Genes that exhibit allelic expression imbalance and imprinted genes play important roles in the survival of the embryo and postnatal growth regulation. In this study, the porcine oxysterol-binding protein-related 5 (OSBPL5) gene was examined, and the 2140G>A mutation (rs318687202) was found in its coding region by a comparison of Laiwu and Landrace pigs. By allele-specific expression analysis based on a specific single-nucleotide polymorphism (SNP), the imprinting status of OSBPL5 gene in skeletal muscle from both neonate and adult pigs was determined. The results showed that the OSBPL5 was paternally imprinted in skeletal muscle from adults but biallelically expressed with predominantly maternal imprinting in neonates. The distribution of the 2140G>A SNP in four pig populations was analyzed, which showed that GG genotype was dominant in Duroc and Dapulian populations, whereas the AG genotype was dominant in Junmu-1 and Laiwu populations. Pigs with the GG genotype had significantly larger litters and greater cannon bone circumferences but a lower average daily gain than pigs with the AA genotype. In conclusion, we determined the difference in the allelic expression of OSBPL5 between adult and neonate pigs and identified an SNP in its coding region that is associated with production traits.

Prolactin plays an important regulatory function in mammary gland development, milk secretion, and expression of milk protein genes. Hence the PRL gene is a potential genetic marker of production traits in dairy cattle. The gene was mapped on chromosome 23 by Hallerman et al. (1988). It consists of 5 exons and four introns (Camper et al. 1984) encoding the 199-amino-acid mature protein (Wallis 1974). On the basis of sequence analysis of four different cDNA clones, seven possible nucleotide substitutions were described by Sasavage et al. (1982). One of them, recognized by RsaI endonuclease, has become a popular genetic marker used for genetic characterization of cattle populations by means of PCR-RFLP (Mitra et al., 1995). Two allelic variants (B and b) have been distinguished at the DNA level, based on RsaI polymorphism in the third exon of the coding region. It has been suggested that prolactin alleles correlate with milk yield (Lewin et al., 1992).

Cellular retinol-binding protein 2 (CRBP2), a vitamin A binding protein expressed specifically in small intestinal villus absorptive cells, plays a pivotal role in the intestinal vitamin A absorption, transport, and metabolism pathways. In this study, we cloned the entire coding region of chicken CRBP2 gene. The amplified fragment contains entire coding region sequence with 408 nucleotides, which putatively codes 135 AA. By comparing nine vertebrates, the homology of nucleotide sequences is from 52.3% to 99.8%, while the similarity of AA sequence ranged from 72.4% to 99.3%. Results showed that the CRBP2 gene was conservative among different animal species. This work constructed the basis for further research on the molecular nature and genetic markers of CRBP2 for improving egg production traits in chicken.

DGAT1 polymorphism in Riverine buffalo, Swamp buffalo and crossbred buffalo

This Research Communication describes the polymorphisms in the coding region of DGAT1 gene in Riverine buffalo, Swamp buffalo and crossbred buffalo, and associations between polymorphisms and milk production performance in Riverine buffalo. Two polymorphisms of DGAT1were identified, located in exon 13 and exon 17, respectively. The distribution of the genotypes of the two SNP loci in different buffalo population varied, especially the polymorphism located in exon 13 which was not found in the Swamp buffalo. Moreover, SNP located in exon 17 was a nonsynonymous switch resulting in the animo acid sequence changed from an arginine (Arg) to a histidine (His) at position 484. Both SNPs were in Hardy–Weinberg equilibrium, and the polymorphism of g.8330T>C in the exon 13 was significantly associated with peak milk yield, total milk yield and protein percentage. The C variant was associated with an increase in milk yield and peak yield but less in protein percentage compared with the T variant. The polymorphisms of g.9046T>C in exon 17 were significantly associated with fat percentage, in that the buffaloes with TT genotype had a significantly higher fat percentage than those with CC genotype. These findings reveal the difference in the genetic evolution of the DGAT1 between Riverine buffalo and Swamp buffalo, and provide evidence that the DGAT1 gene has potential effects for Riverine buffalo milk production traits, which can be used as a candidate gene for marker-assisted selection in buffalo breeding.

Testis is the primary organ of the male reproductive tract in mammals that plays a substantial role in spermatogenesis. Improvement of our knowledge regarding the molecular mechanisms in testicular development and spermatogenesis will be reflected in producing spermatozoa of superior fertility. Evidence showed that N6-Methyladenosine (m6A) plays a dynamic role in post-transcription gene expression regulation and is strongly associated with production traits. However, the role of m6A in bovine testis has not been investigated yet. In this study, we conducted MeRIP-Seq analysis to explore the expression profiles of the m6A and its potential mechanism underlying spermatogenesis in nine bovine testes at three developmental stages (prepuberty, puberty and postpuberty). The experimental animals with triplicate in each stage were chosen based on their semen volume and sperm motility except for the prepuberty bulls and used for testes collection. By applying MeRIP-Seq analysis, a total of 8,774 m6A peaks and 6,206 m6A genes among the studied groups were identified. All the detected peaks were found to be mainly enriched in the coding region and 3′- untranslated regions. The cross-analysis of m6A and mRNA expression exhibited 502 genes with concomitant changes in the mRNA expression and m6A modification. Notably, 30 candidate genes were located in the largest network of protein-protein interactions. Interestingly, four key node genes (PLK4, PTEN, EGR1, and PSME4) were associated with the regulation of mammal testis development and spermatogenesis. This study is the first to present a map of RNA m6A modification in bovine testes at distinct ages, and provides new insights into m6A topology and related molecular mechanisms underlying bovine spermatogenesis, and establishes a basis for further studies on spermatogenesis in mammals.

Abstract Gremlin (GREM1, GREM2) genes are the known bone morphogenetic proteins (BMPs) inhibitors, but their genetic diversity in animal species remains unknown. The current study was conducted to investigate single nucleotide polymorphisms (SNPs) in chicken GREM1 and GREM2 genes, and their association with egg production traits using polymerase chain reaction-single strand conformation polymorphism (PCR-SSCP) and deoxyribonucleic acid (DNA) sequencing. The results discovered novel SNPs and, among these variations, C/T transition at position 436 in exon 1 of the GREM1 gene leads to synonymous substitution of amino acids, and T/C transition at position 690 in the coding region of the GREM2 gene leads to a non-synonymous substitution of amino acids (valine acid 114-to-alanine acid). Association analysis established that at the age of 43, 57 and 66 wks, hen-house egg production (HHEP) was more highly significantly associated (P<0.05) with the AA genotype in the GREM1 gene. In the GREM2 gene, the TC genotype was remarkably linked with higher HHEP at the age of 30, 57 and 66 wks. Our results provide evidence that the GREM1 and GREM2 genes have potential effects on HHEP in chickens. SNPs determined in this work may be utilised as favourable potential DNA markers for improving of egg-laying performance traits.

Background: The avian prolactin gene is highly conserved, located on chromosome number 2 and most sequence polymorphisms occurs in the 5’ flanking region, 3’ flanking region, and the coding region of signal peptide. The present study was aimed at the identification of SNP C-2402T of prolactin gene and its association with production traits in White Leghorn chicken. Methods: A total of 200 birds of White Leghorn were selected from All India Co-ordinated Research Project on Poultry improvement (AICRP) farm, Mannuthy. Genomic DNA was isolated from venous blood. Polymerase chain reaction (PCR) followed by restriction fragment length polymorphism (RFLP) analysis was done to identify the SNP C-2402T of prolactin gene. Result: All the birds were observed with the same genotype CC and the frequency of the C allele was one.

The most common sheep breeds of Egypt are Ossimi, Rahmani, and Barki breeds. The latter one is well adapted to the challenging desert environment, characterized by food shortage and a high temperature fluctuation. Growth performance of Barki sheep has an important economic value in terms of minimizing the shortage of mutton meat in Egypt. Further, milk production is of great importance for feeding newborn lambs. Eight candidate genes, recently associated with production traits in different breeds, were used to study the effect of genotype on lamb growth and ewe milk traits. The examined genes were LEP, IGF1, DGAT1, STAT5A, PRL, CSN1S2, GHR, and GHRHR, of which one representative single nucleotide polymorphism (SNP) located in the coding region was selected for genotyping. Data from 251 Barki sheep were used in this study. Association analysis between SNPs and lamb growth traits identified rs420693815 of the LEP gene to be significantly associated with weaning weight and average daily gain. In ewes, significant effects on milk yield and composition have been estimated for LEP (rs420693815), STAT5A (rs161082816), PRL (rs422713690), and GHRHR (rs414991449). The results indicated that these genes might be considered as interesting candidates for further investigations to improve growth and milk performance in Barki sheep.
