<p>The pro-antioxidant balance in the liver and muscles of sterlet under the influence of artificial carbonic dioxyde hibernation and anaesthesia have been studied. In experiments were used two-years old sterlet weighing 170–210 g which were grown under conditions of a closed water supply. Four groups of five copies of each fish were formed: Control group I (fish remained intact); Group II (the clove oil was used for fish anaesthesia, which was added to the water); Group III (fish were hibernated by carbon dioxide); Group IV - control of hibernation (after complete recovery from carbon dioxide hibernation, these groups of fish were returned to the pool of incubation shop for her follow–up). The research of intensity of the formation products of peroxide oxidation of lipids (POL) in the body of sterlet in the experimental conditions were conducted by determining the content of thiobarbituric acid active products (TBA–active products) in the muscles and liver that are generated at the final stages of lipid peroxidation. The content of malonic dialdehyde (MDA), one of the intermediate products of lipid peroxidation of membrane was determined photometrically by the concentration of colored complex formed by its reaction in the acidic environment of the two molecules of thiobarbituric acid (TBA). Catalase activity was determined by the ability of hydrogen peroxide to form the stable colored complex with Molybdenum salts. The results support the adaptive character in fluctuations in the antioxidant protection system by the actions of carbon dioxide hibernation and anaesthesia on the sterlet body. This also contributes to usage of these conditions in fish transportation at long distances without stress factors.</p>

AbstractThe association between obesity and cardiovascular diseases has a multifactorial pathogenesis, including the synthesis of inflammatory molecules, the increase in oxidative stress and the dysregulation of the matrix metalloprotease (MMP) concentration and activity. In a group of adults with obesity, divided in 2 subgroups according to the body mass index (BMI), we examined lipid peroxidation, expressed as thiobarbituric acid-reactive substances (TBARS), protein oxidation, expressed as protein carbonyl groups (PCs), plasma gelatinases (MMP-2 and MMP-9), and their tissue inhibitors (TIMP-1 and TIMP-2). In the whole group, as well as in the 2 subgroups (with BMI 30–35 or BMI>35) of obese subjects, we observed an increase in TBARS, PCs, MMP-2, and MMP-9, and also TIMP-1 and TIMP-2 in comparison with the control group. A positive correlation between TBARS and PCs emerged in obese subjects and persisted after dividing obese subjects according to BMI. The correlation between MMP-2 and TIMP-2 was not statistically significant, while a significant correlation was present between MMP-9 and TIMP-1. The correlations between the markers of oxidative stress (TBARS and PCs) and those of the MMP/TIMP profile indicated a more marked influence of protein oxidation on MMPs and TIMPs in comparison with TBARS. The innovative aspect of our study was the simultaneous evaluation of oxidative stress markers and MMP/TIMP profile in adult obese subjects. We observed significant alterations and correlations that may negatively influence the clinical course of the disease.

Modern environmental conditions dramatically increased the level of radical forming processes in the body. Heat exposure stimulates the generation of reactive oxygen species inducing peroxidation of lipids as a result of the development of hypoxia. In experimental conditions the possibility to correct free radical lipid oxidation of rats’ organism membranes was studied with the oral introduction of the phytoadaptogens that contain the complex of natural antioxidants. The animals were divided into 5 groups and each of them had 30 rats: intact animals (1) which were held in standard conditions of vivarium; the control group (2) in which rats were exposed to heat during forty-five minutes daily; the experimental groups (3, 4, 5) in which before the effects of heat animals had a daily oral intake of the extracts of Eleutherococcus, Rhodiola, liquorice in a dose of 1 ml/kg. It was found out that in the liver tissue of experimental animals a daily heat exposure during forty-five minutes contributes to the increase of lipid hydroperoxides level (by 34-41%), of diene conjugate (by 45-50%), and of malonic dialdehyde (by 62-74%) against the decrease of antioxidant system activity in the liver of intact animals. The introduction of phytoadaptogens to rats in the conditions of heat exposure contributes to the reliable decrease in the liver of lipid hydroperoxides by 22-28%, of diene conjugates by 22-32%, and of malonic dialdehyde by 30-37% in comparison with the rats of the control group. While analyzing the effect of the phytoadaptogens on the activity of the components of antioxidant system it was shown that the level of ceruloplasmin in the liver of animals was reliably higher by 68-82%, of vitamin E by 42-65% in comparison with the same parameters of the rats of the control group. So, the application of phytoadaptogens in the conditions of long heat exposure of the organism of animals under experiment leads to the stabilization of the processes of peroxidation against the increase of antioxidant system activity.

Objective: Bacoside A is a major bioactive constituent of Bacopa monnieri L having antioxidant property. The objective of this study was to evaluate the effect of Bacoside A, on lipid peroxidation in brain, heart and liver during induced aging.Methods: Male Swiss albino mice, Mus musculus was used for the present investigation. Four experimental groups were used as Group I-Normal adult, Group II-D-galactose induced, Group III-D-galactose induced plus Bacoside A treated and Group IV-Natural aging. The effect of Bacoside A was studied against lipid peroxidation during induced aging. The level of lipid peroxidation in the form of MDA formation was determined and measured in brain, heart and liver.Results: The statistical data obtained were analyzed using one way ANOVA, control vs other groups and results were expressed as mean±SE. In Bacoside A treated group the lipid peroxidation level in heart, brain and liver was significantly decreased (p<0.001) compared to control group. A significant increase (p<0.0001) in the level of lipid peroxidation was observed in D-galactose induced mice. In natural aging group highly significant increase (p<0.0001) in initial lipid peroxidation, ascorbate dependent lipid peroxidation and spontaneous lipid peroxidation was observed.Conclusion: The observations revealed that, lipid peroxidation was reversed in Bacoside A treated group which may be due to antioxidant property of Bacoside A. Thus Bacoside A is able to ameliorate the stress induced changes in lipid peroxidation during aging. The findings also provide a theoretical basis for the development of novel therapeutic formulations, such as antioxidant supplementation to boost antioxidant defenses in the body.

Infrared spectroscopy of saliva is an express and non-invasive method of analysis, applicable for diagnostics of various diseases and for studying metabolic processes and adaptive changes in the body. The goal of this study was to determine possibility of analyzing the products of lipid peroxidation (LPO) by using IR spectroscopy of saliva on the example of oncological diseases. The study involved 203 patients with lung cancer (n = 40), breast cancer (n = 50) and the control group (n = 113). Saliva samples were collected in the morning after overnight fast. The content of LPO products (conjugated dienes, and trienes, Schiff bases, malonic dialdehyde MDA) was determined in all samples and the IR absorption spectra were recorded in the range of 500–4000 cm–1. In the IR spectra, an increase in the intensity of the absorption bands of lipids was observed; it corresponded to an increase in the total lipid content and correlated with the content of MDA, and a decrease in the intensity of vibrations of oxygen-containing groups, which corresponded to a negative correlation with secondary LPO products. Apparently, on the IR spectra, we simultaneously register both primary, intermediate, and final LPO products. Statistically significant regression equations were obtained, allowing to estimate the content of intermediate LPO products - conjugated triene, and Schiff bases. The proposed method allows to monitor LPO processes, as well as to characterize the direction of the equilibrium shift in these processes.

Reactive oxygen species (ROS) are essential for proper functioning of the body. It is important to strike a balance between the production of ROS and their removal by the action of the antioxidant system. The deterioration of this balance may lead to oxidative stress, which causes damage to cell components of all – of lipids, proteins and DNA. Lipid peroxidation is a chain and free-radical process of lipid oxidation, in the course of which are involved ROS. The lipid peroxidation may play an important role in the pathogenesis of psoriasis. The aim of the study was to determine the concentration of thiobarbituric acid reactive substances, malondialdehyde and conjugated dienes, and the activity of superoxide dismutase in patients with psoriasis. Material and methods: The study group consisted of 12 women and 24 men with psoriasis – patients of the Department of Dermatology, Sexually Transmitted Infections and Immunodermatology CM UMK. The control group consisted of 16 healthy volunteers. The material for the study was venous blood. The concentration of TBARS and CD was determined in plasma and erythrocytes. The concentration of MDA was determined in plasma and SOD activity in the erythrocytes. Results: Statistically significantly higher concentration of MDA and statistically significantly higher concentration of TBARS was revaled in patients with psoriasis as compared to control group. There were no statistically significant differences in plasma concentrations of CD among patients with psoriasis and control group. In erythrocytes of patients concentration of CD was twice as high. There were no statistically significant differences in the activity of SOD between the patient group and the control group. Conclusions: The most important product of lipid peroxidation responsible for the formation of psoriatic lesions is MDA. Lack of changes in the concentration of the primary products of lipid peroxidation, with significant changes in the concentration of secondary products of the process in blood of studied subjects can provide a significant degree of severity of damage to the cells.

Chronic atrophic gastritis and gastric cancer represent distinct steps of one pathogenic process. The risk of developing cancer of the stomach is directly proportional to the degree of atrophic changes simultaneously detected in antral segment and in the body of the stomach. The role of immune system in transformation of precancerous diseases into cancer is beyond doubt. During development of the malignant disease, the changes in lipid peroxidation systems – antioxidant defense become significant and contribute to the progression of the tumor and the development of metastases. A simultaneous study of lipid peroxidation and antioxidant defense indices along with phagocytic activity will allow us to evaluate relative contribution of these processes to development of chronic atrophic gastritis and gastric cancer. Purpose of the present study was to assess correlations between the lipid peroxidation indices, i.e. antioxidant protection, and chemiluminescent activity of neutrophilic granulocytes and monocytes in chronic atrophic gastritis and gastric cancer. Forty patients with chronic gastritis, 22 patients with chronic atrophic gastritis and 40 patients with gastric cancer were examined. The control group consisted of 50 practically healthy age-matched volunteers. Evaluation of spontaneous and induced production of reactive oxygen species by neutrophils and monocytes was carried out by chemiluminescent analysis. The parameters of lipid peroxidation/ antioxidant protection were determined by spectrophotometric methods. Statistical data processing was carried out using the Statistica v. 8.0 program (StatSoft Inc., USA). The normal distribution of indices was tested using the Kolmogorov–Smirnov method (adjusted by Lillefors). Quantitative indicators, given the normal distribution, were described using the median (Me) and interquartile scatter (Q0.25-Q0.75). To study statistical significance of differences between quantitative characteristics, the Mann–Whitney test was used. To study strength of relationships of these indicators, the Pearson rank correlation coefficient (r) was calculated. The critical significance level (p) when testing statistical hypotheses was taken equal to 0.05. Correlation analysis showed that the weight of positive correlations increases in patients with chronic atrophic gastritis, and it decreases in patients with gastric cancer, the strength of the correlation dependence and new relationships appear between chemiluminescent activity of neutrophils and monocytes in a spontaneous and induced state, and the amounts of malonic dialdehyde, enzyme activities of superoxide dismutase and catalase. In chronic atrophic gastritis and gastric cancer we have established the features of correlation patterns between lipid peroxidation/antioxidant protection indices, and activity of neutrophils and monocytes.

The proportion of patients with multiple unsuccessful attempts of assisted reproductive technology (ART) is about 30% of all patients treated with the use of ART. Women with history of unsuccessful ART attempts - a special category of patients who require emergency attention and a thorough examination at the stage of preparation for superovulation stimulation,the selection of embryos and endometrium preparation for embryo transfer. The objective: to distinguish high-risk group of unsuccessful attempts based on a detailed analysis of anamnestic and clinical data of infertile women with repeated unsuccessful ART attempts that requires more in-depth study of hormonal features, ovarian reserve and condition of the endometrium. Materials and methods. For better understanding of the problem of repeated unsuccessful ART attempts and сreation of efficient infertility treatment algorithms for these couples we conducted a thorough analysis of anamnestic data of three groups of infertile women (105 patients), which were distributed by age: group I – younger than 35, the II group – from 35 to 40, the III group - over 40 years. These groups of patients were compared with each other and with the control group of healthy women (30 persons). Results. Leading stress factors in the percentage three times prevailed in the group of infertile women and had a direct connection with the fact of procedure «fertilization in vitro» and chronic stressors caused by prolonged infertility. Primary infertility was observed significantly more frequent in patients younger than 35 years (p <0.05), secondary infertility - mostly in the second and third experimental groups (p <0.05). Noteworthy significant percentage of wellknown causes of infertility and idiopathic factor in all groups, and the prevalence of tubal-peritoneal factor in the second and third experimental groups, and endocrine dysfunction in the I experimental group. The most common disorder among this category of woman was polycystic ovary syndrome. Frequency of usual miscarriage among patients of I ana II groups was two times higher than in the third group (p <0.05). Among the experimental groups the leading place belongs urinary tract infection, respiratory tract diseases, pathologies of the cardiovascular system. Data of the stratified analysis show an increase likelihood of repeated unsuccessful ART attempts under the influence of constant chronic stress (odds ratio OR=2.06; 95% CI: 0.95–3.17; p<0.05). Conclusions. Among infertile patients with repeated unsuccessful ART attempts must be separated a high risk group of failures. The identity depends on the duration of infertility, female age and leading combination of factors. Key words: repeated unsuccessful ART attempts, anamnesis, infertility, high risk.

Resistance of erythrocytes from Brown trout (Salmo trutta m. trutta L.) affected by ulcerative dermal necrosis syndrome In the present work we evaluated the effect of ulcerative dermal necrosis (UDN) syndrome on resistance of erythrocytes to haemolytic agents and lipid peroxidation level in the blood from brown trout (Salmo trutta m. trutta L.). Results showed that lipid peroxidation increased in erythrocytes, as evidenced by high thiobarbituric acid reactive substance (TBARS) levels. Compared to control group, the resistance of erythrocytes to haemolytic agents was significantly lower in UDN-positive fish. Besides, UDN increased the percent of hemolysated erythrocytes subjected to the hydrochloric acid, urea and hydrogen peroxide. Results showed that UDN led to an oxidative stress in erythrocytes able to induce enhanced lipid peroxidation level, as suggested by TBARS level and decrease of erythrocytes resistance to haemolytic agents.

Intraperitoneal injection of ferric nitrilotriacetate (Fe-NTA) to rats and mice results in iron-induced free radical injury and cancer in kidneys. This study was designed to investigate the effect of catechin, a bioflavonoid with antioxidant potential, on Fe-NTA-induced nephrotoxicity in rats. Four groups were employed in the present study. Group I served as control group, Group II animals received Fe-NTA (8 mg iron/kg body weight i.p.), Group III animals were given 40 mg/kg catechin p.o. twice a day for 4 days and on the 5th day Fe-NTA was challenged, and Group IV animals received catechin alone for 4 days. Renal function was assessed by measuring plasma creatinine and blood urea nitrogen. The oxidative stress was measured by renal malondialdehyde levels, reduced glutathione levels and by enzymatic activity of catalase, glutathione reductase and superoxide dismutase. One hour after a single intraperitoneal (i.p.) injection of Fe-NTA (8 mg iron/kg), a marked deterioration of renal architecture, renal function and severe oxidative stress was observed. Pretreatment of animals with catechin markedly attenuated renal dysfunction, reduced elevated thiobarbituric acid reacting substances (TBARS), restored the depleted renal antioxidant enzymes and normalized the renal morphological alterations. These results clearly demonstrate the role of oxidative stress and its relation to renal dysfunction, and suggest a protective effect of catechin on Fe-NTA-induced nephrotoxicity in rats.

The aim of this study was to investigate the clinical efficacy of vanadium complexes on triglycerides (TG), total cholesterol (Chol), uric acid (UA), urea (U), and antioxidant parameters: nonenzymatic (FRAP—ferric reducing ability of plasma, and reduced glutathione—GSH) and enzymatic (glutathione peroxidase—GPx, catalase—CAT, and GPx/CAT ratio) activity in the plasma of healthy male Wistar rats. Three vanadium complexes: [VO(bpy)2]SO4⋅2H2O, [VO(4,4′Me2bpy)2]SO4⋅2H2O, and Na[VO(O2)2(bpy)]⋅8H2O are administered by gavage during 5 weeks in two different diets such as control (C) and high fatty (F) diets. Changes of biochemical and antioxidants parameters are measured in plasma. All three vanadium complexes statistically decrease the body mass growth in comparison to the control and fatty diet. In plasma GSH was statistically increased in all vanadium complexes-treated rats from control and fatty group in comparison to only control group. Calculated GPX/CAT ratio was the highest in the control group in comparison to others.
