The leaves of Juniperus sabina (Cupressaceae) are used in traditional Uygur medicine for the treatment of rheumatism and arthritic pain. This study aimed to investigate the analgesic and anti-inflammatory effects of total flavonoids from leaves of Juniperus sabina (JSTF) on rodents. The anti-inflammatory activity was investigated using the carrageenan, egg albumin, or histamine-induced rat paw edema as well as xylene-induced ear edema, capillary permeability, and cotton pellet granuloma while the antinociceptive activity was evaluated using the mouse writhing, formalin, and hot-plate tests. JSTF (125, 250, 500 mg/kg) significantly inhibited xylene-induced ear edema in mice (inhibition ratio as 16.22%, 40.67%, and 51.78%, respectively) and also significantly ameliorated acetic acid increased vascular permeability in mice (inhibition ratio as 11.63%, 32.56%, and 53.49%, respectively). JSTF (250 and 500 mg/kg) gave significant reduction of carrageenin-induced paw oedema at the interval of 1 h and 5 h. Administration of JSTF (500 mg/kg) caused a significant anti-inflammatory effect against oedema induced by egg albumin or histamine at the interval of 0.5 h and 4 h, and both which induced the paw oedema were also inhibited by JSTF (250 mg/kg) at a point in 1, 2, or 3 h after the inflammation. Furthermore, JSTF (125, 250, and 500 mg/kg) produced time-course increase in pain threshold in hot-plate test also. JSTF produced concentration-dependent inhibition on cyclooxygenase-2 (COX-2) or 5-lipoxygenase (5-LO) activities in vitro, and their IC50 values were 31.92 and 129.26 μg/mL, respectively. Moreover, JSTF significantly caused a significant dose-dependent inhibition on acetic acid induced writhing response in mice (inhibition ratio as 23.27%, 36.91%, and 50.76%, respectively). JSTF also significantly ameliorated formalin-induced pain in mice in the late phase on dose-dependent way. These results confirms the clinical use of J. sabina for treatment of rheumatoid arthritis in ethnomedicine, and its effective mechanism will be further studied in the future.

Sophora viciifolia Hance is an edible plant used in traditional Chinese medicine. Sophocarpine, a tetracyclic quinolizidine alkaloid, is one of the most abundant active ingredients in Sophora viciifolia Hance. Here, we study the analgesic and anti-inflammatory effects, as well as the acute toxicity of sophocarpine from Sophora viciifolia Hance in mice. Sophocarpine (20, 40, and 80 mg/kgbw) significantly prolonged the delay period before a hot plate reaction occurred (all P < 0.05 ), and the delay before a tail-flick response was induced by a warm bath ( P < 0.05 ; P < 0.01 ). Sophocarpine (40, 80 mg/kg) resulted in dose-dependent inhibition of the writhing reaction induced by acetic acid in mice ( P < 0.05 ; P < 0.001 , respectively). Sophocarpine (80 mg/kg) reduced the total duration of a formalin-induced pain response ( P < 0.05 ). Sophocarpine prolonged the foot-licking latency of mice after the hot plate reaction, and this effect was antagonized by calcium chloride and enhanced by verapamil. Sophocarpine (20, 40, and 80 mg/kg) significantly inhibited xylene-induced ear edema ( P < 0.01 ; P < 0.001 ; P < 0.001 , respectively) and the penetration of acetic acid-induced dye into the peritoneal cavity ( P < 0.01 ; P < 0.01 ; P < 0.001 , respectively). It also reduced the levels of proinflammatory cytokine interleukin (IL)-1β, IL-6, and prostaglandin E2 ( P < 0.05 , P < 0.01 , P < 0.001 ) and those of serum nitric oxide ( P < 0.05 ). The results of this study suggest that sophocarpine possesses certain analgesic and anti-inflammatory activities, which may be related to calcium and inhibition of the secretion of inflammatory factors.

Summary Introduction: Potato (Solanum tuberosum L.) is an important vegetable crop in Syria. Potato tuber moth Cussonia barteri is a small tree that grows in the sub-Saharan part of Africa. Various parts of the plant are used for the treatment of a variety of ailments in ethno-medicine. Objective: To evaluate the anti-inflammatory and analgesic effect of the methanol leaf extract of Cussonia barteri. Material and methods: The leaves were air-dried, powdered and repeatedly extracted with methanol using a Soxhlet apparatus. The resulting methanol extract (100, 200 and 400 mg/kg) was evaluated for anti-inflammatory activity using carrageenan-induced paw oedema, xylene-induced ear oedema and formalin-induced arthritis tests. Analgesic effect was evaluated using acetic acid-induced mouse writhing, hot plate and tail flick tests. Results: All doses of the extract significantly (p<0.05) reduced carrageenan-induced paw oedema, however the 400 mg/kg dose gave a sustained effect. The extract significantly inhibited xylene induced ear oedema at all doses. There were no significant (p>0.05) reductions in paw swellings due to formalin. In the acetic acid induced writhing test, the extract significantly (p<0.05) decreased writhing at 400 mg/kg only. Reaction times were not significantly different from the control in the hot plate and tail flick tests. Conclusion: This study has shown that the methanol extract possesses acute anti-inflammatory and peripherally mediated analgesic effects.

Background: Smilax zeylanica Linn (Smilaceae) is a woody climbing shrub native to Asia's tropical and subtropical highlands. Traditionally, this plant has been widely used to cure a variety of ailments. Smilax zeylanica stem bark was extracted with ethanol to test its antinociceptive and anti-inflammatory properties. Objective: To study the antinociceptive and anti-inflammatory activities of ethanolic stem bark extract of Smilax zeylanica Linn. in Wistar rats. Methods: The antinociceptive activity has been assessed in mice by formalin-induced paw licking, Eddy’s hot-plate, and acetic-acid induced abdominal stretching models. Carrageenan-induced paw oedema in rats has been conducted to explore the anti-inflammatory activity. Tween 80 (10ml/kg, p.o) was given to the control group, test drug group received ethanol extract of Smilax zeylanica (EESZ) orally at doses of 200 and 400 mg/kg, and standard drugs morphine (5 mg/kg, s.c), diclofenac sodium (10 mg/kg, i.p) were injected to the standard drug group. Results: Oral administration of ethanolic extract of Smilax zeylanica (EESZ) at test doses (200 and 400 mg/kg), produced a significant decrease of the paw licking time for 2 phases (neurogenic and inflammatory) in the formalin test, upsurge of the reaction time in Eddy’s hot plate test, and significantly decreased the abdominal stretching in the acetic acid-induced  writhes test. Ethanol extract of Smilax zeylanica significantly decreased the carrageenan-induced paw oedema. Conclusion: The ethanol extract of Smilax zeylanica possesses antinociceptive, anti-inflammatory activity, probably involving central and peripheral pathways

Uapaca bojeri is an endemic Malagasy plant used by the local population. This work aimed to evaluate antioxidant, anti-inflammatory, and antidiabetic activities of the methanol extracts of U. bojeri leaves and stems and to report their total phenolic content and the bioactive compound content by HPLC methods. Antioxidant capacity was determined by DPPH and ferric reducing antioxidant power (FRAP) assays. An in vivo carrageenan-induced paw oedema and acetic acid-induced writhing test in mice were used for anti-inflammatory activity evaluation. An oral glucose tolerance test was performed in mice to evaluate antidiabetic activity. The total bioactive compound content of leaves was higher than that of stems. Stem methanol extract inhibited the free radical DPPH more than the leaf methanol extract. Leaf methanol extract inhibited, in a dose-dependent manner, the carrageenan-induced paw oedema more than the stem extract, but their inhibition of the pain symptoms caused an acetic acid-induced decrease similar to the number of writhes in the dose-dependent case. The leaf and stem methanol extracts significantly reduced blood glucose levels after 30 min of glucose loading in mice compared to the control group blood glucose reduction. The presence of several bioactive compounds in U. bojeri contributed to the different biological activities, but isolation and identification of these bioactive molecules are necessary to confirm these pharmacological properties.

Symptoms of Coronavirus Disease-2019 include pulmonary hyper-inflammation; managing local and systemic inflammatory responses may be key in treatment. This study evaluated the anti-inflammatory, analgesic and antipyretic effects of a mixture (Cov-Pla 2) containing five medicinal plants in laboratory animals. The antinociceptive activity of Cov-Pla 2 was evaluated using acetic acid-induced writhing, Brewer’s Yeast Pyrexia test in rats and hot plate tests in mice. The egg albumin-induced rat paw oedema test was employed to evaluate the extract’s anti-inflammatory activity. The extract produced a dose-dependent (125-500 mg/kg, p.o.) inhibition of pain response elicited by acetic acid, compared to normal saline and increased reaction latency in the hot plate test. The antiinflammatory test showed a significant (P<0.05) reduction in paw size diameter at 125 mg/kg from 2 hours, compared to control. In the antipyretic test, the extract produced a significant (P<0.05) time-dependent decrease in rectal temperature at 125 mg/kg after 2 and 3 hours and at 250 and 500 mg/kg after 3 hours, compared to normal saline. The extract did not produce mortality up to 5000 mg/kg per oral. The results indicated that the extract possesses antinociceptive and anti-inflammatory properties, which support its recommendation for trials in the treatment of symptoms associated with COVID-19. Aim: This research was designed to evaluate the anti-inflammatory and analgesic activities of Parinari curatellifolia methanol leaf extract in albino rats. Methodology: Phytochemical screening was carried out using standard methods. Anti-inflammatory activity of the extract was done using egg albumin and formalin induced hind paw edema model. Analgesic effect was evaluated using hot plate induced pain and acetic acid induced writhing test. For each model twenty (20) rats were used, divided into five (5) groups of four (4) rats each. Results: Parinari curatellifolia revealed the presence of alkaloids, flavonoids, tannins and phenols while steroids, anthraquinone, terpenoids and glycoside were not detected. For the egg albumin induced inflammation, the group treated with the standard drug (indomethacin) and the group that received the highest dose of the extract were significantly lower (P<0.05) than all the other groups with percentage inhibitions at 25.56% and 24.44% respectively there was no significant difference (P>0.05). For the formalin induced anti-inflammatory activity, at the 1st hour, the normal control group had its paw volume significantly different (P<0.05) from the treated groups. This trend was observed at the 2nd, 3rd and 4th hour. The hot plate method results revealed significant increased (P<0.05) in the analgesic activity of PCMLE at 400mg/kg body weight and the drug treated group when the control was compared with the treated groups with percentage inhibition of 34.32% and 52.94% respectively. The acetic acid induced writhing test revealed that the extract at the three doses of 100, 200 and 400 mg/kg body weight, showed a significant (P<0.05) percentage inhibition of 32.31%, 36.92% and 47.69%, respectively compared to negative control. Conclusion: This justifies the use of Parinari curatellifolia locally in the management of pain and inflammation.

Anti-inflammatory potential of the ethanol leaf extract of Sphenocentrum jollyanum (ELESJ) was assessed in-vivo in mice by using carrageenan-induced paw oedema, egg albumin-induced paw oedema, and xylene-induced ear oedema model at doses of 474.34, 948.68, and 1423.03 mg/kgbw respectively. A total of seventy-five matured mice of both sexes weighing between 20 g and 24 g (divided into three groups of 25 mice each for the 3 different studies) were used for this study. Treatment was done intraperitoneally. Distilled water (10 mL/kgbw), acetylsalicylic acid (ASA) (100 mg/kgbw) or dexamethasone (4 mg/kgbw) served as the normal and standard drugs respectively. The results showed that ELESJ produced significant (p < 0.05), dose-dependent and time-dependent reductions in mean paw thickness in groups III-V compared with control group. Similarly, the extract caused a dose-dependent increase in percentage inhibition of carrageenan-induced paw oedema and egg-albumin induced paw oedema (34.49%, 36.71%) at the highest dose (1423.03 mg/kg), comparable with the anti-inflammatory effects of the standard drug ASA (34.69%, 31.54%). The ELESJ also caused significant (p < 0.05), dose-dependent decreases in xylene-induced ear oedema weight in all the test groups relative to control group and standard drug group. In addition, ELESJ showed a dose-dependent percentage inhibitory effect (44%) at the highest dose (1423.03 mg/kg) comparable with that of the standard drug, dexamethasone (48%). The results obtained from this study support the use of the leaf in folk medicine as it has significant anti-inflammatory properties and this may be attributed to the phytochemical constituents of the leaf.

Antinociceptive and anti-inflammatory potentials of methanolic extract ofBridelia retusafruit (BRME) were evaluated against different animal models in rodents. Antinociceptive effects of BRME were assessed in mice using the acetic acid-induced writhing and formalin test. Anti-inflammatory effects of BRME in three different doses, namely, 100, 200, and 400 mg/kg, were evaluated by utilizing different animal models representing various changes associated with inflammation, namely, carrageenan-induced paw oedema, histamine and serotonin-induced paw oedema, arachidonic acid-induced paw oedema, formalin-induced paw oedema, TPA-induced ear oedema, acetic acid-induced vascular permeability, total WBC count in paw fluid, and myeloperoxidase assay. Also BRME was phytochemically evaluated using chromatographic method. The BRME did not exhibit any signs of toxicity up to a dose of 2000 mg/kg. The extract showed statistical significant inhibition of induced nociception and inflammation in dose dependent manner. The higher dose of extract significantly inhibited pain and inflammation against control (P<0.001). HPLC results revealed the presence of gallic acid and ellagic acid as phytoconstituents in BRME and it was proven as anti-inflammatory agents. The present study scientifically demonstrated the antinociceptive and anti-inflammatory potential of fruit ofB. retusamethanolic extract. These effects may be attributed to the presence of polyphenolic phytoconstituents in the extract.

The present study was aimed to evaluate the analgesic and anti-inflammatory properties ofCastanospermum australeand to profile phytochemicals by GC-MS. The ethanolic extracts were prepared by successive solvent extraction using Soxhlet apparatus. The analgesic activity was analyzed by hot plate method and acetic acid-induced writhing test whereas anti-inflammatory study was done by carrageenan induced paw oedema model. The acute toxicity study revealed that ethanol extracts of leaf and bark ofC. australewere safe even at a higher dose of 2000 mg/kg whereas ethanol extract of seed was toxic at the same dose. In both hot plate method (5.85 s) and acetic acid-induced writhing test (57%), the leaf ethanol extract exhibited significant analgesic activity (P<0.001) at a dose of 400 mg/kg. The anti-inflammatory activity of leaf extract was exhibited by the reduction in paw linear diameter by 64.76% at 400 mg/kg in carrageenan induced paw oedema. The GC-MS analysis of the ethanol extract of leaf revealed sixteen major compounds of which 1,7-dimethyl-4,10-dioxa-1,7-diazacyclododecane, (+)-N-methylephedrine, and permethylspermine were found to be pharmaceutically and the most important. These findings justify thatC. australecan be a valuable natural analgesic and anti-inflammatory source which seemed to provide potential phytotherapeutics against various ailments.

The aim of the study was to assess the polyphenolic profile, anti-inflammatory and antinociceptive activity of Arctium lappa, a medicinal plant traditionally used in the treatment of gout, hepatitis and other inflammatory disorders. Polyphenolic profile of a hydro-glycero-ethanolic extract from Arctium lappa roots (ALE) was evaluated by HPLC-MS method. Anti-inflammatory effect of the Arctium lappa extract (ALE) was determined by carrageenan-induced rat paw oedema test, while antinociceptive effect was determined by acetic acid induced writhing test in mice and Randall Selitto test in rats. HPLC-MS analysis of the extract showed the presence of chlorogenic acid (158.9 μg/mL) and quercitrin (14.4 μg/mL). The administration of ALE reduced the oedema formation in the carrageenan-induced rat paw oedema test, especially at dose of 500 mg/kg, the results being statistically significant and dose-dependent. Also, ALE showed statistically significant and dose-dependent antinociceptive effects in the acetic acid induced writhing test in mice and Randall Selitto test in rats. The results of the pharmacological experiments suggest that the anti-inflammatory and antinociceptive effects of the Arctium lappa extract (ALE) may be related to the ability of polyphenols such as chlorogenic acid to inhibit the synthesis and release of some pro-inflammatory mediators. Our experimental data justify the traditional use of this plant in the management of some inflammatory disease.
