<p>Fish skin is one of marine byproducts potential for alternative source of collagen. This study<br />investigated the effectiveness of alkaline and acetic acid pretreatment on the characteristics of<br />collagen from skin snakehead fish. The concentrations of alkaline pretreatment were 0.05; 0.1; 0.15<br />and 0.2 M for 2, 4, 6, 8, 10 and 12 hours, acetic acid concentrations were 0.05 M, 0.1 M, 0.15 M and<br />0.2 M for 1 and 2 hours. The experimental design used for alkaline and acetic acid pretreatment<br />was factorial completely randomized design. The result showed that the concentration of alkaline<br />0.05 M for 6 hours have significant effect on the elimination of non-collagen protein (p&lt;0.05),<br />whereas for the optimum acetic acid at a concentration 0.1 M for 2 hours significantly different<br />on solubility and swelling. Extraction yields of collagen was 16%, with characteristics of whiteness<br />66.67%, protein content 96.21%, viscosity 10 cP, Tmax 159.9oC and glass transition temperature<br />78.55oC. The dominant amino acid composition were glycine (27.11%), proline (13.87%) and<br />alanine (12.58%). Functional groups collagen from skin snakehead fish has β-sheet structure<br />which is a characteristic of collagen.</p>

Swim bladders of catfish contain high protein, therefore it is can be used as a raw material for collagen. The study aims to determine the chemical characteristics of swim bladders, the pretreatment of non-collagen, extraction of collagen dissolves acid and to evaluate the characteristics of collagen. The method of this study is KOH pretreatment with a concentration of 0,05; 0,1; and 0,15 for 12 hours. The extraction process is done by soaking the sample in a solution of acetic acid with a concentration of 0,25; 0,5 and 0,75 M (ratio 1:10; b/v) and extraction time for 24; 48; and 72 hours at 4oC. The experimental design used for alkaline and acetic acid pretreatment were factorial completely randomized design. The result showed that the protein content of swim bladder was 85,26% (db), the profile of amino acids were dominated by three amino acids namely glycine (56,85 mg/g), prolyne (31,03 mg/g), and alanyne (23,85 mg/g). Using 0,05 M KOH for 8 h was selected as the best pretreatment method for collagen extraction. Extraction method using 0,50 M acetic for 48 h resulted he best collagen which revealed the existence of a triple helix structure and had Tg 84oC.

The horn snail (Telescopium telescopium) is a gastropod living in mangrove forests and is considered as a pond pest. This study was aimed to determine the effects of alkaline and acetic acid pretreatment on the characteristics of collagen from the horn snail meat. The immersion of the meat in NaOH 0.20% for 10 hours gave a significant reduction in non-collagen protein content (α = 0.05). Further immersion with acetic acid 0.05% for 6 hours also significantly affected (α = 0.05) the swelling degree and the collagen solubility. The extraction yield was 1.08±0.21%, with 72.23% of brightness, and the viscosity was 16.34 cP. The proline content of the collagen was 9.21%.

The purpose of this study was to determine the effect of the slaughtering age of goats on goat bone gelatin quality. The study was conducted using a completely randomized design (CRD). Data showing influence are further analyzed using the Duncan test. Treatment consisted of age-old 1-year goat bone (T1), 2-year-old goat bone (T2), and 3-year-old goat bone (T3). Each treatment was repeated 3 times. The results showed that the younger the goat age (1 year), the higher the value of yield, protein content, water content, yellowish color level (b), and viscosity. While the fat content, ash content, the level of brightness (L), and redness (a) in the colors show variable results. Each parameter showed a significant effect (P <0.05). Conclusion, 1-year-old goat leg bone gelatin with acetic acid pretreatment (CH3COOH) concentration of 5% has good quality compared to ages 2 and 3 years.

Pankreas kambing mengandung enzim proteolitik yang berpotensi sebagai bating agent dalam proses penyamakan kulit.Tujuan penelitian ini adalah mengetahui pengaruh penggunaan bating agent dari pankreas kambing terhadap karakteristik fisik kulit ikan nila tersamak dan mengetahui konsentrasi terbaik dari penggunaan bating agent pankreas kambing. Materi yang digunakan dalam penelitian ini adalah kulit ikan niladan pankreas kambing segar. Metode penelitian ini bersifat experimental laboratories menggunakan Rancangan Acak Lengkap (RAL) dengan tiga kali pengulangan dengan 4 konsentrasi bating agent  yaitu (0,5%, 1%, 1,5% dan 2%) kulit ikan tersamak diuji dengankekuatan tarik, kemuluran, kekuatan sobek, kadar krom oksida, dan suhu kerut. Hasil penelitian menghasilkan bahan bating agent berpengaruh terhadap karakteristik kulit ikan tersamak (P<0,05). Konsentrasi bating agent sebesar 2% memberikan hasil yang terbaik sebagai berikut: kekuatan tarik 1300,45 N/cm², kemuluran 87,70%, kekuatan sobek 273,90 N/cm², kadar krom oksida 3,46 %, dan suhu kerut 99,75°C. Kulit ikan nila tersamak hasil penelitian semua perlakuan  memenuhi persyaratan SNI 06-4586-1998 kecuali untuk parameter kemuluran tidak memenuhi syarat. Goat pancreas, containing proteolytic enzymes which the potential as a bating agent in the process of tanning the skin. The purpose of this study was to determine the effect of using bating agent from goat pancreas on the physical characteristics of tilapia skin and knowing the best concentration of using the goat pancreatic agent. The material used in this study is the skin of tilapia and fresh goat pancreas. This research method is experimental laboratories using Completely Randomized Design (CRD) with three repetitions with 4 concentrations of bating agent namely (0.5%, 1%, 1.5% and 2%) the skin of tanned fish is tested with tensile strength, elongation, tear strength, chrome oxide content, and wrinkle temperature. The results of the study resulted in bating agent material affecting the characteristics of tanned fish skin (P <0.05). Bating agent concentration of 2% gave the best results as follows: tensile strength 1300.45 N / cm², elongation 87.70%, tear strength 273.90 N / cm², chrome oxide content 3.46%, and wrinkle temperature 99, 75 ° C. The results of this study produced a total nitrogen of 2.65% of the goat pancreas which has great potential to be used as bating material. The skin of tanned tilapia from the results of this study meets the requirements of SNI 06-4586-1998 except for elongation parameters that do not meet the requirements.

Fish skin is one of the alternative sources contained high protein  to isolate collagen. Fish skin generally extracted by the method of acid, alkali and enzymes. The study aim to determine the effectiveness of NaOH<br />and acetic acid on catfish (Pangasius sp.) skin extraction  process.  The concentrations of alkaline pretreatment were 0,05; 0,1; 0,15 and 0,2 M with the soaking time of 2, 4, 6, 8 and 10 h by NaOH replacement in every 2 h. The concentrations of acetic acid for hydrolisis process were 0.05; 0.1; 0.15 and 0.2 M with the soaking time of 1, 2, and 3 h. The experimental design used for pretreatment process is split splot, while for the hydrolysis process is factorial completely randomized design. The results showed that pretreatment with a concentration of 0.05 M NaOH for 4 h has a significant effect for eliminating non-collagen protein (p&lt;0.05). The acetic acid concentration of 0.15 M for 1 h also has a significant effect on fish skin swelling. The yield of striped catfish collagen was 17.272%, the protein content was 86%, and the viscosity was 12 cP. Fish skin extract was identified as type I collagen by functional groups and electrophoretic analysis. Collagen from striped catfish skin has α1 and α2 and protein structure with the molecular weight of α chain were 94 and 98 kDa, meanwhile the molecular wheight of β chain was 204 kD.

ABSTRACT The objective of this work was to evaluate the effect of six indole-3-acetic acid (IAA) concentrations and three immersion times in IAA solutions on the vegetative propagation of Spondias dulcis and Spondias tuberosa by stem cuttings. The experiment was conducted in a greenhouse, using a completely randomized design, with a 6×3 factorial arrangement consisting of six indole-3-acetic acid concentrations (0, 2, 4, 6, 8, and 10 g L-1) and three immersion times (8, 16, and 24 seconds), with ten cuttings of S. dulcis and S. tuberosa per plot, and three replications. Percentage of surviving cuttings, and percentage of rooted cuttings, number of leaf buds, number of roots per cutting, root length, and total fresh weight were evaluated at 180 days after planting. Data were subjected to analysis of variance by the F test and to regression analysis. Vegetative propagation from woody cuttings of S. dulcis is not viable to produce seedlings, thus, new studies on this technique for this species are needed. Vegetative propagation from woody cuttings of S. tuberosa is satisfactory, but with low efficiency for improvement and emergence of new shoots and roots; it is more effective when using a concentration of 10 g L-1 of indole-3-acetic acid and immersion time in the solution of 16 seconds.

This research is about application IBA and NAA with some cutting materials of “Madu Deli Hijau” water apple using NFT hydroponic system was aim to study the growth of water apple’s cuttings was treated by IBA and NAA 500 ppm and study the best kind of cutting materials of water apple that can growth the most.  This research was conducted using completely randomized design in two factors.  The first was consisted of 7 treatments; control (without Auxin), IBA 500 ppm, and NAA 500 ppm. The second was consisted of 4 treatments; woody branch (hardwood), semi woody (semi hardwood), and soft branch, softwood, and shoots. The results showed that the added auxin in the form of IBA and NAA 500 ppm can increase the success of madu deli hijau water apple cuttings and the cuttings from the shoots are the best for the success of madu deli hijau water apple in NFT hydroponic system.

The objective was to evaluate the effect of inclusion of acetic acid during the ensiling of sugarcane (Saccharum officinarum) on fermentation quality and nutritional value. The treatments consisted of sugarcane silages, variety (IAC 86-2480) additives with four inclusion levels of glacial acetic acid (1.5, 3.0, 4.5 and 6.0 % in natural matter) plus the control. A completely randomized design was used, being 5 treatments and 6 replicates. The glacial acetic acid showed a pH 2.9 (0,1M). There was a reduction of 0.07 units in the pH of the sugarcane silage for each 1% inclusion of acetic acid (P < 0.01). The N-NH3 concentration was not modified in the ensiled mass of the treatments, with a mean of 0.45% in dry matter (P =0.91). The averages of effluent losses were adjusted to the quadratic regression model with the application of acetic acid (P < 0.01). There was a reduction in the yeast population in the order of 0.44 log UFC/g of silage (P < 0.01). The addition of acetic acid in sugarcane silage reduces fermentative losses, the yeast population and improves the nutritional value in doses from 1.5% of natural matter.

The research of callus induction in potato (S. tuberosum  L.) from Dombu Village with indole-3-acetic-acid (IAA) was conducted from Januari to April 2019 in Laboratory of Tissue Culture, Biology Department, Faculty of Mathematics and Natural Science, Tadulako University. The research was aimed to determine of concentration IAA of the best to induce callus. This study was designed based on Completely Randomized Design (CRD) with 5 treatments, 3 replications and 3 explants on each unit. The treatments were P1= MS + 0 ppm, P2= MS + 0.5 ppm, P3= MS + 1.0 ppm, P4= MS + 2.0 ppm, P5= MS + 3.0 ppm. The results showed that  the best and the efficient treatments for induction of callus treatment was P4= MS + 2.0 ppm with to induce callus up to 100%, the callus colour was yellow to brown and intermediate texture.

Background: Hotong seeds are a growing and fertile plant on Buru Island and have been exploited by local people as an alternative source of carbohydrates. However, its utilization is still very simple as a result of not knowing the pattern of utilization in other form. Hotong seeds are the smallest cereal seeds weighing 15.2 kg each. Hotong seeds have the highest nutrient content, diameter hot seeds at the base of the larger than the tip and center. The color of seeds are varied hot yellow, brownish and red. Method: This study was an experimental study using Completely Randomized Design (RAL) with long storage treatment (0, 6, 8, and 12 months). Each treatment was repeated three times. The parameters observed were fat content in hotong seeds. Analysis of fat content using descriptive analysis method. Results: The results showed that the fat content of Hotong seeds of Waelo Village differed in control and on each treatment of storage duration. In the control of low fat content of 3.3591%, fat content increased in storage of 6 months and 8 months that is 3.5491% and 3.7242% and then decreased at 12 months ie 3.9322%, where from each treatment fat content the highest was obtained at 12 months storage treatment. Conclusion: There are different amounts of fat content in Hotong seeds of Waelo Village on the controls as well as on any storage treatment duration.
