An experiment was conducted to evaluate the expression of insulin-like growth factor-I (IGF-I) and IGF-binding proteins (IGFBPs -II and -III) in response to a 10% inclusion of caramel plant wastewater (CPWW) in weaning pig diets; the objective was to assess associations between those growthrelated proteins to feed intake (Fl) and body weight gain (BWG). Sixteen purebred Landrace piglets were randomly distributed among eight pens (a gilt and boar per pen) and assigned to one of two treatments: 0% (control) and 10% inclusion of CPWW. During four consecutive weeks, live weight and Fl were recorded. Blood samples were drawn by jugular venipuncture during the first, second, and third weeks of the experiment and serum levels of IGF-I, IGFBP-II and IGFBP-III were determined. Feed intake, BWG and feed efficiency (FE) were not affected (P > 0.05) by the addition of 10% CPWW tothe diet, nor was animal health status visibly affected. Serum IGF-I levels were higher in control animals (P < 0.05) and increased from d 14 to d 28 of the experimental period (P < 0.05). Weekly increases were observed for IGFBP- III (P < 0.05) whereas IGFBP-II circulating levels decreased from d 14 to d 28 of the post-weaning test period. Simple correlation analysis revealed that there was a positive association between circulating levels of IGF-I and IGFBP-III (r = 0.88; P < 0.0001). However, the opposite was observed between these two and IGFBP-II (r = -0.84, P < 0.0001; r = -0.67, P < 0.0025, respectively). The changes observed in circulating levels of IGF-I, IGFBP-III and IGFBP-II were associated with weekly increases in Fl and BWG that occurred during the entire experimental period (P < 0.05).

In total, 32 pregnant goats were assigned randomly to four diets fed from Day 100 of pregnancy to Day 30 after parturition, to determine the effects of metabolisable energy (ME) and metabolisable protein (MP) supplementation levels on feed intake, subsequent colostrum and milk production and expression of genes regulating mammary-cell proliferation and apoptosis. Diets were as follows: (1) diet with ME and MP provided according to NRC recommendations (control), (2) diet with extra 10% ME, (3) diet with extra 10% MP, and (4) diet 1 with 10% extra of both ME and MP. Mammary biopsies were obtained from each udder half 24 h after parturition. Feed intake (g/day), and colostrum (kg/day) and milk (kg/month) production increased when the extra ME and MP were provided together prepartum and in early lactation (P &lt; 0.05). Relative mRNA expressions significantly increased in the mammary gland of insulin-like growth factor 1 (IGF-1, 4.3-fold), IGF-1 receptor (IGF-1R, 3.6-fold) and B-cell lymphoma 2 (Bcl-2, 4.6-fold), whereas insulin-like growth factor binding protein 3 (IGFBP-3, 3.2-fold), Bcl-2-associated X protein (Bax, 16.7-fold) and the ratio of Bax:Bcl-2 expressions significantly decreased (69.8-fold) with increased ME and MP levels fed in late gestation. In conclusion, colostrum production and milk yield in the early lactation period are sensitive to nutrient supply during gestation, where increased dietary ME as well as MP supplementation levels during late gestation will favour mammary development, by increasing expression of genes stimulating cellular proliferation (IGF-1, IGF-1R, Bcl-2) and reduced those stimulating apoptosis (IGFBP-3, Bax).

Abstract. Insulin-like growth factor I, other hormones and blood metabolites were measured in growing heifers before, during and after a 3-day period of normal feed intake and a corresponding period of reduced feed intake. In addition, 0.1 or 0.5 mg recombinant bovine GH/kg was injected daily for 5 days during normal or during and following reduced feed intake. During reduced feed intake blood concentrations of insulin-like growth factor I, insulin, T4, T3, glucose and urea-nitrogen decreased, whereas those of non-esterified fatty acids, albumin and protein increased (P< 0.05). GH, insulin-like growth factor I and insulin increased, whereas urea-nitrogen decreased in response to exogenous GH when heifers were adequately fed (P< 0.05). In contrast, insulin-like growth factor I did not change during GH injections while heifers received reduced amounts of feed. Therefore, during insufficient energy and (or) protein intake, characterized by low glucose, insulin and thyroid hormone levels and increased non-esterified fatty acid concentrations, insulin-like growth factor I concentrations and responses to GH administration were markedly reduced.

We infused recombinant human insulin-like growth factor I (IGF-I) for 4 h at 12.3 micrograms.h-1.kg live weight-1 directly into the left femoral artery and measured the rates of synthesis, degradation, and gain of protein by the treated and contralateral limbs of well-fed (n = 8), feed-restricted (n = 10), and fasted (n = 9) castrated male lambs. Reducing feed intake decreased net protein gain of hindlimb muscle, reduced hindlimb glucose uptake, and lowered arterial plasma concentrations of IGF-I, insulin, glucose, phenylalanine, tyrosine, and isoleucine. The effect of nutrition on IGF-binding proteins (IGFBP) was generally small; IGFBP-2 was more abundant in fasted lambs. Infusion of IGF-I into the left femoral artery increased plasma levels of IGF-I 2- to 4-fold in the left femoral vein and by 1.5- to 3-fold in the artery and right femoral vein. In the treated limb, IGF-I reduced protein degradation, increased protein gain, and increased glucose uptake without altering blood flow or oxygen uptake, regardless of feed intake. Systemically, IGF-I reduced plasma insulin, phenylalanine, tyrosine, isoleucine, and leucine in all nutrition groups. Plasma IGFBP-3 was increased by 4 h of IGF-I treatment in fasted but not in fed lambs. In fed but not fasted lambs, IGF-I increased blood glucose concentration. Effects of IGF-I on protein metabolism in the contralateral limb were affected by nutrition, generally more so in fasted than in unrestricted fed lambs.
