A general purpose controlled release (CR) intra-ruminal device (CRD) containing chromium sesquioxide (Cr2O3) was used to measure mean daily fecal dry matter excretion (FE). The FE of penned sheep, estimated using the CRD, was 1.02 ± 0.04 of actual FE, independent of diet. A single, uniform release rate for the CRD was observed in grazing and penned sheep. SummaryFaecal output of 287 beef cows consisting of Hereford (H × H), Simmental × Hereford (S × H), Friesian × Hereford (F × H) and Brahman × Hereford (B × H) breeding was estimated using controlled release of chromic oxide (Cr2O3). All cows were from 6 to 10 years of age and had grazed on one of three pasture systems in the subtropics of New South Wales since they were 7–8 months of age. These pastures were designated as being of high, medium and low nutritive value. Faecal organic matter output (FOMO) was estimated in each of four periods (early lactation, mid lactation, late lactation, and non-lactating) during one annual production cycle.Herbage mass exceeded 2 t/ha on all pasture systems, but estimates of organic-matter digestibility, nitrogen content and pasture composition indicated large differences in pasture quality (high > medium > low). Live weight of cows averaged 604, 488 and 393 kg live weight on the high, medium and low pastures, respectively. There were significant differences in live weight between genotypes on the medium (B × H > F × H and H × H) and low (S × H ≡ F × H ≡ B × H > H × H) pastures but not on the high pasture).FOMO and dry-matter intake (DMI) differed significantly between genotypes among lactating cows on all pasture systems. Rankings on high pastures were reasonably consistent in all periods: S × H ≡ F × H ≥ B × H ≥ H × H, as were those on low pastures, B × H ≥ S × H ≡ F × H ≥ H × H. Differences on medium pastures were significant only during mid-lactation when B × H ≡ S × H ≡ F × H > H × H. Significant differences between genotypes were observed among non-lactating cows on high pastures only, where H × H ≡ S × H ≡ F × H > B × H. Lactating females had significantly higher FOMO and DMI than non-lactating females when compared across genotypes during early lactation on all pastures.

Thirty-two dairy cows (663 ± 70; kg of BW ± SD) were allotted to a 2 × 2 factorial arrangement of treatments to evaluate the controlled release alkane capsule (CRC; C32 and C36) to estimate digestibility and dry matter (DM) intake in early and late lactation for diets based on low (LC, 30%) or high (HC, 60%) proportions of concentrate. Cows were dosed orally with a CRC into the rumen and digestibility was measured by total collection and estimated from the ratios of alkanes in the diet and faeces. Recoveries of endogenous (C31 and C33) and exogenous (C32 and C36) alkanes were higher and lower, respectively, for LC compared with HC. The effects of diet and the interaction between diet and lactation stage were well estimated (P < 0.05) when measured released rates of C32 and C36 were used to estimate DM digestibility although estimation with both alkanes indicated an effect of lactation stage, which was not observed when using data of measured DM digestibility. The faecal ratio of C31/C32 or C33/C32 showed no diet effect on DM intake estimation while measured DM intake was 22.6% higher for cows fed HC than for those fed LC. In conclusion, estimation of DM intake was inadequate although digestibility of DM was estimated from alkanes contained in the CRC. Key words: Dairy cows, alkane, intake

Background: Venlafaxine HCl is a selective serotonin reuptake inhibitor which is given in the treatment of depression. The delivery of the drug at a controlled rate can be of great importance for prolonged effect. Objective: The objective was to prepare and optimize the controlled release core in cup matrix tablet of venlafaxine HCl using the combination of hydrophilic and hydrophobic polymers to prolong the effect with rate controlled drug release. Methods: The controlled release core in cup matrix tablets of venlafaxine HCl were prepared using HPMC K5, K4, K15, HCO, IPA, aerosol, magnesium sterate, hydrogenated castor oil and micro crystalline cellulose PVOK-900 using wet granulation technique. Total ten formulations with varying concentrations of polymers were prepared and evaluated for different physicochemical parameters such FTIR analysis for drug identification, In-vitro drug dissolution study was performed to evaluate the amount of drug release in 24 hrs, drug release kinetics study was performed to fit the data in zero order, first order, Hixson–crowell and Higuchi equation to determine the mechanism of drug release and stability studies for 3 months as observed. Results: The results of hardness, thickness, weight variation, friability and drug content study were in acceptable range for all formulations. Based on the In vitro dissolution profile, formulation F-9 was considered to be the optimized extending the release of 98.32% of drug up to 24 hrs. The data fitting study showed that the optimized formulation followed the zero order release rate kinetics and also compared with innovator product (flavix XR) showed better drug release profile. Conclusion: The core-in-cup technology has a potential to control the release rate of freely water soluble drugs for single administration per day by optimization with combined use of hydrophilic and hydrophobic polymers.

AbstractDetermination of the optimal fertilization method is crucial to maximize nitrogen use efficiency and yield of different rice cultivars. Side-deep fertilization with controlled-release nitrogen, in conjunction with machine transplanting and subsequent topdressing, was applied to Indica–japonica hybrid rice ‘Yongyou1540’ (YY1540) and indica hybrid rice ‘Tianyouhuazhan’ (TYHZ). Four nitrogen treatments were applied in 2018 and 2019: traditional nitrogen application with quick-release nitrogen (T1), single-dose deep fertilization at transplanting with 100% controlled-release nitrogen (T2), and deep fertilization of 70% controlled-release nitrogen and topdressing of 30% quick nitrogen at tillering (T3), or at panicle initiation (T4). Side-deep fertilization reduced the fertilizer application frequency without causing yield loss, T4 enhanced the yield of YY1540 by increasing the number of productive tillers and number of spikelets per panicle compared with T1, T2 and T3. The yield of TYHZ showed no significant difference among treatments. The T4 treatment decreased the number of tillers at the tilling peak stage and increased the percentage productive tillers and number of differentiated spikelets. Compared with the other treatments, T4 increased dry matter accumulation and leaf area index during panicle initiation and grain ripening, and contributed to enhanced nitrogen uptake and nitrogen utilization in YY1540. On average, nitrogen uptake and utilization in YY1540 were highest in T4, but no significant differences among treatments were observed in TYHZ. Dry matter accumulation and nitrogen uptake from panicle initiation to heading of YY1540 were correlated with number of spikelets per panicle, but no significant correlations were observed for TYHZ. Supplementary topdressing with quick-release nitrogen at the panicle initiation stage was required to increase yield of indica–japonica hybrid rice, whereas single-dose deep fertilization with controlled-release nitrogen is satisfactory for the indica hybrid cultivar.

Whole barley was treated with sodium hydroxide (NaOH) in laboratory trials. Dry matter disappearance from nylon bags in the rumen of whole barley treated with 2.5, 3.5, or 4.9% NaOH for 30 h was 59.6, 72.4, and 93.0%, respectively, compared with 82.2% for untreated ground barley. In a subsequent lactation trial, 24 Holstein cows (eight per treatment) were fed high moisture ground ear corn, high moisture rolled barley or high moisture whole barley treated with 3.5% NaOH. Milk persistencies tended to be greater for cows fed high moisture rolled barley, next for ground ear corn and least for NaOH-treated barley. Milk composition was similar for all treatments. Dry matter intake was greatest for cows fed ground ear corn and lower for those fed the barley diets. Alpha-linked glucose and pH of feces were similar for cows fed ground ear corn and high moisture rolled barley diets, but fecal pH was lower and alpha-linked glucose concentrations three times greater for NaOH-treated barley. Digestibility percents of dry matter, acid detergent fiber and nitrogen were 61.4, 25.3, 64.7 for ground ear corn; 64.4, 38.0, 67.1 for high moisture rolled barley; and 56.8, 43.2, 54.8 for NaOH-treated barley, respectively. Rumen grain turnover estimated by excretion of ytterbium in feces was greatest for NaOH-treated barley (9.09%/h), intermediate for ground ear corn (6.10%/h) and lowest for high moisture rolled barley (4.93%/h). A controlled release system was prepared, it based on UF modified PCC cells in which TEB are loaded into cells. It can control the drug release rate, depress the initial “burst effect”, and was efficacious in controlling wheat powdery mildew.

Five timothy (Phleum pratense L.) cultivars, Clair, Champ, Milton, Climax and Bounty, were harvested at first flower stage during 2 consecutive years and in the regrowth for 1 yr. The forages were each offered as hay to 15 young rams to evaluate voluntary intake. Five animals were used to determine initial harvest dry matter digestibility. The cultivars varied significantly in voluntary intake at first flower stage. The highest intake (g kg−0.75 d−1) averaged for the years was obtained in the early cultivar Champ (65.9), which was 10% more than that of the medium-late cultivar Climax. The cultivars did not differ significantly in dry matter digestibility. In the regrowth, the relative intakes of the cultivars were essentially reversed from that of the initial harvest. The voluntary intake was substantially higher but the differences among forages were less marked. Climax regrowth voluntary intake was higher (P < 0.05) than all cultivars, except Bounty. A 15% range in digestible dry matter intake was present among cultivars. The ranking of the culivars for digestible dry matter was different than for dry matter yield.
