Film Thickness-Profile Measurement Using Iterative Peak Separation Structured Illumination Microscopy

The surface and thickness distribution measurement for transparent film is of interest for electronics and packaging materials. Structured illumination microscopy (SIM) is a prospective technique for measuring film due to its high accuracy and efficiency. However, when the distance between adjacent layers becomes close, the peaks of the modulation depth response (MDR) start to overlap and interfere with the peak extraction, which restricts SIM development in the field of film measurement. In this paper, an iterative peak separation algorithm is creatively applied in the SIM-based technique, providing a precise peak identification even as the MDR peaks overlap and bend into one. Compared with the traditional method, the proposed method has a lower detection threshold for thickness. The experiments and theoretical analysis are elaborated to demonstrate the feasibility of the mentioned method.

Long-Term Safety and Efficacy of Orelabrutinib Monotherapy in Chinese Patients with Relapsed or Refractory Mantle Cell Lymphoma: A Multicenter, Open-Label, Phase II Study

Background: In spite of years of effort, Mantle Cell Lymphoma (MCL) remains a clinical challenge. The clinical significance of Bruton's Tyrosine Kinase (BTK) inhibitor has been validated with the approvals by FDA for the treatment of multiple subtypes of NHL including refractory and relapse (r/r) MCL. However, some serious adverse events (AEs) due to poor target selectivity (inhibition of EGFR, TEC, BMX and others), such as diarrhea, sever bleeding and atrial fibrillation, remain as challenges in clinic. Orelabrutinib is a novel, potent irreversible BTK inhibitor with high selectivity for BTK. Results of early clinical study showed that it has excellent safety/tolerability profiles and favorable pharmacokinetic/pharmacodynamic properties. Sustained ~100% BTK occupancy at 24 hours was achieved with once daily dosing regimen of 50 mg and above. In this presentation, we will report an updated analysis of orelabrutinib in Chinese patients with r/r MCL with minimum of 12 cycles of treatment. Aims: To evaluate the sustained efficacy and long-term safety of orelabrutinib in Chinese patients with r/r MCL. Methods: This is an open-label, multicenter, two stages, phase II study. The primary endpoint was objective response rate (ORR) assessed per Lugano criteria (2014). Safety and other efficacy (DOR, PFS, OS) evaluations were chosen as secondary endpoints. Results: A Total 106 patients, were enrolled in this study with median follow up time of 15.0 months. 79.2% of the patients were male and median age of 62.0 years old. Most patients were at advanced stage (73.6% were at stage IV and 20.8% were at stage III). According to per protocol analysis, 87 (87.9%) patients achieved ORR and 93.9% patients achieved disease control. The median duration of response (DOR) was not reached, the DOR rate at 12 months was 73.7%, as expected the median PFS and OS were not reached, the PFS and OS rates at 12-month were 70.8% and 88.7% respectively. Comparing to the results of previous analysis, the CR rate, by conventional CT method, increased to 27.4% and it was expected a higher rate of in depth response may occur with prolonged treatment. Further analysis showed orelabrutinib was efficacious in all subgroups (age, gender, status, stage, prior therapy, etc.). Orelabrutinib demonstrated excellent safety profile in r/r MCL patients. The frequently reported treatment related adverse events (TRAE) were primarily hematological toxicities including thrombocytopenia, neutropenia, leukopenia, and hypertension. The frequently reported grade 3 or higher AEs of any cause was thrombocytopenia . No treatment related ≥grade 3 GI and cardio toxicity, nor severe bleeding, were observed. Of the 106 patients, 32 experienced serious AEs; and 17 of them, mainly hematological toxicities and / or infections were treatment-related. Comparing to the safety data of median follow up of 10.5 months, there was only a mild increase of adverse events rate after extended treatments; the safety profiles were essentially the same. These results suggested that safety events primarily occurred during early treatment and it appeared less eventful with orelabrutinib continue treatment. Conclusion: Orelabrutinib showed continuous efficacious in treating patients with r/r MCL. In addition, orelabrutinib is safe and well tolerated with no treatment related grade 3 or higher diarrhea, atrial fibrillation/flutter or severe bleeding in this study. Results of prolong treatment expected to produce a higher rate of in depth response without altering its safety profiles support orelabrutinib being a better selection for BTKi therapy. The improved safety as a resulting of high target selectivity, and the convenience of daily dosing regimen provides orelabrutinib as preferred therapeutic choice for B cell malignancy. Identification of Plastic Properties From Conical Indentation Using a Bayesian-Type Statistical Approach

The plastic properties that characterize the uniaxial stress–strain response of a plastically isotropic material are not uniquely related to the indentation force versus indentation depth response. We consider results for three sets of plastic material properties that give rise to essentially identical curves of indentation force versus indentation depth in conical indentation. The corresponding surface profiles after unloading are also calculated. These computed results are regarded as the “experimental” data. A simplified Bayesian-type statistical approach is used to identify the values of flow strength and strain hardening exponent for each of the three sets of material parameters. The effect of fluctuations (“noise”) superposed on the “experimental” data is also considered. We build the database for the Bayesian-type analysis using finite element calculations for a relatively coarse set of parameter values and use interpolation to refine the database. A good estimate of the uniaxial stress–strain response is obtained for each material both in the absence of fluctuations and in the presence of sufficiently small fluctuations. Since the indentation force versus indentation depth response for the three materials is nearly identical, the predicted uniaxial stress–strain response obtained using only surface profile data differs little from what is obtained using both indentation force versus indentation depth and surface profile data. The sensitivity of the representation of the predicted uniaxial stress–strain response to fluctuations increases with increasing strain hardening. We also explore the sensitivity of the predictions to the degree of database refinement.

Individual Differences in Responsiveness to Acupuncture: An Exploratory Survey of Practitioner Opinion

Background: Previous research has considered the impact of personal and situational factors on treatment responses. This article documents the first phase of a four-stage project on patient characteristics that may influence responsiveness to acupuncture treatment, reporting results from an exploratory practitioner survey. Methods: Acupuncture practitioners from various medical professions were recruited through professional organisations to complete an online survey about their demographics and attitudes as well as 60 questions on specific factors that might influence treatment. They gave categorical (“Yes”, “No”, and “Don’t know”) and free-text responses. Quantitative and qualitative (thematic) analyses were then conducted. Results: There were more affirmative than negative or uncertain responses overall. Certain characteristics, including ability to relax, exercise and diet, were most often considered relevant. Younger and male practitioners were more likely to respond negatively. Limited support was found for groupings between characteristics. Qualitative data provide explanatory depth. Response fatigue was evident over the course of the survey. Conclusions: Targeting and reminders may benefit uptake when conducting survey research. Practitioner characteristics influence their appreciation of patient characteristics. Factors consistently viewed as important included ability to relax, exercise and diet. Acupuncture practitioners may benefit from additional training in certain areas. Surveys may produce more informative results if reduced in length and complexity.

Effect of Properties and Turgor Pressure on the Indentation Response of Plant Cells

The indentation of plant cells by a conical indenter is modeled. The cell wall is represented as a spherical shell consisting of a relatively stiff thin outer layer and a softer thicker inner layer. The state of the interior of the cell is idealized as a specified turgor pressure. Attention is restricted to axisymmetric deformations, and the wall material is characterized as a viscoelastic solid with different properties for the inner and outer layers. Finite deformation, quasi-static calculations are carried out. The effects of outer layer stiffness, outer layer thickness, turgor pressure, indenter sharpness, cell wall thickness, and loading rate on the indentation hardness are considered. The calculations indicate that the small indenter depth response is dominated by the cell wall material properties, whereas for a sufficiently large indenter depth, the value of the turgor pressure plays a major role. The indentation hardness is found to increase approximately linearly with a measure of indenter sharpness over the range considered. The value of the indentation hardness is affected by the rate of indentation, with a much more rapid decay of the hardness for slow loading, because there is more time for viscous relaxation during indentation.

The objective of the work was the evaluation of the soybean no-till system as a mean of reducing production cost without affecting the crop yield also toperform an economical analysis comparing this system with the traditional one. There was evaluated the performance of three different furrow opener under no-till system; the results showed that the better crop response was obtained with the chisel type furrow opener compared with the double disc. Also was tested the effect of planting depth and seed density over the effect of final stablished plant, these results showed that it is necessary to increase by 15% the seed dendity compared with the traditional system and the better planting depth response was obtained between three to four centimeters. The economical analysis showed that with the no-till system was possible to obtain a higher marginal return rate of investment in the order of 127% compared with 105% of the traditional system.

Research Data Management as a “wicked problem”

Purpose – The purpose of this paper is to explore the usefulness of the concept to thinking about Research Data Management (RDM). The concept of “wicked problems” seeks to differentiate very complex, intractable challenges from tamer issues where approaches to problem solving are well-understood. Design/methodology/approach – The paper is based on and co-authored by a collaboration of practitioners from libraries, information technology and research administration, with facilitators from the Sheffield Information School. Participants worked together in two-day-long workshops to understand the wicked problem concept and advice on leadership in wicked problem contexts. Findings – Participants concurred that RDM had many features of a wicked problem and most of Grint’s advice on leadership for wicked problems also resonated. Some elements of the issue were simple; participants were optimistic about improving the situation over time. Participants were resistant to the more negative or fatalistic connotations of the phrase “wicked problem”. Viewing RDM as a wicked problem is an interesting way of looking at it as a challenge for support professionals. Practical implications – The notion of a wicked problem is a generative concept that can be usefully added to professional vocabulary. Originality/value – The paper captures an in-depth response from practitioners to the notion of wicked problems as a lens for examining RDM.
