BACKGROUND: High-fat diet (HFD) is associated with dyslipidemia which is a risk factor for atherosclerosis. Dyslipidemia causes oxidative stress which induces vascular cell adhesion molecule-1 (VCAM-1). Oxidative stress also triggers the thickening of tunica intima-media (IMT) and Perivascular Adipose Tissue (PVAT). Xanthone compound in ethanolic extract of Mangosteen pericarp (EEMP) has an antioxidant property to overcome the oxidative stress. AIM: The objective of this study is to investigate the effect of dietary EEMP administration on the expression of VCAM-1 and thickness of PVAT and IMT in atherosclerotic rat model fed with HFD. METHODS: This experimental laboratory study uses 25 Wistar strain Rattus norvegicus which were divided into 5 study groups. Negative Control group (GT1) was given a normal diet, Positive Control group (GT2) was treated with HFD, and three treatment groups were each treated with HFD with Mangosteen pericarp extract of 200 mg/kg BW (GT3), 400 mg/kg BW (GT4), and 800 mg/kg BW (GT5). Measurements of VCAM-1 expression were performed using immunofluorescence. PVAT and IMT measurements were performed on rat aortic preparations. RESULTS: One-way ANOVA test showed the addition of dietary EEMP significantly (p < 0.05) decreased the expression of VCAM-1 and decreased the thickness of PVAT and IMT in treatment groups as compared with both negative and positive controls. Tukey HSD test showed a dose of 800 mg/kg BW was the most effective dose for decreasing VCAM-1 level, PVAT and IMT. CONCLUSION: Dietary EEMP significantly decreases the expression of VCAM-1, as well as the thickness of PVAT and IMT in Wistar strain Rattus norvegicus treated with HFD.

Introduction: Oral ulcer lesions are one of the most common lesions in the oral cavity. The prevalence of traumatic ulcers is quite high compared to other oral lesions. Brown algae, Sargassum sp. is one of the algae contains flavonoids, vitamin A, vitamin C, iron, and calcium, which play a role in wound healing and has economic value. This study was aimed to determine the effectiveness of Sargassum sp. ethanolic extract on traumatic ulcers healing in the labial mucosa of Wistar strain (Rattus norvegicus). Methods: 28 Wistar strain (Rattus norvegicus) weighed 150-200 grams. The samples were divided into 4 groups (K1, K2, P1, P2); each group consisted of 7 mice. K1 group was administered with Hydroxy Propyl Methyl Cellulose (HPMC), K2 was administered with 0.2% hyaluronic acid gel, P1 was administered with 50% Sargassum sp. extract gel, and P2 was administered with 75% Sargassum sp. extract gel. In each group, the ulcer was made on the mice labial mucosa; then the ulcer diameter was measured and observed on day 1 and day 7. Results: The mean diameter ± standard deviation of traumatic ulcer calculations were K1 (1.09 ± 0.40), K2 (1.81 ± 0.67), P1 (1.39 ± 0.43) and P2 (2.16 ± 0.49) respectively. One way ANOVA test results showed significant differences in K1, K2, P1, and P2 groups. Conclusion: Ethanol extract of Sargassum sp. is effective in accelerating the traumatic ulcers healing at the concentration of 75%.Keywords: Traumatic ulcer, Sargassum sp., wound healing.

<p>Teratogenic effect of herbicide glyphosate-Roundup, sold under the name Glifotox on Wistar rats was evaluated. The biological material was treated intraperitoneally with glyphosate at concentrations of 100, 125, and 150 mg/kg from gestation day nine. Hysterectomy was performed on day 18 of gestation, and the uterine horns where the embryos were located, in addition to recording the percentage of malformed embryos by modifying the method of Wilson were observed. The liver was removed and quantified by spectrophotometry with transaminase activity showed higher concentrations malformation rate and higher enzyme activity was 125 mg/kg, below is the average of 100 mg/kg and higher concentrations such as 150 mg/kg a large number of resorptions was obtained. It is concluded that glyphosate is toxic affecting the liver and liver enzymes involved in the formation of amino acids also produce delay in embryonic development.</p>

High salt diet and prednisone administration will increase blood pressure which chronically causes heart muscle remodeling so that it appears macroscopically as left ventricular hypertrophy. Bay leaf extract contains flavonoids which can lower blood pressure and decrease left ventricular thickening. To determine the effect of bay leaf extract (Syzygiumpolyanthum) on the left ventricle thickening of rat heart (Rattus norvegicus wistar strain) in hypertensive model.  True experimental post-test only controls group design with simple random sampling technique. The subjects of this study were 25 male Wistar rats aged 2-3 months weighing 150-200 grams. Rats were induced with prednisone 1.5 mg/ KgBW / day in 2% NaCl for 6 weeks followed by administration of bay leaf extract at a dose of 70 mg/ KgBW, a dose of 140 mg/ KgBW, and a dose of 280 mg/ KgBW. Data analyzed by One Way ANOVA, Post Hoc Bonferroni, followed by Linear: One Way ANOVA test showed significant differences between treatments (p = 0.000). The Bonferroni Post Hoc test concluded that there were significant differences in the treatment of the P2 and P3 groups. Linear regression test obtained R square 0.663 and obtained the equation Y = 1965.838 - 1.275X. The extract of bay leaf (Syzygiumpolyanthum) has an effect on decreasing the left ventricle thickening of the white male rat heart (Rattus norvegicus wistar strain) hypertension model.

The purpose of this study was to prove that the administration of Jati (Guazuma ulmifolia Lamk) leaves extract improves the lipid profile of dyslipidemic male Wistar rats. Subjects were 20 rats (Rattus norvegicus), male, Wistar strain, dyslipidemia (total cholesterol ≥ 200 mg dl-1), aged 2 months old, weighing 180-200 grams. The control group (10 rats) were given a placebo of 3 ml aquadest (P0) and the treatment group was given extracts of the Jati (Guazuma ulmifolia Lamk) leaves extract of 25 mg kg-1 BW (P1). Before and after treatment for 14 days, total cholesterol, triglyceride, LDL, and HDL levels were examined. The results showed that in the P0 group there were no changes in total cholesterol, triglyceride, LDL, and HDL levels (p>0.05), whereas the P1 group experienced a decrease in total cholesterol, triglyceride and LDL levels (p<0.05) and an increase in HDL levels (p<0.05). The results of this study indicated that the Jati leaves extract was effective to improve the lipid profile of dyslipidemic rats. It was necessary to compare the effectiveness of Jati leaves extract with synthetic dyslipidemia drugs used in the community such as statin.

Tujuan: untuk mengetahui dosis efektif enoxaparin dalam mencegah terjadinya trombosis pada anastomosis mikrovaskular. Metode: penelitian ini bersifat eksperimental dengan rancangan the randomized post test only control group design. Terdapat 33 tikus jantan Rattus norvegicus Wistar strain yang dikelompokkan menjadi tiga perlakuan yaitu perlakuan A (enoxaparin 0,75 mg/kg), B (enoxaparin 1  mg/kg), dan C (kontrol). Tuck model anastomosis dilakukan pada arteri femoralis, kemudian luas trombus yang terjadi pada pembuluh darah dibandingkan dengan diameter lumen pembuluh darah diukur dengan graticule lens dan dinyatakan dalam persen. Hasil: trombus terbentuk pada semua subyek penelitian baik pada kelompok perlakuan maupun kontrol. Rerata persentase luas trombus pada kelompok enoxaparin 0,75 mg/kg adalah 24,3%, enoxaparin 1 mg/kg sebesar 19,8% dan kelompok NaCl 0,9% sebesar 79,4%. Terdapat perbedaan antara perlakuan pemberian enoxaparin dosis 0,75 mg/kg dan 1 mg/kg dengan kontrol, namun tidak ada perbedaan bermakna rerata persentase luas trombus diantara kelompok enoxaparin dosis 0,75 mg/kg dan dosis 1 mg/kg (p=0,624). Perlu dilakukan penelitian secara klinis guna melihat efektivitas enoxaparin dalam meningkatkan patensi anastomosis pada free flap maupun replantasi. Simpulan: pemberian enoxaparin dosis 0,75 mg/kg dan enoxaparin dosis 1 mg/kg secara subkutan efektif mengurangi persentase luas trombus pada anastomosis arteri femoralis tikus. Tidak terdapat perbedaan efektivitas yang bermakna dalam mengurangi persentase luas trombus pada anastomosis arteri femoralis tikus setelah diberikan dosis enoxaparin 0,75 mg/kg dan 1 mg/kg secara subkutan.

Mangosteen, or Garcinia mangostana L., has merged as an emerging fruit to be investigated due to its active compounds, especially xanthone derivatives such as α -mangostin (AM), γ-mangostin (GM), and gartanin (GT). These compounds had been reported to exert some pharmacological activities, such as antioxidant and anti-inflammatory, therefore, the development of an analytical method capable of quantifying these compounds should be investigated. The aim of this study was to determine the correlation between FTIR spectra and HPLC chromatogram, combined with chemometrics for quantitative analysis of ethanolic extract of mangosteen. The ethanolic extract of mangosteen pericarp was prepared using the maceration technique, and the obtained extract was subjected to measurement using instruments of FTIR spectrophotometer at wavenumbers of 4000–650 cm−1 and HPLC, using a PDA detector at 281 nm. The data acquired were subjected to chemometrics analysis of partial least square (PLS) and principal component regression (PCR). The result showed that the wavenumber regions of 3700–2700 cm−1 offered a reliable method for quantitative analysis of GM with coefficient of determination (R2) 0.9573 in calibration and 0.8134 in validation models, along with RMSEC value of 0.0487% and RMSEP value 0.120%. FTIR spectra using the second derivatives at wavenumber 3700–663 cm−1 with coefficient of determination (R2) >0.99 in calibration and validation models, along with the lowest RMSEC value and RMSEP value, were used for quantitative analysis of GT and AM, respectively. It can be concluded that FTIR spectra combined with multivariate are accurate and precise for the analysis of xanthones.

Cardiovascular disease is a disease caused by impaired functions of heart and blood vessel, one of those is coronary heart disease. This study aimed to identify the effect of purple sweet potato (Ipomoea batatas L) extract on MDA (Malondialdehyde) levels of male white rat (Rattus norvegicus Wistar strain) model of atherosclerosis. This research was an experimental study using Post-Test Only Control Group Design method, used 5 groups (positive control group, negative control, treatment 1, treatment 2, and treatment 3), each group contained 5 rats plus 2 reserve rats. ANOVA one way test obtained a significance of 0.00 (p <0.05). The regression test results showed the coefficient of determination R² = 0.71, which means a decrease in serum MDA levels of experimental animals is influenced by the dose of purple sweet potato extract administration as much as 71% while the remaining 29% can be influenced by factors outside the study. It can be concluded that purple sweet potato extract can affect serum MDA levels of experimental animals.
