Response of sunflower germplasms viz. cultivated sunflower H. annuus and two breeding lines H. annuus x T. rotundifolia and H. annuus x V. encelioides developed after wide hybridization were used for identification of drought tolerant sunflower genotypes at the seedling growth stage. Three water stress levels of zero (control), -0.4, and -0.8 MPa were developed using polyethyleneglycol-6000 (PEG-6000). Physiological and biochemical stress determining parameters such as root and shoots length, fresh weight, antioxidant enzyme activities (superoxide dismutase (SOD), catalase (CAT), guaiacol peroxidase (GPO), ascorbate peroxidase (APX) and antioxidant metabolite content (total antioxidant capacity, total phenols and total flavonoids content) were compared between seedlings of all three genotypes. Results revealed that sunflower genotypes have similar responses at two osmotic potentials for shoot and root length and fresh weight. The data also showed that drought stresss could induce oxidative stress, as indicated by the increase level of ascorbate peroxidase and guaiacol peroxidase at -04 MPa in H. annuus cv 1114. Although the activity of ascorbate peroxidase and guaiacol peroxidase was differentially influenced by drought, the changes of antioxidant enzyme activities such as catalase, superoxide dismutase, guaiacol peroxidase, and ascorbate peroxidase subjected to drought stress follow a similar pattern in both breeding lines, indicating that similar defense systems might be involved in the oxidative stress injury in sunflowers. Increase in content of phenols and flavonoids were detected for all three genotypes under stress, which showed that these were major antioxidant metabolites in scavenging cellular H2O2.

The Impact

Phytotron experiments were conducted to examine the impact of elevated atmospheric CO2 level (750 μmol mol−1) on the drought tolerance of winter barley (Petra), durum wheat (Mv Makaroni) and spring oat (Mv Pehely) varieties. Under drought stress conditions, the durum wheat variety was found to be unaffected by CO2 enrichment, as neither the biomass or grain yield nor the antioxidant enzyme activities changed compared to those at ambient CO2. Despite the fact that the spring oat variety had similar grain yield loss due to drought at both CO2 levels, it exhibited reduced antioxidant enzyme activities under less severe drought, indicating a slightly increased tolerance to drought. Winter barley, which exhibited an extremely positive reaction to CO2 enrichment at the control water supply level, also showed increased drought tolerance in response to high CO2. It had low glutathione reductase, glutathione-S-transferase and ascorbate peroxidase activities even at the most severe drought stress levels, while it could also fully compensate for the negative effects of drought on biomass and grain yield parameters when grown at elevated CO2.

Soybean is one of the most profitable advanced crops in agricultural production in Ukraine and the world as a whole. Therefore, studies of means of regulation and increase in the adaptive capacity of soybeans in symbiosis with nodule bacteria under the action of unfavourable environmental factors are relevant and should be aimed at the use of complex bacterial compositions involving modern nanotechnological approaches. Nanocarboxylates of ferrum, molybdenum and germanium metals were used as components of rhizobia inoculation suspension for soybean seed treatment to study the effectiveness of their complex effect on the regulation of the activity of the key antioxidant enzyme superoxide dismutase in plants under drought. Various symbiotic systems were used, which included soybean plants and inoculation suspensions based on the active, virulent Tn5-mutant Bradyrhizobium japonicum B1-20 by adding nanoparticles of ferrum, germanium and molybdenum carboxylates to the culture medium in a ratio of 1: 1000. Citric acid was the chelator. A model drought lasting 14 days was created during the period of active fixation of atmospheric molecular nitrogen by root nodules of soybeans in the budding and flowering stages, by means of controlled watering of plants to 30% of the total moisture content. In the stage of bean formation, watering of plants was resumed to the optimal level – 60% of the total moisture content. The control was soybean plants, the seeds of which were inoculated with a suspension of rhizobia without the addition of chelated metals. The following research methods were used in the work – microbiological, physiological and biochemical. According to the results, it was found that when nanoparticles of carboxylates of ferrum, molybdenum and germanium were added to the inoculation suspension of rhizobia, there was an increase in superoxide dismutase activity in root nodules and a decrease in soybean leaves under optimal water supply conditions of plants. This indicates the initial changes in the activity of the antioxidant enzyme in these symbiotic systems, induced by the influence of chelated metals in combination with the rhizobia of the active Tn5-mutant B. japonicum B1-20. Prolonged drought induced an increase in the overall level of superoxide dismutase activity in soybean nodules and leaves, compared to plants grown under optimal watering conditions. The symbiotic system formed by soybeans and B. japonicum with molybdenum carboxylate nanoparticles was the most sensitive to long-term drought exposure, compared to two other soybean-rhizobial symbioses using ferrum and germanium nanocarboxylates. This was manifested in the unstable reaction of the enzyme to the action of drought – suppression or intensification of the level of its activity in the root nodules and leaves of soybeans inoculated with rhizobia containing molybdenum carboxylate nanoparticles. In symbiotic systems with the participation of germanium and ferrum nanocarboxylates, slight changes were revealed in superoxide dismutase activity in root nodules and leaves of plants during drought and restoration of enzyme activity to the level of plants with optimal watering after water stress. It is concluded that the addition to the culture medium of rhizobia Tn5-mutant B1-20 of nanocarboxylates of germanium or ferrum is an effective means of regulating the activity of the antioxidant enzyme superoxide dismutase in soybean root nodules and leaves, which can contribute to an increase in the protective properties and adaptation of plants to the action of dehydration.

Studies were carried out in order to determine the presence of damaged intracellular proteins in sperm from water buffalo as a result of toxic action of free radicals and their attitude towards the vitality of the gametes. Experiments were performed with thawed semen samples as the trial has been added antioxidant enzyme superoxide dismutase. It was found that added to the test sample exogenous antioxidant makes inoffensive much of the available free radicals in sperm, respectively reduces the amount of damaged intracellular proteins and helps protect the vitality of sperm.

Summary This study was conducted to compare and estimate the genetic variability among durum wheat lines in response to drought according to their grain yields and physiological traits. The use of fluorescence parameters as drought selection criteria for durum wheat was investigated in a population of 249 recombinant inbred lines (RILs) derived from a cross between two cultivars of durum wheat: ‘Svevo’ and ‘Kofa’. The durum wheat RILs considered were analyzed to determine the relationship between their grain yields and chlorophyll fluorescence parameters at the grain filling stage under drought stress (rainfed) and supplemental irrigation (irrigated) conditions during the 2004 and 2005 seasons at the ICARDA center. The results obtained show that the durum wheat line performance was influenced by drought stress, indicating significant differences between the grain yields and fluorescence parameters recorded under drought rainfed (RF) and irrigation (IR) conditions. Significant differences between the experimental groups of durum wheat RILs (P<0.05) were only recorded under drought conditions, with the exception of leaf water potential (Lwp). Under such conditions, nearly all the parameters examined were significantly increased in the high-yielding group (with the exception of the Que parameter), thus revealing the genetic variability of the durum wheat lines considered in response to drought stress. The potential quantum efficiency of photosystem II (Fv/Fm) was found to be positively associated with the grain yield parameter. The mean values of Fv/Fm in both the high- and low-yielding groups significantly dropped under drought stress (0.71 and 0.68, respectively) compared to the Fv/Fm values recorded under irrigated conditions (0.80 and 0.81). Under drought conditions, slopes were highly significant (P<0.001) nearly for all the fluorescence parameters examined (with the exception of CHLSPAD) compared to those recorded under irrigation conditions. It was concluded that chlorophyll content (SPAD), F0, Fm, Fv, Fv/Fm, Lwp, and Que could be used as additional indicators in screening wheat germplasm for drought tolerance.
