Fungi excrete a variety of extracellular enzymes capable of breaking down complex nutrient sources into products that can be directly metabolized by the fungi. An extracellular proteinase and lipase produced by the sap-staining fungus, Ophiostoma piceae, have been purified and the specificities of these enzymes for a variety of substrates have been determined. In this work, we show that these enzymes were able to hydrolyze natural substrates isolated directly from wood. Polyclonal antibodies were raised against proteinase K and were used to immunolocalize the major extracellular proteinase of the sap-staining fungus Ophiostoma piceae (Münch) H. and P. Sydow. Immunodot blotting showed that the IgG antibodies recognized both enzymes but reacted more strongly with proteinase K than with the O. piceae proteinase. Immunogold labelling and transmission electron microscopy revealed that the O. piceae proteinase was localized in the cell walls of O. piceae grown either in liquid media or wood. The development of ascoma, ascus, and ascospore in Ophiostoma piceae was studied ultrastructurally and compared with that of other Ascomycetes. Ascospore delimitation commenced with the formation of double delimiting membranes in the ascus. The ascospore wall, consisting of the primary and secondary walls, was deposited between these membranes. The elongate ascospores of O. piceae differed from other species having similarly shaped ascospores, with respect to the shape and arrangement of asci in the ascoma, the number of wall layers of the ascospore, and formation of the secondary wall. Asci in O. piceae are spindle shaped, arranged along the periphery of the ascomatal wall. The ascospores have three layered walls, whereas some other species in Ceratocystis s.l. also with elongated ascospores probably have only two wall layers. There are differences in the extracellular enzymes produced from species of wood-rotting fungi and their activity due to variation in nutritional conditions such as carbon sources and nitrogen sources, as well as environmental conditions such as incubation temperature and pH. In this study, to determine the methods to promote the secretion of especially cellulase with high activity among the extracellular enzymes of wood-damaging fungi, the optimal nutrient sources and incubation conditions for the production of cellulase with high activity were investigated using response surface methodology based on a broth culture from Acanthophysium sp. KMF001, a novel strain of wood-damaging fungus. The nutrient sources that were optimal for the cellulase production with high activity from Acanthophysium sp. KMF001 were cellulose as a carbon source and tryptone:yeast extract (7:3) as a nitrogen source. The optimal incubation conditions were a temperature of 30 °C and a pH of 6. The optimal concentrations of carbon and nitrogen sources were cellulose at 31.1 g∙L-1 and tryptone:yeast extract (7:3) at 15 g∙L-1, with pH of 5.9.

Gastrointestinal stromal tumors (GIST) are the most common mesenchymal tumors of the digestive tract (1%). These tumors express the CD 117 in 95% of cases. The stomach is the preferential localization (70%). Diagnosis is difficult and sometimes late. Progress of imaging has greatly improved the management and the prognosis. Computed tomography (CT) is the gold standard for diagnosis, staging, and treatment follow-up. The increasing recognition of GIST’s histopathology and the prolonged survival revealed some suggestive imaging aspects. Key words: gastro-intestinal stromal tumors; computed tomography; diagnosis

This article aims at describing the obseroance of wasiat wajibah (compulsory bequeathment) in the Islamic court of Banten, analyzing the provision of the substitute heir and adopted children in the Compilation of  Islamic Law (KHI). In addition, the ideal laws to manage the innheritance rules in Indonesia. This article is library research by using doctrinal approach and using case study and survey methods. The data was, then, analyzed by using analytical descriptive and analytical correlative methods. The result showed that the observance of wasiat wajibah in the Islamic court of  Banten employed by judges is by using the Mawali Hazairin’s Doctrine. Five phosphorus rates (0, 5, 10, 20 and 30 kg P/ha) were tested with four rice genotypes in Boro (BRRI dhan36, BRRI dhan45, EH1 and EH2) and T. Aman (BRRI dhan30, BRRI dhan49, EH1 and EH2) season. Phosphorus rates did not influence grain yield irrespective of varieties in T. Aman season while in Boro season P response was observed among the P rates. Application of P @ 10 kg/ha significantly increased the grain yield. But when P was applied @ 20 and 30 kg P/ha, the grain yield difference was not significant. The optimum and economic rate of P for T. Aman was 20 kg P/ha but in Boro rice the optimum and economic doses of P were 22 and 30 kg/ha, respectively. Hybrid entries (EH1 and EH2) used P more efficiently than inbred varieties. A negative P balance was observed up to 10 kg P/ha.
