Rhodamine B thiohydrazide (RBS) was firstly employed as turn-on fluorescent probe for hypochlorite in aqueous solution and living cells. It exhibits a stable response to hypochlorite from 1.0×10-6to 1.0×10-5M with a detection limit of 3.3×10-7M. The response of this probe to hypochlorite is fast and highly selective compared with other reactive oxygen species (such as.OH,1O2, H2O2) and other common anions (such as X-, ClO2-, ClO4-, NO3-, NO2-, OH-, Ac-, CO32-, SO42-).

We reported here a turn-on fluorescent probe (1) for the detection of cysteine (Cys) by incorporating the recognition unit of 2,4-dinitrobenzenesulfonyl ester (DNBS) to a coumarin derivative. The structure of the obtained probe was confirmed by NMR and HRMS techniques. The probe shows a remarkable fluorescence off-on response (∼52-fold) by the reaction with Cys in 100% aqueous buffer. The sensing mechanism was verified by the HPLC test. Probe 1 also displays high selectivity towards Cys. The detection limit was calculated to be 23 nM. Moreover, cellular experiments demonstrated that the probe is highly biocompatible and can be used for monitoring intracellular Cys.

Hydrazine (N2H4) is one of the most widely used industrial chemicals that can be utilized as a precursor of pesticides, pharmaceutics, and rocket propellant. Due to its biological and environmental toxicity with potential health risks, various sensing tools have been developed. Among them, fluorescence-based molecular sensing systems have been highlighted due to its simple-operation, high selectivity and sensitivity, and biocompatibility. In our recent report, we disclosed a ratiometric type fluorescent probe, called HyP-1, for the detection of hydrazine, which is based on ortho-methoxy-methyl-ether (o-MOM) moiety assisted hydrazone-formation of the donor (D)-acceptor (A) type naphthaldehyde backbone. As our follow-up research, we disclose a turn-on type fluorescent probe, named HyP-2, as the next-generation hydrazine probe. The sensing rational of HyP-2 is based on the o-MOM assisted retro-aza-Henry type reaction. The dicyanovinyl moiety, commonly known as a molecular rotor, causes significant emission quenching of a fluorescent platform in aqueous media, and its cleavage with hydrazone-formation, which induces a significant fluorescence enhancement. The high selectivity and sensitivity of HyP-2 shows practical explicabilities, including real-time paper strip assay, vapor test, soil analysis, and real water assay. We believe its successful demonstrations suggest further applications into a wide variety of fields.

A new 3-hydroxyphthalimide-based turn-on fluorescent probe is designed and synthesized. This probe can be used to determine the presence of Hg2+ ions by fluorescence spectroscope with high selectivity over other metal ions in aqueous solution. The analytical detection limit for Hg2+ is as low as 6.5 × 10−7 M. The recognition mechanism is attributed to Hg2+-promoted carbonothioate group cleavage and a subsequent excited-state intramolecular proton transfer mechanisms.
