3135, 3127 and 1243 ml (SE 390) for SBM, RSM and protein-free diets respectively.3. Amounts of dry matter digested in the small intestine were 730, 669 and 809 g/d for SBM, RSM and protein-free diets respectively, all values being significantly different (P < 0·001).4. Nitrogen intakes were 32·6, 29·9 and 5·9 g/d, and amounts digested in the small intestine were 25·7, 20·2 and 1·6 g/d for SBM, RSM and protein-free diets respectively, all values being significantly different (P < 0·001). Amounts digested in the large intestine were 2·6, 3·7 and 0·7 g/d.5. Total amino acid intakes and amounts collected at the ileum and in the faeces were (g/d): SBM, 177, 24 and 18; RSM, 149, 28 and 22; protein-free 3, 9 and 12. Digestibility in the small intestine was higher for SBM than RSM for seventeen of the eighteen amino acids estimated. Greater quantities of arginine, methionine, cystine and tyrosine were voided in the faeces than passed through the ileal cannulas for pigs receiving the SBM and RSM diets. For those receiving the protein-free diet this was true for each amino acid except proline.6. Significant differences were found between all diets in the concentration of some amino acids in ileal and faecal amino-N, and endogenous protein secretions did not mask the differences between diets.7. Differences in digestibility between SBM and RSM were greater at the ileum than in the faeces. Amino acid fermentation in the large intestine obscured or reduced differences between SBM and RSM.

A study was made of the effect on nitrogen metabolism of increasing the rectal temperature of Brahman cross and British steers on controlled feed intake. When the rectal temperature of the British steers was 1.3 °C higher than that of the Brahman cross steers, they excreted 13.5 g urinary nitrogen per day more than the Brahman cross. When the Brahman cross steers had a rectal temperature 1.3 °C higher than the British, they excreted 8.7 g urinary nitrogen per day more than the British steers. At the same nitrogen intake there was no significant breed difference in the increase in urinary nitrogen per degree rise in rectal temperature. Increasing the rectal temperature had a small effect on appetite but did not affect the apparent nitrogen digestibility; the concentration of nitrogen in the faecal dry matter was higher under the heat treatment. The ambient temperature at which rectal temperatures were increased by 1.3 °C was about 31°C for the British steers and 45° for the Brahman cross steers. At the increased rectal temperature the Brahman cross steers drank significantly more water than the British steers although faecal and urinary water losses were similar.

Quantitative estimates of disappearances of dry matter, nitrogen and amino acids anterior to the mid-point and terminal ileumof the small intestine were made in pigs prepared with re-entrant cannulas and offered six diets containing wheat alone or supplemented with either meat meal, fish meal, peanut meal, soybean meal or sunflower meal. Apparent digestibilities of dietary dry matter and nitrogen were also estimated by faecal measurements. Disappearance of dry matter and nitrogen was greater anterior to the mid-point than at the end of the small intestine and the rectum. Amounts disappearing at each location and between the three locations differed significantly between protein sources. Up to 10% of dietary dry matter and nitrogen disappeared in the large intestine. For lysine, methionine, threonine and valine, absorption was greater anterior than posterior to the midpoint of the small intestine for all diets except that containing only wheat. Measurements made of the disappearance of amino acids anterior to the large intestine indicated that the apparent availability of the majority of ammo acids of peanut meal, followed by soybean meal, was greater than those of meat meal (52 % crude protein) and fish meal (50 % crude protein).
