Foot-and-mouth disease (FMD) is the most highly contagious disease affecting livestock resulting in a significant adverse economic impact worldwide. Disease outbreaks in previously FMD-free countries are initially controlled by the culling of infected and in-contact animals, restriction of susceptible animal movement, and vaccination with an inactivated whole-virus antigen preparation. With increasing trade, commerce globalization, and people migration, it is likely that more inter-pool viral exchanges and spillovers will occur posing a greater threat to both, endemic and non-endemic regions, mostly due to the limited antigenic coverage of current vaccines. Currently available inactivated FMD vaccines have a number of disadvantages, including incomplete inactivation of the virus, they require multiple vaccination to maintain good levels of immunity and periodic inclusion of new viral strains into the vaccine formulation to cover new FMDV subtypes against which existing vaccines no longer protect and lack differentiation of infected from vaccinated animals (DIVA). The essential aim of DIVA strategy is realization of the so-called “vaccinate-to-live” policy, which is based on the principles that vaccinated animals exposed to FMDV will not transmit the virus. To address the shortcomings of inactivated vaccines, many efforts are currently devoted to developing novel FMD vaccines, including attenuated and marker inactivated vaccines, recombinant protein vaccines, synthetic peptide vaccines and empty capsid vaccines. Novel vaccine platforms offer promising alternatives for effective FMD control. It is likely that in the near future, multiple FMD vaccine approaches will compete for diverse markets, providing fit-for purpose solutions to evolving challenges in preventing and controlling FMD worldwide.

Background and Aim: Signs of dysautonomia were frequently observed in calves that died during foot-and-mouth disease (FMD) virus (FMDV) outbreaks in Egypt from 2015 to 2018. This study aimed to describe the clinical and histopathological features of the central nervous system in malignant cases of FMD and excluding possible concurrent bacterial, and bovine herpes virus 4 (BHV4) infections or both. Materials and Methods: In this study, 335 FMDV-infected buffalo calves aged 1-22 months were clinically examined and followed until recovery or death. Of the 335 calves, 134 died (malignant group) and 201 recovered after exhibiting classic symptoms of FMD (recover group). The calves were subjected to clinical examination. For the malignant group, several laboratory trials were conducted to assess the possible cause/s of dysautonomia-related viral, bacterial, or concurrent infections. Koch's postulates and polymerase chain reaction were employed. Postmortem and histopathological examinations of nervous tissue were performed. Results: In the malignant group, signs of dysautonomia were observed before death, including partial or complete gut dysfunction, loss of anal sphincter tone, rapid breathing sounds, fluctuating body temperature, and cardiac arrhythmias. In the malignant group, histopathological examination of the spinal cord, pons, medulla oblongata, hypothalamus, cerebellum, and cerebrum revealed demyelination, neuronal degeneration, and focal areas of malacia and gliosis. The nervous tissue and heart samples from malignant cases were positive for serotype O FMDV. Conclusion: Findings revealed in this study support the existence of neurodegeneration induced by FMDV infection in buffalo calves.

Three Foot and Mouth Disease (FMD) outbreaks in northern Thailand that occurred during the implementation of the national FMD strategic plan in 2008–2015 are described to illustrate the lessons learned and to improve the prevention and control of future outbreaks. In 2008, during a FMD outbreak on a dairy farm, milk delivery was banned for 30 days. This was a part of movement management, a key strategy for FMD control in dairy farms in the area. In 2009, more than half the animals on a pig farm were affected by FMD. Animal quarantine and restricted animal movement played a key role in preventing the spread of FMD. In 2010, FMD infection was reported in a captive elephant. The suspected source of virus was a FMD-infected cow on the same premises. The infected elephant was moved to an elephant hospital that was located in a different province before the diagnosis was confirmed. FMD education was given to elephant veterinarians to promote FMD prevention and control strategies in this unique species. These three cases illustrate how differences in outbreak circumstances and species require the implementation of a variety of different FMD control and prevention measures. Control measures and responses should be customized in different outbreak situations.
