Experiments have been carried out on explanted chick embryos to test certain widely accepted concepts about the role of Hensen's node in somite formation. The relationship between elongation of the area pellucida and regression of Hensen's node has also been investigated. We conclude from these experiments that: (a) The timing of somite formation is not controlled by the regression of Hensen's node, nor by the shearing of the mesoderm into right and left halves. (b) Somite size and shape are probably controlled by local conditions in the chick embryo. (c) Elongation and regression are two different events. (d) The position of the somites probably depends on mechanical tensions in the area pellucida. (e) The notochord is not required for the stability of somites in vivo.

AbstractInterleukins, a group of cytokines participating in inflammation and immune response, are proved to be involved in the formation and development of pulmonary fibrosis. In this article, we reviewed the relationship between interleukins and pulmonary fibrosis from the clinical, animal, as well as cellular levels, and discussed the underlying mechanisms in vivo and in vitro. Despite the effects of interleukin-targeted treatment on experimental pulmonary fibrosis, clinical applications are lacking and unsatisfactory. We conclude that intervening in one type of interleukins with similar functions in IPF may not be enough to stop the development of fibrosis as it involves a complex network of regulation mechanisms. Intervening interleukins combined with other existing therapy or targeting interleukins affecting multiple cells/with different functions at the same time may be one of the future directions. Furthermore, the intervention time is critical as some interleukins play different roles at different stages. Further elucidation on these aspects would provide new perspectives on both the pathogenesis mechanism, as well as the therapeutic strategy and drug development.

To investigate the possible contributions to dysphoria of interactions among attributional dimensions, 105 freshmen and sophomores were administered the Attributional Style Questionnaire and the Beck Depression Inventory. Analyses examined the relationship to dysphoria of (a) the traditional composite score; (b) multiple regression analyses including interactions among the various dimensions; and (c) indices of behavioral self-blame, characterological self-blame, and external blame. The results provided modest support for the specific hypothesized interactional model and, to a large extent, appeared to support the validity of the standard manner in which dysphoric attributional style is viewed. Refinements of the traditional model are suggested, involving the self-blame construct, the possible role of the stability dimension, and the relationship between controllability and positive event attributions.

This article reflects the peculiarities of the psychological connection between a financial fraudster and his potential victim. The process of forming a stressful situation depending on the type of financial fraud is described, the reasons for its occurrence and the result of implementation associated with a decrease in critical thinking are indicated. The essence is also revealed, including from the perspective of the relationship between the fraudster and the potential victim, and the types of financial fraud and practical examples of their manifestation are considered. The psychological portrait of a financial fraudster and his transformation in connection with the changing preferences of consumers of financial services are described. The role of the Bank of Russia in reducing the activity of financial fraud and ensuring the stability of the financial market is reflected.

This article investigates the relationship between the perception of violence and the spending on security goods and services in households. Individual microdata from a random national survey on family budget carried out in Brazil in 2008-2009 were used for modeling the household spending using two instrumental variables. The stability of results was checked by applying the Lasso-Gaussian regularization method in the selection of the statistically significant variables. Positive relationships were found between household spending on security goods and services and (i) the fear of insecurity at the household level, (ii) the neighbors’ spending on security, and (iii) the registered criminality, but no evidence was found on the relationship between the role of police on household spending on security goods and services.

The effect of leukocyte antibodies detected under different conditions on the fate in vivo of granulocytes was studied using 111-indium- labeled granulocytes. Sera from patients were tested by granulocyte agglutination (GA), granulocytotoxicity (GC), granulocyte immunofluorescence (GIF), lymphocytotoxicity (LC), and antibody- dependent lymphocyte-mediated granulocytotoxicity. Granulocytes from donors to be studied were labeled with 111-indium and injected. Then the intravascular recovery and survival or tissue localization was determined in 93 studies. Antibodies detected by granulocyte agglutination were associated with a significant reduction in recovery (6.7% v 30.8% in controls; P less than .001) and t1/2 (0.3 hours v 5.6 hours in controls; P = .002). When all possible combinations of serum reactivity were considered, reactivity in the GA plus GIF assays had the best correlation with decreased recovery (R2 = .49; P less than .001) and t1/2 (R2 = .73; P less than .001). When the relationship between the strength of antibody reactivity and the recovery and t1/2 were analyzed, the best relationship was between the combination of LC and GIF with recovery (R2 = .62; P = .001). Because of the general availability of the HLA (LC) testing, the role of LC reactivity was investigated in other ways. There was a strong relationship between sera highly reactive by LC and those reactive by GIF. These highly reactive sera were also associated with reduced recovery and t1/2. The influence of specific HLA antigen mismatches was also studied. When donor and recipient were mismatched for the HLA-A2, B8, or BW44 antigens, there was a significant reduction in either recovery, t1/2, or both. Tissue localization was studied by body scans in patients with and without known sites of inflammation. Antibodies detected by a combination of GA and GIF caused abnormal pulmonary sequestration of granulocytes (three cases) and failure of granulocytes to localize at known sites of inflammation (three cases). HLA (LC) antibodies did not alter tissue localization despite the presence of the corresponding HLA antigens on granulocytes. It appears that GA, GIF, or a combination of these tests is the most effective predictor of altered in vivo fate of granulocytes. However, sera highly reactive by LC and GIF probably define a group of highly immunized patients in whom granulocyte recovery and t1/2 are also reduced. Mismatching for certain HLA antigens is also associated with reduced granulocyte recovery and survival. At present, GA, with or without the immunofluorescence assay, is the most effective predictor of altered in vivo granulocyte activity.(ABSTRACT TRUNCATED AT 400 WORDS)

AbstractN6-methyladenosine (m6A) modification is the most prevalent modification in eukaryotic RNAs while accumulating studies suggest that m6A aberrant expression plays an important role in cancer. HNRNPA2B1 is a m6A reader which binds to nascent RNA and thus affects a perplexing array of RNA metabolism exquisitely. Despite unveiled facets that HNRNPA2B1 is deregulated in several tumors and facilitates tumor growth, a clear role of HNRNPA2B1 in multiple myeloma (MM) remains elusive. Herein, we analyzed the function and the regulatory mechanism of HNRNPA2B1 in MM. We found that HNRNPA2B1 was elevated in MM patients and negatively correlated with favorable prognosis. The depletion of HNRNPA2B1 in MM cells inhibited cell proliferation and induced apoptosis. On the contrary, the overexpression of HNRNPA2B1 promoted cell proliferation in vitro and in vivo. Mechanistic studies revealed that HNRNPA2B1 recognized the m6A sites of ILF3 and enhanced the stability of ILF3 mRNA transcripts, while AKT3 downregulation by siRNA abrogated the cellular proliferation induced by HNRNPA2B1 overexpression. Additionally, the expression of HNRNPA2B1, ILF3 and AKT3 was positively associated with each other in MM tissues tested by immunohistochemistry. In summary, our study highlights that HNRNPA2B1 potentially acts as a therapeutic target of MM through regulating AKT3 expression mediated by ILF3-dependent pattern.

Menggunakan metode penelitian yuridis normatif dengan pendekatan perundang-undangan dan pendekatan konsep. We explored the relationship in chick embryos between somitogenesis and the onset of somite myogenesis by immunodetection of the muscle-specific intermediate filament protein desmin. Early somite desmin expression was detected by whole-mount in situ confocal microscopy. No detectable somite desmin was observed in embryos of 15 somites (Stage 12) or younger. In embryos having between 16 and 26 somites (Stages 12-15), desmin could be detected in somites positioned increasingly more caudal in the embryo. Finally, in embryos of 27 somites (Stage 16) and older, somite desmin expression was consistently present in all but the caudal-most six somites. Although the rate of somite formation is fairly constant, the rate of observed somite desmin expression progressing caudally in the embryo is greater initially than the rate of segmentation. After an embryo has formed about 27 somites, the rate of desmin appearance parallels the rate of segmentation at a distance of about six somites. This result suggests that very early somite myogenesis is not linked to somitogenesis.

: Copper is an essential element for most aerobic organisms, with an important function as a structural and catalytic cofactor, and in consequence, it is implicated in several biological actions. The relevant aspects of chemistry and biochemistry and the importance of copper compounds in medicine give us a comprehensive knowledge of the multifaceted applications of copper in physiology and physiopathology. In this review, we present an outline of the chemistry and the antitumor properties of copper complexes on breast, colon, and lung cancer cells focus on the role of copper in cancer, the relationship between structure-activity, molecular targets, and the study of the mechanism of action involved in its anticancer activity. This overview is expected to contribute to understanding the design, synthesis, uses of copper complexes as antitumor agents in the most common cancers.

The blastulae of unincubated eggs of the quail, Coturnix coturnix japonica, have been bisected in ovo, using the technique of Lutz (1949). Some embryos were harvested after 24 hand found to possess two primitive streaks. Most were fixed at 48 h or 72 h. Some were found to have regulated to form almost normal single axes, whilst others had developed into duplicitas anterior embryos, separate twins or collided axes. All three types of twinned embryos were smaller than the control embryos. The number of somites was not however reduced in the shorter embryos. This finding corresponds with a similar result obtained by Cooke (1975) who reported that if a Xenopus blastula is reduced in size, it nevertheless develops the correct number of somites. The quail however adjusts the shape of the individual somites so that they fit into the reduced body length, whereas Xenopus reduces the size of somites. No miniaturized somites were ever seen in these quail embryos. As a result of the present experiments, it was concluded that the length of incubation time does not directly control the rate of somite formation, because different numbers of somite were found in twins which possessed identical genomes and had developed in almost the same environment for identical periods. In addition, the size of the area pellucida does not appear to control somite formation. Probably, the most important influence is the regression of the node.
