Thermal lesions were produced in 12 male Wistar rats, positioning a massive aluminum bar 10 mm in diameter (51 g), preheated to 99°C ± 2°C/10 min. on the back of each animal for 15 sec. After 7, 14, 21, and 28 days, animals were euthanized. The edema intensity was mild, with no bubble and formation of a thick and dry crust from the 3rd day. The percentage of tissue shrinkage at 28 days was 66.67 ± 1.66%. There was no sign of infection, bleeding, or secretion. Within 28 days reepithelialization was incomplete, with fibroblastic proliferation and moderate fibrosis and presence of modeled dense collagen fibers. It is concluded that the model established is applicable in obtaining deep second-degree thermal burns in order to evaluate the healing action of therapeutic agents of topical use.

This study aimed at evaluating the use of hydrogel isolectin in the treatment of second-degree burns. Twenty male rats were randomly divided into two groups (G1 = treatment with hydrogel containing 100 μg/mL Cramoll 1,4 and G2 = Control, hydrogel). After 7, 14, 21, 28, and 35 days, animals were euthanized. On the 7th day, G1 showed intense exudates, necrosis and edema. On the 14th day, G1 showed tissue reepithelialization and moderate autolysis. On the 21st day, G1 showed intense fibroblastic proliferation, presence of dense collagen, and moderate fibrosis. On the 28th day, G1 showed complete tissue epithelialization. On the 35th day, G1 showed modeled dense collagen. The significant wound contraction was initiated from day, 14 in the G1. There were no significant differences in biochemical and hematological parameters analyzed. These results extend the potential of therapeutic applications for Cramoll 1,4 in the treatment of thermal burns.

The cancerogen 1,2-dimethylhydrazine (DMH), widely used in the experimental animal model of carcinogenesis, affects various organs, but its effect on muscle fibers is unknown. To evaluate the effect of 15-week DMH treatment on the fiber size and myosin heavy chain (MyHC) isoforms, which substantially determine fiber types and their contractile characteristics, pure and hybrid fiber types were immunohistochemically determined according to the MyHC isoform expression in soleus, extensor digitorum longus, gastrocnemius medialis and lateralis muscles of DMH-treated and control male Wistar rats. Whereas the size of fibers was mostly unaffected, the MyHC isoform expression was partially affected in both gastrocnemius samples, but not in the soleus and extensor digitorum longus of DMH-treated rats. The lower proportions of hybrid fiber types and especially that of type 1/2x in most gastrocnemius samples of DMH-treated rats resulted in a shift towards a single MyHC isoform expression, but the extent and pattern of the MyHC isoform shift varied across the different gastrocnemius samples. Such variable response to DMH treatment across muscles indicates that each muscle possesses its own adaptive range. These findings are essential for an accurate evaluation of skeletal muscle characteristics in DMH animal model.

Ocimum gratissimum(OG) is known as a food spice and traditional herb, which has been recommended for the treatment of various diseases. To investigate the hepatoprotective effect of OG aqueous extract (OGAE), male Wistar rats challenged by carbon tetrachloride (CCl4) were used as the animal model of chronic hepatic injury. Significantly increased serum catalase and DPPH levels were detected in CCl4-administrated rats that were treated with OGAE or silymarin as compared to those rats that were treated with saline or CCl4. In contrast, significantly decreased stress proteins including HSP70 and iNOS were observed in livers of CCl4-administrated rats that were treated with OGAE or sylimarin as compared to those rats that were treated with saline or CCl4. Moreover, significant decreases of MMP-9/MMP-2 ratio, uPA, phosphorylated ERK (p-ERK) and NF-κB (p-P65) were detected in livers of CCl4-administrated rats that were treated with OGAE or sylimarin as compared to those rats that were treated with saline or CCl4. These findings imply that OGAE can efficiently inhibit CCl4-induced liver injuries in rats and may therefore be a potential food or herb for preventing liver injuries.

ABSTRACTBackground: Ramania leaf (Bouea macrophylla Griff) extract gel has secondary metabolites in the form of flavonoids, steroids, phenols and terpenoids which have a role as antioxidant. They will protect the body from excessive production of reactive oxygen species (ROS) by increasing endogenous antioxidants SOD, CAT and GPX, so that wound healing will not be inhibited and the process of collagen synthesis can run smoothly. Objective: To analyze the effect of ramania leaf extract gel that was applied topically with 5%, 10% and 15% concentration on collagen fibers density in incisional wound of male Wistar rats (Rattus norvegicus) on the 7th and the 14th day. Method: This research is a pure experimental study with a posttest-only control group design, using 24 rats which were divided into 4 groups: the treatment groups given ramania leaf extract gel of 5%, 10%, 15%, and the control group given placebo gel. The application of the extract gel was done once within 24 hours. The collagen level was measured with a spectrophotometer on the 7th and the 14th day. Results: Two-Way ANOVA test results on the 7th and the 14th day of each group showed a significant difference with p=0.000 (p<0.05). The Bonferroni Post-hoc Test showed a significant difference with p<0.05 between the placebo gel group and the groups of ramania leaf extract gel of 5%, 10%, 15% on the 7th and the 14th day. Conclusion: There is an effect of ramania leaf extract gel on collagen fibers density with the most effective concentration of 15%.Keywords: Antioxidant, , Collagen, Ramania Leaf Extract Gel

The glycaemic indices (GI) of food paste made from wheat, corn, yam, flours and garri in apparently healthy rats using glucose as standard control and water as normal control were determined through  Laboratory-experimental design. This was achieved based on the effect of these different selected carbohydrate diets on postprandial glycaemia of the animal model which resulted on variable glucose response. The proximate analysis of the processed diet from wheat, corn and yam showed higher fat and protein contents than that of gari diet. Also, yam diet showed the lowest carbohydrate. The fibre content was shown to be higher in gari with the lowest in yam containing diet.  Each of these selected carbohydrate diets contained 56.85% starch. A total of eighteen (18) adult male wistar rats divided into six groups which consisted of three rats each were used in the course of this study. Animals were starved  throughout the night for twelve (12) hours and their blood glucose level tested at time zero, before the test food containing 2 grammes of carbohydrate per kilogramme body weight were cautiously administered. After a time interval of 15, 30, 45, 60, 90 and 120 minutes, the blood glucose was determined. α-amylase digestibility of the yam, corn, and wheat diets were higher than that of garri diet. The glycaemic index of yam diet (73.8%) was the highest, followed by that of corn diet (70.4%), wheat diet (70.2%) and garri (61.5%). The glycaemic indices of all the rat diets were less than that of the glucose standard (100%). Comparatively, the glycaemic indices of yam, wheat and corn diets did not differ significantly (p>0.05), while that of garri was significantly (p<0.05) lower.

Abstract Context Parathyroid hormone (PTH) replacement is a promising approach in the management of hypoparathyroidism but long-acting analogues need to be developed. To date, animal models for testing PTH required parathyroidectomy by surgery. We have developed a nonsurgical rodent hypoparathyroid model and tested a delayed-clearance PTH molecule (DC-PTH). Objective The aim of this study was to use cinacalcet to suppress calcium levels in normal rats and to reverse these effects with the administration of PTH or PTH analogues Methods Male Wistar rats were gavaged with either 30 mg/kg cinacalcet-HCl (cinacalcet) or vehicle only. Animals were then dosed with either single or repeated subcutaneous doses of PTH 1-34 or a DC-PTH at 20 nmol/kg. Control animals received vehicle only. Serum samples were analyzed for ionized calcium (iCa), phosphate, PTH, and DC-PTH. A pharmacokinetic-pharmacodynamic (PK-PD) model was built for cinacalcet, PTH 1-34, and DC-PTH using Phoenix64. Results Cinacalcet reduced iCa levels between 2 and 24 hours, returning to baseline by 72 hours post dose with nadir at 8 hours (analysis of variance P &lt; .001), associated with a fall in rat PTH. For phosphate there was a variable biphasic response. Single-dose PTH abrogated the cinacalcet-induced fall in iCa for up to 2 hours. DC-PTH prevented the fall in iCa from 4 hours post dose and gave a prolonged response, with iCa levels quicker to return to baseline than controls. DC-PTH has a half-life of 11.5 hours, approximately 44 times longer than human PTH 1-34. The PK-PD models defined the reproducible effect of cinacalcet on iCa and that DC-PTH had prolonged biological activity. Conclusion The administration of cinacalcet provides a robust and reproducible nonsurgical animal model of hypoparathyroidism. DC-PTH holds promise for the treatment of hypoparathyroidism in the future.

Abstract Objective: the aim of this study was to test the hypothesis that synthesis of nitric oxide (NO) and activation of CB1 receptors have opposite effects in a behavioural animal model of panic and anxiety. Methods: to test the hypothesis male Wistar rats were exposed to the elevated T maze (ETM) model under the following treatments: L-Arginine (L-Arg) was administered before treatment withWIN55,212-2, a CB1 receptor agonist ; AM251, a CB1 antagonist, was administered before treatment with L-Arg. All treatments were by intraperitoneal route. Results: the CB1 receptor agonist, WIN55,212-2 (1 mg/kg), induced an anxiolytic-like effect which was prevented by pretreatment with an ineffective dose of L-Arg (1 mg/kg). Administration of AM251 (1 mg/kg), a CB1 antagonist before treatment with L-Arg (1 mg/kg) did not produce anxiogenic-like responses. Conclusion: altogether, this study suggests that the anxiolytic-like effect of cannabinoids may occur though modulation of NO signaling.

Objective : To evaluate the smooth muscle-collagen fibers ratio in the bladder wall of castrated male wistar rats. Material & Method : Sixteen wistar rats were equally assigned to castrated group (n=8) and sham group (n=8). All wistar rats were subjected to a one-week acclimatization period before undergoing treatment. After 60 days, cystectomy was performed in all rats in both groups and subsequent specimens of the bladder were taken to semi quantitative pathology analysis using haematoxylineosin (HE) staining under Olympus CX20 microscope by a uropathologist. Results were then grouped into parametric data and analyzed using independent T-test. Results: Smooth muscle fibers were significantly lower in the castrated group 0,615±0,13 (p=0,01). There was higher amount of collagen fibers in the castrated group 0,385±0,18 (p=0,04). Smooth muscle to collagen ratio in the castrated group was also significantly lower compared to the control group 1,597±0,25 (p=0,03). Conclusion : Testosterone deprivation significantly reduces the smooth muscleand smooth muscle to collagen fibers ratio in bladder wall of castrated rats.

PURPOSE: To compare tissue cicatrization in the tongue of rats using electrosurgery and the cold blade. METHODS: Ten adult male Wistar rats were divided into two groups of 5 animals each, according to the time of sacrifice (3 and 7 days). Each animal had two incisions, one made with a cold blade and the other with an electric blade, both of which were approximately 0.5 mm in length. Following sacrifice of the animals, the tongues were submitted to a histological study in order to classify the presence of angiogenesis, fibroblastic proliferation, epithelial proliferation and inflammatory cells as good, moderate and weak. RESULTS: The wound made with electric blade presented a delay in the healing process and a greater inflammatory response as compared with the cold blade, despite the fact that there was no statistically significant difference (p<0.05). CONCLUSION: There were no significant statistical differences between electrosurgery and the cold blade in relation to angiogenesis, fibroblastic proliferation, epithelial proliferation or the presence of inflammatory cells at any time of evaluation.
