Phosphatidylinositol phosphates are key regulators of cell signalling pathways and membrane trafficking in eukaryotic cells, and there is a need for new chemical probes to further understand how they interact with lipid-binding proteins. Here, the synthesis of phosphatidylinositol phosphate analogues containing adamantyl carboxylic ester groups, in place of the natural lipid side chains, is described. These derivatives are considerably more soluble in water than analogues containing other lipid side chains and do not form large aggregates such as liposomes or micelles. These adamantyl analogues bind to known phosphoinositide-binding proteins with similar affinities to native ligands and will facilitate future studies on the substrate specificities of these proteins involving cocrystallisation studies with proteins.

ABSTRACTA low-molecular-weight liquid crystalline amphiphilic compound (Az6Bu) and a liquid crystalline polymethacrylate [P(Az6Bu)] with amphiphilic-mesogenic side-chains are reported in this present paper. A polyethyleneimine chain with 2-hydroxyethyl side-groups was used as the water-soluble spacer. Az6Bu exhibited a lyotropic columnar phase and no thermotropic mesophase. P(Az6Bu) showed thermotropic and lyotropic liquid crystalline properties. P(Az6Bu) formed thermotropic smectic A and C mesophases on the heating and cooling processes. The lyotropic systems consisting of P(Az6Bu) and water exhibited a smectic C lyomesophase with oilystreak and schlieren textures. The layered structures formed in the thermotropic and lyotropic mesophases are composed of a sublayer produced by the anisotropic alignment of aromatic-mesogenic groups and a sublayer of hydrophilic groups.

The combination of chemical cross-linking and mass spectrometry is currently a progressive technology for deriving structural information of proteins and protein complexes. In addition, chemical cross-linking is a powerful tool for stabilizing macromolecular complexes for single particle cryo-electron microscopy. Broad pallets of cross-linking chemistry, currently available for the majority of cross-linking experiments, still rely on the amine-reactive N-hydroxysuccinimide esters targeting mainly N-termini and lysine side chains. These cross-linkers are divided into two groups: water soluble and water insoluble; and research teams prefer one or another speculating on the benefits of their choice. However, the effect of cross-linker polarity on the outcome of cross-linking reaction has never been studied. Herein, we use both polar (bis(sulfosuccinimidyl) glutarate) and non-polar (disuccinimidyl glutarate) cross-linkers and systematically investigated the impact of cross-linker hydrophobicity on resulting distance constraints, using bovine serum albumin as a model protein.

Cell wall changes during ripening have a major effect on fruit texture. The cell walls isolated using phenol-Tris buffer were sequentially extracted to give polysaccharide fractions that contained mainly water-soluble pectin, chelator-soluble (CDTA) pectin, hemicelluloses (0.05 M Na2CO3 followed by 1M and 4M KOH) and cellulose. The fractions were analyzed colorimetrically for uronic acid, total neutral sugar and cellulose contents. The component sugars of each fraction were determined as their alditol acetates by GC. Then was a decrease in the two pectin fractions during ripening. The pectins appear to have arabinan and galactan side chains. Pectic galactose decreases during ripening. The weight of the combined hemicellulose fractions did not change during ripening, nor did the cellulose level. At least two types of arabinan are present. Pectins were found in all cell wall fractions. Nashi cell walls contain a relatively large amount of xylan compared to other fruit.
