Avoparcin and monensin effects on the digestion of nutrients in dairy cows fed a mixed diet

An in vivo study was carried out to evaluate the effect of avoparcin on fiber, starch and nitrogen (N) digestion compared with monensin. Three dairy lactating cows with ruminal, duodenal and ileal cannulas were used in a 3 × 3 Latin square design. Cows were fed three diets; the control diet had no additive, and the others contained 33 ppm of either avoparcin or monensin-sodium. There were no treatment effects on ruminal pH and total volatile fatty acid levels; molar proportion of propionate tended to be higher (27.9–29.9 vs. 23.7%) and of butyrate tended to be lower (9.9–12.9 vs. 15.4%) for the antibiotic-supplemented diets. Also, both additives decreased ruminal ammonia N concentration (77.2–105.6 vs. 164.1 mg L−1). The supply of avoparcin or monensin reduced the ruminal breakdown of fiber (36.1 and 37.1 vs. 45.9%) starch (57.4 and 54.7 vs. 63.6%), and dietary N (59.5 and 53.6 vs. 67.3%); efficiency of bacterial protein synthesis was unchanged. Passage of bacterial N to the small intestine was not modified; flows of fiber, starch, nonammonia N and dietary N were improved by the both additives. In comparison with the control, monensin caused a greater quantity of glucose (2100 vs. 1500 g d−1) and total amino acids (AA) (1381 vs. 1047 g d−1) to be digested in the small intestine; avoparcin had a less pronounced effect on the intestinal absorption of glucose (1700 g d−1) and AA (1152 g d−1). Only monensin increased the plasma glucose levels (3.6 vs. 3.1 mM). Both additives did not affect total tract digestibilities of organic matter, fiber, starch, and N. Postruminal digestion of fiber was greater (16.0 vs. 7.3%) for antibiotic-supplemented diets, compensating for the lower forestomach digestion. In comparison with monensin dietary avoparcin had a less pronounced effect on the alteration of nutrient digestion. Key words: Avoparcin, monensin, rumen, intestines, amino acids, digestion, cattle

Four castrated pigs, each fitted with a re-entrant cannula in the terminal ileum, were used to study digestion in the small intestine. A nitrogen-free diet was used for the estimation of metabolic nitrogen and amino acids. Faba beans, as the sole source of dietary protein, were used raw or after autoclaving for 30 or 60 min. The four diets were fed to the pigs in a 4 × 4 latin square experiment. The pigs were fed each diet for 6 days prior to a 24-hr collection of total ileal contents. Autoclaving of faba beans had no significant effect on digestibility of dry matter, gross energy, nitrogen and individual amino acids except arginine, which was significantly increased. The intestinal uptake of arginine was the highest and of cystine the lowest in all faba bean diets. It was concluded that autoclaving faba beans had no beneficial effect on the digestion of nutrients in the small intestine of the pig.

1. Six 45 kg pigs with re-entrant ileal cannulas were used in two 3 × 3 Latin-square design experiments to study the site of absorption of protein and amino acids. Semi-purified diets containing soya-bean meal (SBM), rapeseed meal (RSM) or no protein source (protein-free) were offered at the rate of 1 kg dry matter/d.2. Flow-rates of ileal contents for 24 h collection periods, corrected for recovery of marker, were 3135, 3127 and 1243 ml (SE 390) for SBM, RSM and protein-free diets respectively.3. Amounts of dry matter digested in the small intestine were 730, 669 and 809 g/d for SBM, RSM and protein-free diets respectively, all values being significantly different (P < 0·001).4. Nitrogen intakes were 32·6, 29·9 and 5·9 g/d, and amounts digested in the small intestine were 25·7, 20·2 and 1·6 g/d for SBM, RSM and protein-free diets respectively, all values being significantly different (P < 0·001). Amounts digested in the large intestine were 2·6, 3·7 and 0·7 g/d.5. Total amino acid intakes and amounts collected at the ileum and in the faeces were (g/d): SBM, 177, 24 and 18; RSM, 149, 28 and 22; protein-free 3, 9 and 12. Digestibility in the small intestine was higher for SBM than RSM for seventeen of the eighteen amino acids estimated. Greater quantities of arginine, methionine, cystine and tyrosine were voided in the faeces than passed through the ileal cannulas for pigs receiving the SBM and RSM diets. For those receiving the protein-free diet this was true for each amino acid except proline.6. Significant differences were found between all diets in the concentration of some amino acids in ileal and faecal amino-N, and endogenous protein secretions did not mask the differences between diets.7. Differences in digestibility between SBM and RSM were greater at the ileum than in the faeces. Amino acid fermentation in the large intestine obscured or reduced differences between SBM and RSM.

AbstractThe effects of anAspergillus oryzaeextract containing alpha-amylase activity (Amaize™, Alltech Inc., Nicholasville, KY) were examinedin vivoandin vitro. A lactating cow study employed 20 intact and four ruminally fistulated Holstein cows in a replicated 4 × 4 Latin-square design to examine the effects of four concentrations of dietary Amaize™ extract on milk production and composition, ruminal fermentation and serum metabolite concentrations. The treatment diets contained 0, 240, 480 or 720 alpha-amylase dextrinizing units (DU) per kg of total mixed ration (TMR) (dry-matter basis). The supplemental alpha-amylase increased the yields of milk (P= 0·02), fat (P= 0·02) and protein (P= 0·06) quadratically. The maximum milk yield was obtained when 240 DU per kg of TMR were offered. Ruminalin situstarch disappearance was not affected by alpha-amylase supplementation in lactating cows or ruminally cannulated steers. Supplemental alpha-amylase extract reduced the molar proportion of propionate in the rumen of steers (P= 0·08) and lactating cows (P= 0·04), and in rumen-simulating cultures (P= 0·04). The supplement also increased the molar proportions of acetate (P= 0·06) and butyrate (P= 0·05), and the serum beta-hydroxybutyrate (P= 0·01) and non-esterified fatty acid (P= 0·03) concentrations in lactating cows. The improvements in milk production appear to be a consequence of the effects of alpha-amylase on ruminal fermentation and the potential changes in nutrient metabolism that result from them. We conclude that supplemental alpha-amylase may be given to modify ruminal fermentation and improve milk and component yield in lactating Holstein cattle.

The effect of two levels of fishmeal substitution (50 (FM1) and 150 (FM2) g/kg) of a grass silage control diet (C) on the rumen digestion of organic matter and nitrogen, and the small intestinal disappearance of amino acids was examined in young growing cattle each equipped with simple PVC cannulas in the dorsal sac of the reticulo-rumen, the proximal duodenum and the terminal ileum. The silage was a primary growth of perennial ryegrass (Lnlium pevenne) (+formic acid) with a total N content of 22 g/kg dry matter (DM) (diet C). Fishmeal substitution increased this to 26 (diet FM1) and 34 (diet FM2) g/kg DM. On diets C and FM1, approximately 0.71 of digestible organic matter intake was apparently digested in the rumen, but this was significantly (P < 0.05) reduced on diet FM2 (0.60). Whilst duodenal flows of non-ammonia N and total amino acids were significantly (P < 001) increased at the highest level of fishmeal inclusion only, the synthesis of microbial N was significantly (P < 0.001) reduced by fishmeal inclusion, and feed N degradability declined progressively in response to increased fishmeal. Both levels of fishmeal addition caused a significant (P< 0.05) reduction in the fractional outflow rate of water from the rumen, and on the highest level of fishmeal significant (P< 0.05) increases in rumen ammonia concentration and rumen propionate molar proportions were observed. The net effect of the highest level of fishmeal substitution was to increase amino acid absorption from the small intestine by 0.47 compared with the control diet (P< 0.05), but due to an elevated ileal flow of amino acid no such effect was detected at the lowest level of fishmeal substitution. Composition of the absorbed amino acid fraction was relatively unaffected by the treatments imposed, despite large changes in the composition of the duodenal protein. The apparent non-linearity of response to fishmeal substitution is discussed and the amino acid supply findings are compared with the protein retention findings obtained in an earlier study by Gill et al. (1987). By two methods of calculation it was estimated that the amino acid N fraction disappearing from the small intestine was utilized with an efficiency of between 0.51 and 0.53 and no apparent effects due to diet or level of amino acid supply were detected.

Four rumen-fistulated sheep, 62 kg average body weight (BW), were assigned four pelleted diets containing 0 (control), 15, 30 and 45% steam-processed aspen (SPA) in a 4 × 4 Latin square design. All diets were formulated to be isonitrogenous to the control diet which contained 45% ground alfalfa hay as a roughage source. Intake of dry matter (DM), crude protein (CP), crude fiber (CF) and gross energy (GE) per kg BW0.75 were not affected (P > 0.05) by addition of SPA to pelleted sheep diets. However, digestible energy (DE) consumption apparently decreased with SPA diets. Apparent digestibilities of DM and GE significantly (P < 0.05) decreased for the SPA diets; however, CP and CF digestibility coefficients and N retention were not different (P > 0.05) among the four treatments. Ruminal pH for all treatments fell rapidly within 1 h post feeding and remained low to 12 h, whereas ammonia (NH3) levels were significantly (P < 0.05) elevated for all aspen-fed animals, from 1 h through 12 h post feeding. Total VFA concentrations and molar concentrations of acetic, propionic and butyric acids in ruminal fluid were comparatively lower for sheep fed the 45% SPA diet versus those receiving the other treatments. Blood serum glucose concentration was significantly (P < 0.05) depressed when 45% SPA was fed to mature sheep.

The objective of this study was to evaluate the ruminal and intestinal digestion, the ruminal fermentation and the performance of beef cattle fed diets containing Stylosanthes silage (StS) and corn silage (CS). The treatments consisted of diets containing StS with different concentrate levels, based on dry matter (DM) (g/kg): 200 (StS-200), 400 (StS-400), 60 (StS-600), and a Control treatment with 600 g/kg CS and 400 g/kg concentrate (CS-400). In Experiment 1, 32 Nellore bulls, 364.28 ± 2.82 kg bodyweight, were distributed in a complete randomised design with eight replicates. Four Holstein × Zebu bulls (average bodyweight of 429 ± 15 kg) were used in Experiment 2, distributed in a 4 × 4 Latin square design. Nutrient intake was similar between StS-400 and CS-400 (P > 0.05), however, only StS-600 showed the same intake of total digestible nutrients as CS-400 (P < 0.05). Only the animals fed the StS-600 diet had similar (P > 0.05) average daily gain of carcass in comparison with those fed the Control diet. The StS-200 and StS-400 diets showed lower (P < 0.05) total tract digestibility of DM and crude protein than CS-400 (P < 0.05). Ruminal pH values of StS-200 and SE-400 were higher (P < 0.05) than in CS-400. The StS-600 diet showed a similar productive performance to CS in diets with 600 g/kg of concentrate, highlighting the potential use of this legume in diets for beef cattle in tropical conditions.

Possible interactions between the Fusarium toxin deoxynivalenol and lipopolysaccharides on in vivo protein synthesis were investigated in selected porcine tissues. A total of 36 male castrated pigs (initial weight of 26 kg) were used. 24 pigs were fed a control diet and 12 a Fusarium-contaminated diet (chronic oral deoxynivalenol, 3.1 mg/kg diet) for 37 days. Tissue protein synthesis was measured in pigs fed control diet after intravenous infusion of deoxynivalenol (100 µg/kg live weight/h), lipopolysaccharides (7.5 µg/kg live weight/h) or a combination of both compounds on the day of the measurements, while six pigs from the chronic oral deoxynivalenol group were intravenously treated with lipopolysaccharides (7.5 µg/kg live weight/h). Deoxynivalenol challenge alone failed to alter protein synthesis parameters. Fractional protein synthesis rates were exclusively reduced in liver, spleen and small intestine of lipopolysaccharides-treated pigs. Intravenous deoxynivalenol co-exposure enhanced the impacts of lipopolysaccharides on protein synthesis parameters in the spleen and the small intestine to some extent, while a chronic oral pre-exposure with deoxynivalenol relieved its effects in the spleen. Whether these interactions occur in other tissues and under other study conditions, especially toxin doses and route of entry into the body, needs to be examined further.

Abstract The exposure of pigs to heat stress (HS) appears to damage their intestinal epithelia, affecting the absorption of amino acids (AA). Arg is involved in the restoration of intestinal epithelial cells but HS reduces Arg intake. The effect of dietary supplementation with Arg on morphology of intestinal epithelia, AA transporter gene expression, and serum concentration (SC) of free AA in HS pigs were analyzed. Twenty pigs (25.3 ± 2.4 kg BW) were randomly assigned to two dietary treatments: control (0.81% Arg), wheat-soybean meal diet supplemented with L-Lys, L-Thr, DL-Met and L-Trp, and the experimental diet where 0.16% free L-Arg was supplemented to a similar control diet (+Arg). All pigs were individually housed and exposed to HS, fed at libitum with full access to water. The ambient temperature, recorded at 15-min intervals during the 21-d trial, ranged on average from 29.6 to 39.4 °C within the same day. Blood samples were collected on d18 at 1600 h (ambient temperature peak); serum was separated by centrifugation. At the end of the trial, five pigs per treatment were sacrificed to collect samples of mucosa scratched from each small intestine segment. The expression of AA transporters in intestinal mucosa and the SC of AA were analyzed. Villi height was higher (P &lt; 0.01) in duodenum, jejunum, and ileum but the crypt depth did not differ between the control and the +Arg pigs. Supplementation of L-Arg increased the mRNA coding for the synthesis of the cationic AA transporter b 0,+ (P &lt; 0.01) and the neutral AA transporter B 0 (P &lt; 0.05) in duodenum by approximately five-folds and three-folds, respectively, but no effect on mRNA abundance was observed in jejunum and ileum. The supplementation of L-Arg increased serum Arg, His, Met, Thr, Trp, and urea (P &lt; 0.05); tended to increase Val (P &lt; 0.10), but did not affect Ile, Lys, Leu, and Phe. These results indicate that supplementing 0.16% L-Arg to the control diet may help to improve the function of the small intestine epithelium, by increasing the villi height, the abundance of AA transporters, and the SC of most indispensable AA in pigs exposed to HS conditions. However, the lack of effect of supplemental Arg on both Lys SC and weight gain of pigs suggests that increasing the Lys content in the +Arg diet might be needed to improve the performance of HS pigs.

The present study was conducted to evaluate the in vitro and in vivo digestibility of dietary starch and its digestive behaviour on the systemic circulating amino acids (AA) in weaned pigs. Eighteen weanling pigs surgically fitted with a catheter in the jugular vein were randomly assigned to three dietary treatment groups. Sticky rice starch (SRS) was hydrolysed more quickly in vitro (P < 0·05) than maize starch (MS) and resistant starch (RS), and was almost completely hydrolysed within 4 h. The in vivo digestibility of dietary starch in different segments of the small intestine was significantly different. SRS was digested (81·9 %; P < 0·05) in the anterior jejunum, but not more than half of the MS and RS was digested in the same segment of the small intestine. The digestibilities of isoleucine, leucine, methionine, phenylalanine, threonine, tryptophan, valine, alanine, aspartate and serine in the SRS group were higher than in the MS group (P < 0·05), and all nutritionally indispensable and dispensable AA in the SRS group were higher when compared with those in the RS group (P < 0·05). The serum concentrations of nutritionally indispensable AA, proline and serine in the three groups were increased to a peak point within 1·5 h postprandially then decreased gradually; however, the time that serum concentrations of alanine, aspartate, glutamate and glycine in each group increased to a peak point was different. The concentrations of nutritionally indispensable AA, including arginine, cystine, histidine, isoleucine, leucine, methionine, phenylalanine, threonine, tryptophan, tyrosine and valine at 09.30 hours and arginine, cystine, histidine, isoleucine, methionine, phenylalanine, threonine, tryptophan, tyrosine and valine at 13.30 hours in the SRS group were higher than in the MS group (P < 0·05); all nutritionally indispensable AA in the SRS group were higher than in the RS group at 09.30 and 13.30 hours (P < 0·05), respectively. We conclude that dietary starches digested rapidly in vitro have higher digestibility in the anterior small intestine of pigs. Diets containing rapidly digestible starch ameliorate the digestive and absorptive function and regulate AA metabolism to beneficially increase the entry of dietary AA into the systemic circulation in pigs.
