Objectives: Liver diseases are among the health challenges facing many people and health care providers worldwide. In their search for solution to these problems, researchers are increasingly advocating the use of herbal preparations with proven efficacy in protecting against hepatic disorders. They also investigate medicinal plants with the aim of developing new drugs.  Russelia equisetiformis is a plant which contains phytoconstituents that were reported to have biological activities, such as anti-inflammatory, antidiabetic, and membrane-stabilizing properties. In this study, the effect of methanol extract of R. equisetiformis (MEREQ) on paracetamol-induced hepatotoxicity was investigated in rats. Method: Rats were pretreated orally with graded doses (100 – 400 mg/kg b.w) of MEREQ for 7 days. On the 8th day, hepatotoxicity was induced in the pretreated rats with a single intraperitoneal injection of paracetamol (2 g/kg b.w). Rats were sacrificed on the 15th day; blood samples were taken for biochemical analysis, and the liver was excised for histopathological study. Biochemical parameters analyzed are alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), total protein, albumin, and bilirubin. Results: Administration of paracetamol in the rats resulted in significant increase (p<0.05) in the serum levels of AST, ALT, ALP, and bilirubin compared with the control. Treatment with MEREQ significantly reduced (p<0.05) the levels of these parameters in a dose-dependent manner, compared with the untreated rats. No significant changes were observed in the serum levels total protein and albumin. Histopathological examination showed that administration of paracetamol caused distortions in the architecture of the liver, but the degree of degeneration of hepatocytes was reduced in the MEREQ-treated rats. Objective: Many plants are consumed as food by humans for growth and survival, but a large number of these plants have not been tested for toxicity potential. Repeated consumption of such plants could lead to accumulation of toxic chemicals in the body and cause health-related problems. Irvingia wombolu kernel is widely consumed by many ethnic groups in Nigeria and some other African countries. The toxicity potentials of Irvingia wombolu kernel extract (IWKE) on the kidney and liver of rats was evaluated in this study. Methods: Three groups of Wistar rats were fed orally with IWKE (50, 100, and 200 mg/kg b.w) daily for 28 days. The fourth group which is the control was treated with distilled water (10 ml/kg b.w) for the same period.  The rats were sacrificed on the 29th day, and blood samples, kidney and liver were harvested for analyses. Serum levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), total protein, albumin, bilirubin, creatinine, and urea were determined. Kidney and liver sections were examined for histopathological changes. Data were subjected to Student’s t-test for statistical analysis. Results: Serum levels of creatinine and urea were not significantly altered in the IWKE-treated rats compared with the control. At 200 mg/kg b.w, the extract significantly increased (p<0.05) serum levels of ALT, AST, total bilirubin, total protein, and albumin compared with the control. Significant distortions were observed in the liver sections of rats treated with 200 mg/kg b.w IWKE compared with control, but the structure of the kidney section of IWKE-treated rats was not significantly different from the control. Objective: Phaseolus vulgaris, like other beans, is endowed with rich nutritional contents. This study evaluated the effects of raw and cooked aqueous and methanol extracts of P. vulgaris on renal function in albino Wistar rats. Methods: Oral acute toxicity (LD50) study of both extracts was conducted in two phases. In the main design, a total 36 Wistar albino rats were used and divided into nine groups of four rats and oral administration lasted for 7 days. Group 1 served as control and 2 – 9 treated groups. Groups 2 and 3; 4 and 5 were administered aqueous extracts while groups 6 and 7; 8 and 9 were administered methanol extracts of 350mg/kg and 550mg/kg body weight raw and cooked P. vulgaris respectively. Results: Results of LD50 of all extracts were greater than 5000mg/kg. Results showed a significant (P<0.05) increase in concentrations of urea and chloride across test groups administered aqueous extracts, than methanol extracts; a significant (P<0.05) increase in serum creatinine in test groups administered methanol extracts; a significant (P<0.05) increase of serum total protein of test groups compared to control; no significant (P<0.05) difference in the concentration of potassium in test groups administered compared to control group. Conclusion: It may be concluded that P. vulgaris portrays potentials capable of improving renal function and its consumption may contribute to the wellness of a person due to its rich nutrients, and based on the duration of this work and standard scale of toxicity; the extracts are practically non- toxic since the LD50 was greater than 5000mg/kg.   Peer Review History: Received 7 June 2020; Revised 25 June; Accepted 3 July, Available online 15 July 2020   UJPR follows the most transparent and toughest ‘Advanced OPEN peer review’ system. Pogostemon cablin(PC) is a herbal medicine traditionally applied to treat not only common cold, nausea and diarrhea but also headache and fever. The aim of this study was to investigate the analgesic and anti-inflammatory properties of standardized PC methanol extract (PCMeOH)in vivo. Investigations were performed in mice with two analgesic models. One was acetic acid-induced writhing response and the other formalin-induced paw licking. The anti-inflammatory effect was tested by λ-carrageenan (Carr)-induced mice paw edema. These analgesic experimental results indicated that PCMeOH (1.0 g/kg) decreased the acetic acid-induced writhing responses and PCMeOH (0.5 and 1.0 g/kg) decreased the licking time in the second phase of the formalin test. Moreover, Carr-induced paw edema inflammation was significantly reduced in a dose-dependent manner when PCMeOH (0.5 and 1.0 g/kg) was administered 3 and 4 h after the Carr injection. Mechanistic studies showed that PCMeOH decreased the levels of malondialdehyde in the edema paw by increasing the activities of anti-oxidant enzymes, such as superoxide dismutase, glutathione peroxidase and glutathione reductase, in the liver and decreasing the cyclooxygenase 2 and tumor necrosis factor-α activities in the edema paw. This study has demonstrated the analgesic and anti-inflammatory effects of PCMeOH, thus verifying its popular use in traditional medicine.

The liver and the kidneys are two very important organs in the body and they are responsible for the metabolism and excretion of drugs respectively amongst several other functions. Severe adverse effects on these organs can lead to organ dysfunction or failure and a consequential effect on wellbeing and can even be life-threatening. This study investigated the effects of hydromethanolic stem extract of Costus afer Ker Gawl. (Costaceae) on liver and kidney function indices and the histopathology of the organs in Wistar rats. Serum liver enzymes which include: alanine amino transferase (ALT), aspartate amino transferase (AST) and alkaline phosphatase (ALP), total protein and albumin as well as serum urea, creatinine, sodium ions, potassium ions, chloride ions, bicarbonate ions were evaluated in biochemical studies. Sections of the liver and kidneys appropriately treated were examined microscopically for pathological lesions.There were decreased serum levels of ALT and ALP, but serum levels of AST increased at 500 and 1000 mg/kg doses. Serum levels of total protein (TP) and albumin concentration as well as urea and creatinine serum levels were not significantly (p>0.05) affected. However, histological examination of the liver and kidneys revealed slight to moderate hepatic necrosis and slight tubular necrosis respectively especially at 500 and 100 mg/kg doses of the extract. The results showed that the extract may be harmful to the liver and to a lesser extent the kidneys on prolonged administration and therefore it should be used with caution in such instances.

Background: Anti-inflammatory and immunomodulatory properties are prerequisites for the success of cell therapy as shown by adult Mesenchymal Stem cells (MSC) by reducing both B- and T-lymphocyte proliferation, in a paracrine dependent manner. Proteomics of neonatal MSCs (nMSCs) has identified the superior quality of secretome containing a significantly higher number of anti-inflammatory (68 vs 21 in aMSCs) molecules. Hypothesis: nMSCs have potent anti-inflammatory and immunomodulatory characteristics as compared to other cell types currently being used as cell therapy. Methods: nMSC were tested and compared to other cell types for: a) Hypo-immunogenicity (expression of MHCII in the presence of IFN-gamma), b) immunogenicity (reduce proliferation of human CD4+ T-cells in a co-culture mixed lymphocyte reaction (MLR), c) anti-inflammatory properties (reduce IL-8 secretion of macrophages in the presence of Nigericin, d) systematic immune response (using rat myocardial infarction (MI) model (IL10, TNF-a and IL12)) and e) Immunomodulation (CD4+ cells to T reg CD4 + /CD25 + /FoxP3 + ). Results: Expression of MHCII on nMSCs, after 72 hours of exposure to IFN-gamma and proliferation of CD4 + cells in MLR was significantly (40±3.8% and 55±4.3%, n=5) reduced as compared to aMSC or CDC. IL8 was measured in the supernatants of nMSC derived secretome reduced the secretion of IL8 from activated macrophages in a dose dependent manner (150 ug protein/ml). Intravenous injection of nMSCs in a rat MI model significantly reduced serum levels of of TNF-α and IL-12 while upregulated IL-10 and improving myocardial function. While BM-MSCs and aMSCs failed to significantly modulate immune system, nMSC significantly increased the number of Treg cells as compared to CDC. Similar results were observed with Pig IRI model. Conclusions: Together, in a head-to-head comparison, nMSCs outperformed all other competitive cell types currently under investigation for their tissue repairing role, including CDCs and BM-MSCs to achieve the highest clinical efficacy.

Most of the synthetic drugs used at present as analgesic and anti-inflammatory agents cause many side effects and toxic effects. Many medicines of plant origin with analgesic and anti-inflammatory activity have been used since long time without adverse effects. The purpose of the study was to explore the anti-pyretic and anti-oxidant activity of fruits of Garcinia pedunculata. The fruit of G. pedunculata was collected from Shillong, Meghalaya and extraction is done by using methanol as a solvent. The methanol extract shows the presence of carbohydrate, glycoside, alkaloids, phenol and flavonoids. Acute anti-pyretic activity for the extract was investigated in pyrexia rat. Temperature level was determined after 1hours, 4 hours, 8 hours and 12 hours after giving the extract dose of 100mg/kg and 200mg/kg body weight, and it was found to produce a significant reduce in fever. DPPH radical scavenging activity (In-vitro anti-oxidant activity of G. pedunculata) of methanolic extract shows 38.31 as IC50 (µg/ml) whereas Ascorbic acid as standard in the same concentration shows 34.81 IC50 (µg/ml). The obtained result justified the traditional use of G. pedunculata as anti-pyretic and antioxidant purpose.

Many research groups are working to find new possible anti-inflammatory molecules, and marine sponges represent a rich source of biologically active compounds with pharmacological applications. In the present study, we tested different concentrations of the methanol extract from the marine sponge,Geodia cydonium, on normal human breast epithelial cells (MCF-10A) and human breast cancer cells (MCF-7). Our results show that this extract has no cytotoxic effects on both cell lines whereas it induces a decrease in levels of VEGF and five proinflammatory cytokines (CCL2, CXCL8, CXCL10, IFN-γ, and TNF-α) only in MCF-7 cells in a dose-dependent manner, thereby indicating an anti-inflammatory effect. Moreover, interactomic analysis suggests that all six cytokines are involved in a network and are connected with some HUB nodes such as NF-kB subunits and ESR1 (estrogen receptor 1). We also report a decrease in the expression of two NFKB1 and c-Rel subunits by RT-qPCR experiments only in MCF-7 cells after extract treatment, confirming NF-kB inactivation. These data highlight the potential ofG. cydoniumfor future drug discovery against major diseases, such as breast cancer.
