The present study was conducted to optimize extraction process for defatted pitaya seed extract (DPSE) adopting response surface methodology (RSM). A five-level central composite design was used to optimize total phenolic content (TPC), total flavonoid content (TFC), ferric reducing antioxidant power (FRAP), and 2,2′-azino-bis (3-ethylbenzothizoline-6-sulfonic acid (ABTS) activities. The independent variables included extraction time (30–60 min), extraction temperature (40–80 °C) and ethanol concentration (60%–80%). Results showed that the quadratic polynomial equations for all models were significant at (p < 0.05), with non-significant lack of fit at p > 0.05 and R2 of more than 0.90. The optimized extraction parameters were established as follows: extraction time of 45 min, extraction temperature of 70 °C and ethanol concentration of 80%. Under these conditions, the recovery of TPC, TFC, and antioxidant activity based on FRAP and ABTS were 128.58 ± 1.61 mg gallic acid equivalent (GAE)/g sample, 9.805 ± 0.69 mg quercetin equivalent (QE)/g sample, 1.23 ± 0.03 mM Fe2+/g sample, and 91.62% ± 0.15, respectively. Ultra-high-performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UPLC-QTOF/MS) analysis identified seven chemical compounds with flavonoids constituting major composition of the DPSE.

The purpose of this study is to investigate the antioxidant properties and total phenolic content in local fruit wastes. Watermelon rinds, mango and banana peels were selected in the experiment. Two parameters involved were the extraction time and temperature. All samples had undergone drying process in an oven at 40°C for 24 hours before being extracted with 80%, v/v methanol on a hot plate together with magnetic stirrer. The extraction time was ranged from 20 to 40 minutes and the extraction temperature was from 30 to 60°C. Ferric Reducing Antioxidant Power (FRAP) assay and Folin-Ciocalteu reagent were used for antioxidant analysis and phenolic content determination, respectively. UV-Vis spectrophotometer was used to detect the antioxidant activity at 593 nm and 765 nm for phenolic content. Response Surface Method was used to find the best extraction condition. The best extraction conditions were found at extraction temperature and time of 58.93°C and 37.86 minutes, respectively.

The consumption of herbal teas has become popular in recent years due to their attractive flavors and outstanding antioxidant properties. The Five-Golden-Flowers tea is a herbal tea consisting of five famous edible flowers. The effects of microwave-assisted extraction parameters on the antioxidant activity of Five-Golden-Flowers tea were studied by single-factor experiments, and further investigated using response surface methodology. Under the optimal parameters (53.04 mL/g of solvent/material ratio, 65.52 °C, 30.89 min, and 500 W), the ferric-reducing antioxidant power, Trolox equivalent antioxidant capacity, and total phenolic content of the herbal tea were 862.90 ± 2.44 µmol Fe2+/g dry weight (DW), 474.37 ± 1.92 µmol Trolox/g DW, and 65.50 ± 1.26 mg gallic acid equivalent (GAE)/g DW, respectively. The in vivo antioxidant activity of the herbal tea was evaluated on alcohol-induced acute liver injury in mice. The herbal tea significantly decreased the levels of aspartate aminotransferase, total bilirubin, and malonaldehyde at different doses (200, 400, and 800 mg/kg); improved the levels of liver index, serum triacylglycerol, and catalase at dose of 800 mg/kg. These results indicated its role in alleviating hepatic oxidative injury. Besides, rutin, chlorogenic acid, epicatechin, gallic acid, and p-coumaric acid were identified and quantified by high performance liquid chromatography (HPLC), which could contribute to the antioxidant activity of the herbal tea.

This research aimed to optimize the total polyphenol content (TPC) extracted from soybean sprout powder under different experimental parameters, including ethanol concentration (60–100% v/v), extraction temperature (40–80 °C), extraction time (15–150 min), material:solvent ratio (1:4–1:10 g/mL), the number extraction cycles (1, 2 and 3 times), the age of sprout (0–7 days), and the used part of the sprout (cotyledon, hypocotyl, or radicle). The obtained results were used in response surface methodology, in combination with a central composite design, to model the total polyphenol content (TPC) with respect to three variables, including ethanol concentration, extraction temperature, and material:solvent ratio. The experimental conditions for optimal recovery of TPC consisted of ethanol concentration of 88% (v/v), extraction temperature of 59 °C, material:solvent ratio of 1:6.5 g/mL, extraction time of 60 min, and 2 cycles of maceration. In addition, for maximal TPC, the sprout should undergo the germination of 5 days and the radicle fraction should be used. Based on the suggested optimum conditions, the obtained and verified TPC was 19.801 mg genistein (GE)/g dry weight (d.w.). The obtained dried extract also exhibited low antioxidant activity.

This study intends to apply enzyme-assisted extraction method to determine the antioxidant activity of round kumquat peel extract. Firstly, chemical composition of the kumquat peel was determined. Then, single factor test was employed to evaluate the effects of enzyme treatment conditions (enzyme amount and extraction time, ethanol concentration, material:ethanol ratio, extraction time and extraction temperature) on total phenolic content (TPC) and antioxidant activity of the kumquat peel extract. The result showed that the content of phenolic compounds in the kumquat peel achieved 1.3%. The peel extract exhibited the maximum TPC of 335.96±16.79 milligrams of gallic acid equivalents (mg GAE)/g dry matter, 2,2'-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) cation radical (ABTS●+) scavenging activity of 1993.26±99.66 μmol Trolox Equivalent (TE)/g dry matter and ferric reducing antioxidant power (FRAP) value of 3202.86±160.14 μmol TE/g dry matter. This extract was obtained under   enzyme treatment condition including enzyme content of 9 U/g dry matter, enzyme-treating time of 90 min and the extraction condition comprising of kumquat peel: 96% ethanol ratio of 1:40 (w/v), extraction time of 150 min and temperature of 50°C. The results revealed that the kumquat peel extract could be used as a potential natural antioxidant in food and/or pharmaceutical products.

Free radical-induced oxidative stress is the root cause of many diseases, such as diabetes, stress and cardiovascular diseases. The objective of this research was to screen GABA levels, antioxidant activities and bioactive compounds in brown rice. In this study, we first fermented brown rice with different lactic acid bacteria (LABs), and the best LAB was selected based on the levels of GABA in the fermentate. Lactobacillus reuterii generated the highest levels of GABA after fermentation. To ascertain whether germination can improve the GABA levels of brown rice, we compared the levels of GABA in raw brown rice (Raw), germinated brown rice (Germ), fermented brown rice (Ferm) and fermented-germinated brown rice (G+F) to identify the best approach. Then, antioxidant activities were investigated for Raw BR, Germ BR, Ferm BR and G+F BR. Antioxidant activity was calculated using a 2,2-diphenyl-1-picryl hydrazile radical assay, 2,2-azino-bis-(3-ethylene benzothiozoline-6-sulfonic acid) radical assay and ferric-reducing antioxidant power. In Ferm BR, DPPH (114.40 ± 0.66), ABTS (130.52 ± 0.97) and FRAP (111.16 ± 1.83) mg Trolox equivalent 100 g, dry weight (DW), were observed as the highest among all samples. Total phenolic content (97.13 ± 0.59) and total flavonoids contents (79.62 ± 1.33) mg GAE/100 g and catechin equivalent/100 g, DW, were also found to be highest in fermented BR. Furthermore, an untargeted metabolomics approach using ultra-high-performance liquid tandem chromatography quadrupole time of flight mass spectrometry revealed the abundance of bioactive compounds in fermented BR, such as GABA, tryptophan, coumaric acid, L-ascorbic acid, linoleic acid, β-carotenol, eugenol, 6-gingerol, etc., as well as bioactive peptides which could contribute to the health-promoting properties of L. reuterii fermented brown rice.

Seeds of Alpinia blepharocalyx contain high amounts of phenolic and flavonoid compounds with potential antioxidant properties. In this study, the effects of the extraction method on the recovery of phenolic and flavonoid compounds from Alpinia blepharocalyx seeds were investigated. Response surface methodology (RSM) has been used to optimize the extraction conditions of total phenolic and total flavonoid from seeds of Alpinia blepharocalyx. A Box-Behnken design was used to investigate the effects of four independent variables, namely extraction temperature (°C), extraction time (min), (v/m) and ethanol concentration (%) on the responses: total phenolic content (TPC) and yield. The optimal conditions obtained from response RSM were 52.66 % v/v for the solvent composition, 62.34 (°C) for extraction temperature, 34.48/1 (ml/g) for solvent/material ratio and 125.42 (min) for extraction time. The experimental values of TPC, TFC and yield were 39.31±0.05 mgGAE/g, 12.75±0.07 mgCE/g and 6.97±0.05%, respectively.

Optimization of the yield, total phenolic content (TPC), and total antioxidant activities (TAA) of polyphenol concentrates extracted from Solanum macrocarpon leaves was studied using response surface methodology. The process variables investigated included extraction temperature (30, 50, and 70°C), extraction time (2, 4, and 6 h), and dried leaf powder : water ratio (1 : 10, 1 : 20, and 1 : 30 w/v). Box–Behnken design resulted in 15 experimental runs. The results showed the following optimum extraction conditions: temperature, 49.05°C; extraction time, 243 min; leaf powder : water ratio, 1 : 22 w/v. The optimized extraction conditions gave polyphenol concentrate yield, TPC, and TAA values of 24.94%, 421.09 mg GAE/g, and 23.81 mg AAE/g, respectively. Results of the in vitro antioxidant activities of the polyphenol concentrate showed 2, 2-diphenyl-2-picrylhydrazyl hydrate, metal chelating ability, and ferric reducing ability values of 76.78%, 80.22%, and 56.46 mg AAE/g, respectively. The study concludes that the experimental values compared closely with the predicted values, which indicates suitability of the model employed for polyphenol extraction optimization from dried S. macrocarpon leaves.

Mediterranean plant Helichrysum italicum represents a rich source of versatile bioactive compounds with potential benefits for human health. Despite extensive research on the plant’s active constituents, little attention has yet been paid to characterizing the relationship between its intra-specific genetic diversity and metabolite profile. The study aimed to determine metabolic profile of H. italicum ssp. italicum (HII) and ssp. tyrrhenicum (HIT) cultivated on the experimental plantation in Slovenia and to compare the chemical composition of extracts regarding the solvent extraction process. Extracts were prepared upon conventional extract preparation procedures: maceration with 50% methanol or ethanol and cold or hot water infusion and analyzed using High Performance Liquid Chromatography-Diode Array Detection-Electrospray Ionization-Quadrupole Time-of-Flight-Mass Spectrometry (HPLC-DAD-ESI-QTOF-MS). One hundred compounds were identified in the samples, among them several isomers and derivatives were reported for the first time, while caffeoylquinic acids and pyrones were the most abundant. Semi-quantitative comparison revealed that the extraction procedure had a greater impact on the chemical profile than genetic variability. All HIT extracts showed a higher total phenolic content compared to HII, while the antioxidant potential evaluated by 1,1-diphenyl-2-picrylhydrazil test was not proportionally higher. In addition, hot water extracts proved to be comparably active as alcoholic ones, confirming high commercial potential of Helichrysum italicum as herbal functional beverages.

Polysaccharides ofPhellinus igniariushave remarkable bioactivities and attracted considerable attention. In this study, extraction process of endo-polysaccharides fromP. igniariuwas investigated. The extraction conditions including ratio of sporocarp to water, extraction time, extraction temperature, extraction times were optimized. The experimental results indicated that the maximal yield of endo-polysaccharides fromP. igniariuwas 2.93 %, and the optimal conditions were as follows: ratio of sporocarp to water 1:30, extraction time 2 h, extraction temperature 90 °C, extraction twice. The best final ethanol concentration was 80 %. In addition, protein removal rate could reach 90.6 % using trichloroacetic acid method in extracting polysaccharides process.

In this paper, the influence of enzymatic pretreatment on the microwave assisted extraction (MAE) of polyphenols from artichoke leaves is described. Prior to enzymatic pretreatment, the influence of different parameters (extraction time, stirring rate, and extraction temperature) on the extraction process was studied. The total phenolic content (TPC) increases with the stirring rate. To avoid degradation of polyphenols, the extraction time and temperature should not be too high. The antioxidant capacity is in concordance with the TPC results. The enzymatic pretreatment, for the best extraction conditions, enhances the concentration of polyphenols compared with the extracts obtained without pretreatment.
