The pathogenesis of chronic spontaneous urticaria (CSU) has not been fully understood; nevertheless, significant progress has been achieved in recent years. The aim of this study was to investigate the possible role of reactive oxygen species (ROS) in the pathogenesis of CSU. Sixty-two children with CSU and 41 healthy control subjects were enrolled in the study. An extensive evaluation of demographic and clinical features was done, and serum oxidative stress was evaluated by plasma total oxidant status (TOS) and total antioxidant status (TAS) measurements. The median value of plasma TOS was found to be 10.49 μmol H2O2 equiv./L (interquartile range, 7.29–17.65) in CSU patients and 7.68 μmol H2O2 equiv./L (5.95–10.39) in the control group. The difference between the groups was statistically significant (p=0.003). Likewise, the median plasma TAS level in the CSU group was decreased significantly compared to that of the control group (2.64 [2.30–2.74] versus 2.76 [2.65–2.86] mmol Trolox equiv./L, resp.,p= 0,001). Our results indicated that plasma oxidative stress is increased in children with CSU when compared to healthy subjects, and plasma oxidative stress markers are positively correlated with disease activity.

Background: Diabetes Mellitus (DM) is a progressive metabolic disorder. Objective: The aim of this study was to investigate the relationship between antioxidant and oxidative stress markers in the saliva of patients with type 2 DM and a healthy control group. Methods: In this study, 20 patients with diabetes and 20 healthy individuals were evaluated. Salivary antioxidants markers consisted of total antioxidant capacity (TAC), uric acid (UA), peroxidase and catalase. Oxidative stress markers included total oxidant status (TOS), malondealdehyde (MDA) and total thiol (SH). Sialochemical analysis was performed with spectrophotometric assay. All the statistical analyses were conducted using STATA software. Results: TAC decreased significantly in patients with diabetes. Although salivary UA and peroxidase were lower in patients with diabetes compared to the control group, the difference was not significant. Salivary catalase in patients with diabetes was significantly lower than that in the control group. MDA and TOS exhibited significantly higher levels in type 2 DM. SH levels were slightly higher in DM. Conclusions: According to the results of the present study, there were some changes in the salivary levels of some antioxidants and oxidative stress markers in patients with type 2 DM and could be measured as an indicator of serum changes..

ObjectiveSchizoaffective disorder is a disease with both affective and psychotic symptoms. In this study, we aimed to compare oxidative metabolism markers of schizoaffective disorder, bipolar disorder and schizophrenic patients. Furthermore, we also aimed to investigate whether schizoaffective disorder could be differentiated from schizophrenia and bipolar disorder in terms of oxidative metabolism.MethodsTotal oxidant status (TOS) and total antioxidant status (TAS) were measured in the blood samples that were collected from schizoaffective patients (n = 30), bipolar disorder patients (n = 30) and schizophrenic patients (n = 30). Oxidative stress index (OSI) was calculated by dividing TOS by TAS.ResultsTOS and OSI were found to be higher in patients with schizoaffective disorder compared with those in schizophrenia and bipolar disorder patients. TAS was not significantly different between the groups.ConclusionSchizoaffective disorder was found to be different from bipolar disorder and schizophrenia in terms of oxidative parameters. This result may indicate that schizoaffective disorder could differ from bipolar disorder and schizophrenia in terms of biochemical parameters. Increased TOS levels observed in schizoaffective disorder may suggest poor clinical course and may be an indicator of poor prognosis.

Objective: Carbon monoxide poisoning (COP) is one of the important causes of morbidity and mortality in toxicological cases. In this study, we aimed to find out more about the pathophysiology of COP by investigating the effects of COP on oxidative stress parameters such as total oxidant status (TOS) and total antioxidant status (TAS). Methods: Eighty-eight patients admitted to the emergency department of our hospital with acute COP and 35 healthy adults as control group were included in this study. Blood samples were collected from all COP patients at the time of initial emergency department evaluation to determine the oxidative stress parameters. Then, serum levels of total antioxidant status and total oxidant status levels were measured. Results: A total of 88 patients poisoned by carbon monoxide (CO; mean age 37.1 ± 18.2 years; 54’% women) were enrolled. TOS and carboxyhemoglobin (COHb) levels in COP patients were increased when compared to control group (p = 0.001). TOS, oxidative stress index (OSI) and COHb levels in COP patients were significantly lower after the treatment. (respectively, p = 0.016; p = 0.023; p = 0.001). On the other hand, no statistical differences were observed in TAS levels of study and control group as well as there were no changes with treatment. Conclusion: Measurements of TOS, TAS and OSI levels may be useful markers to find out the pathophsiology of COP.

Objective. Oxidative stress is considered to be the main factor in the development of diabetic complications and tissue injury. our objective was to investigate and compare the oxidant/antioxidant conditions and detoxification mechanisms of the liver, lung, kidney, cardiac tissues, and mitochondria of rats with diabetes induced by streptozocin (STZ).Methods. Rats with diabetes induced by streptozocin were anesthetized by administering 90 mg/kg ketamine hydrochloride and 3 mg/kg xylazine hydrochloride. Thoracic cavities were incised open; liver, lung, kidney, and cardiac tissues were removed and stored at−70°C. All samples were homogenized and mitochondrial fractions were separated. Total Antioxidant Status (TAS), Total Oxidant Status (TOS), Oxidative Stress Index (OSI), Paraoxonase (PON), Arylesterase, Catalase (Cat), Malondialdehyde (MDA), and Glutathion-S-transferase were measured in each fraction.Results. MDA and TOS levels were significantly increased in liver tissues, and T OS and OSI were increased in the mitochondrial fractions of diabetic rats. These increases were not statistically significant compared to the control group. No significant differences were determined in the antioxidant and GST activities.Conclusion. According to our results, oxidative stress has not developed in rats with diabetes induced by streptozocin. The detoxification system was induced; however, this induction did not differ significantly from the controls.

AbstractOxidative stress appears to play a role in the pathogenesis of several inflammatory gastrointestinal diseases. Increased homocysteine levels may play a role in the pathogenesis of Chron’s disease and ulcerative colitis. The aim of this study was to examine the influence of homocysteine on the antioxidant status of rat intestine and liver. The levels of thiobarbituric acid reactive substances (TBARS), activity of catalase (CAT) and total antioxidant status (TAS) were investigated in the isolated gut and liver of young male rats in the control group (8 rats) and after 3-hоur incubation in high doses of D, L-homocysteine thionolactone (Hcy) (10 μmol/L) (8 rats). Samples of duodenum, ileum, colon and liver were homogenized in sodium phosphate buffer (1:10). Homogenates were centrifuged at 10000 for 10 min at 4° C and the supernatant was taken for biochemical assays. Our results showed that high D, L-homocysteine thionolactone concentration reduced enzymatic catalase activity in homogenates of the isolated segments of duodenum (27.04%) p<0.01; ileum (37.27%), colon (34.17%) and liver (67.46%) p<0.001. Exposition to high D,L-homocysteine thiolactone concentration significantly increased TBARS levels in the duodenum (106.05%), ileum (47.24%), colon (112.75%) and liver (32.07%) (p<0.01). Homocysteine also modifi ed the total antioxidant status of homogenates from the duodenum, ileum, colon and liver, increasing by 20.68% (duodenum), 24.74% (ileum), 14.88% (colon) and 19.35% (liver) (p<0.001). Homocysteine induced a consistent oxidative stress in rat’s intestine and liver (reduced activity of catalase and increased level of TBARS), but the elevated activity of TAS in our experiments could be explained as an adaptive response to the generated free radicals which indicates the failure of the total antioxidant defense mechanism to protect the tissues from damage caused by homocysteine.

Abstract The purpose of this study was to examine the effects of antioxidants, including vitamin C, chitosan or a combination of both, on oxidative stress markers in muscles, as edible tissues of fish, exposed to paraquat. Fish exposed to 0.02 mg/L paraquat for 21 days were fed different diets: a normal diet, a diet containing chitosan (1000 mg/kg diet), a diet with vitamin C (1000 mg/kg diet) or both vitamin C and chitosan. Oxidative stress markers, including the activity of catalase, total antioxidant and malondialdehyde (MDA), as well as biochemical parameters including the activity of aspartate aminotransferase (AST), alanine aminotransferase (ALT), creatine phosphokinase (CPK), lactate dehydrogenase (LDH) and acetylcholinesterase (AChE), were measured in muscles. Fish exposure to paraquat increased LDH, CPK, catalase and MDA activity significantly, while it significantly decreased AST, ALT and AChE activity and total antioxidant capacity in muscles. Administration of vitamin C, combined with chitosan, to fish exposed to paraquat was effective in regulating AChE, AST, ALT, LDH, CPK and catalase activity. A significant increase in the total antioxidant status and a significant decrease in MDA levels were observed in fish fed chitosan-vitamin C complex. In conclusion, it is suggested that combined supplementation with vitamin C and chitosan may improve the detoxification system in the muscles of fish and protect common carp from paraquat toxicity.

Background: There is a well-documented cross-talk between the gut and brain. Evidence is accumulating to suggest beneficial effects of psychobiotics [prebiotics, probiotics or synbiotics] on psychological distress in disease states. However, their role in healthy status remains relatively unclear. Objectives: The present study was aimed to clarify if psychobiotics could influence behavioral responses and physiological stress in healthy rats. Methods: In the present experiment, 28 male Wistar rats were divided into four groups (healthy rats treated by Lactobacillus plantarum (L.plantarum), inulin and their combination (synbiotic) and control group). Then, psychobiotics were administered to the intervention groups for 8 weeks. Behavioral tests (Morris water maze, Elevated plus maze, and Forced swimming test) were performed at the endpoint. Then, serum and brain levels of superoxide dismutase, malondialdehyde, glutathione peroxidase, total antioxidant capacity, brain-derived neurotrophic factor (BDNF), and serotonin were measured. Results: Our finding indicated that unlike inulin, the administration of L.plantarum and synbiotic could ameliorate depression and anxiety-like behavior and cognitive performance (P<0.05). Serum and brain oxidative stress markers were significantly improved by synbiotic consumption. Intake of L.plantarum led to decreased oxidative stress in the hippocampus and amygdala (P<0.05). Moreover, a significant increase in hippocampal serotonin and BDNF concentration was observed after synbiotic and L.plantarum intake (P<0.05). In addition, there was a strong correlation of serum and brain markers with behavioral performance (P< 0.05). Conclusion: Our study suggests that psychobiotics therapy may have favorable effects on the amelioration of psychological disorders.

AbstractObjectiveTo investigate oxidative stress parameters [total oxidant status (TOS), total antioxidant status (TAS), oxidative stress index (OSI), paraoxonase (PON), arylesterase (ARE) and thiol-disulphide homeostasis] in patients who were diagnosed as having somatic symptom disorder in accordance with Diagnostic and Statistical Manual of Mental Disorders-5.MethodsThe study included 41 medication-free patients with somatic symptom disorder and 47 age, sex, and sociodemographic-matched healthy individuals. The patients were administered the Patient Health Questionnaire-15, Somatic Symptom Amplification Scale, Beck Depression Inventory, and Beck Anxiety Inventory. TOS, TAS, OSI, PON, ARE thiol, disulphide levels, and routine biochemical parameters were compared between the two groups.ResultsTOS, OSI, disulphide levels, disulphide/native thiol, and disulphide/total thiol ratios were found significantly higher in the patient group compared with the control group (p < 0.001). There was no significant difference in PON, ARE, and TAS parameters between the two groups (p > 0.05).ConclusionThis study showed that the level of oxidants increased and oxidative balance was impaired in somatic symptom disorder. Oxidative stress may play a role in the aetiopathogenesis of this disorder. This is the first study to report an association between oxidative stress and somatic symptom disorder.

<p><strong>BACKGROUND</strong><strong>: </strong>Oral lichen planus (OLP) is a chronic and inflammation mucosal disease. Reactive Oxygen Species (ROS) and oxidative stress damage might be the cause. Malondialdehyd (MDA), one parameter of lipid peroxidation is appropriate for DNA damage in OLP disease.</p><p><strong>OBJECTIVE</strong><strong>: </strong>To evaluate antioxidants and oxidative stress parameters in patients with oral lichen planus (OLP) diseases and healthy control group with compare on serum and salivary samples.</p><p><strong>MATERIAL </strong><strong>&amp;</strong><strong> METHODS: </strong>The research population included 22 patients with OLP which recently diagnosed and 22 healthy controls matched for periodontal status. Total antioxidant activity (TAA), Malondialdehyde (MDA) or lipid peroxidation product and uric acid (UA) were evaluated in both serum and saliva. The t-tests were used for differences between the two groups in normal distributed variables and also Spearman’s rho correlation coefficient for assessing association between serum and salivary fluids.</p><p><strong>RESULTS</strong><strong>: </strong>TAA levels in OLP patients showed significant result and lower than healthy control group (p=0.39). Also, results in the saliva MDA concentration, was significantly higher in OLP patients than controls. In correlation test, inverse and significant correlation was observed between the MDA and UA values(r=0. 682, P=0.0001) and a significant correlation was found between serum TAA and UA values.</p><p><strong>CONCLUSION</strong><strong>: </strong>This study showed that OLP groups have higher cellular lipid peroxidation in compare to healthy controls and low level of TAA than controls. Patients with OLP are believed to be more at risk of antioxidant-oxidative stress imbalance.</p>
