Viscosity is the key parameter of many biological systems such as live cells. It can be conveniently measured with ‘molecular rotors’ – fluorescent sensors of microviscosity. Here, we investigate one of the most applied molecular rotors BODIPY-C10.

Fluorescent solvatochromic dyes and molecular rotors have attracted considerable attention as fluorogenic probes because of background-free detection of biomolecules in live cells in no-wash conditions.

The fluorescent excited state of a molecular rotor based on the meso-substituted boron-dipyrromethane (BODIPY) core decays rapidly to the ground state via a conical intersection. The fluorescence is strongly increased in viscous solvents, but solvent polarity has only a small effect.

The effect of solvent polarity on the reaction of PCl5 with (Me3Si)2NH has been investigated and it is found that nonpolar solvents promote the formation of Cl3PNSiMe3, whereas the more polar chloroform gives unusual phosphazenes at room temperature.

Norbornene derivatives were validated as probes for cysteine sulfenic acid on proteins and in live cells. Trapping sulfenic acids with norbornene probes is highly selective and revealed a different reactivity profile than the traditional dimedone reagent. The norbornene probe also revealed a superior chemoselectivity when compared to a commonly used dimedone probe. Together, these results advance the study of cysteine oxidation in biological systems.
