Comparison of three microquantity techniques for measuring total lipids in fish

To measure lipids in juvenile and adult fishes, we refined three microquantity approaches (microgravimetric, microcolorimetric sulfophosphovanillan (SPV), and Iatroscan thin layer chromatography – flame ionization detection (TLC–FID)) that were originally developed to measure lipids in small aquatic invertebrates. We also evaluated their precision and comparability by quantifying the total lipid content of age-1+ walleye ( Sander vitreus ), yellow perch ( Perca flavescens ), and lake whitefish ( Coregonus clupeaformis ) collected in Lake Erie (US–Canada), Lake Michigan (USA), and Muskegon Lake (Michigan, USA). Our findings demonstrate that (i) microquantity approaches provide estimates of total lipids in juvenile and adult fishes similar to those of more traditional macroquantity approaches, (ii) the microcolorimetric SPV and microgravimetric approaches produce near identical estimates of total lipid content, and (iii) the Iatroscan TLC–FID approach underestimates total lipids relative to the other approaches for individuals with high lipid levels. Ultimately, our research makes available additional techniques for measuring total lipid content of fishes that are less expensive than traditional techniques, owing to a reduced need for large quantities of samples and solvents.

Five soft coral samples belonging to the same specimen Sinularia brassica were collected in the coastal area of Ha Long Quang Ninh, Nhat Le beach, Quang Binh, Hon Vang - Thanh Hoa and Nha Trang areas (2014–2016), these samples were examined and analyzed by total lipid content, content and composition of lipid and fatty acids. Highest total lipid content was found in the sample SHM 09 collected in Nha Trang bay (1.11%), and the lowest total lipid content was in HL-SHM 07 collected in Ha Long, Quang Ninh (0.68%).The lipid composition of samples contained basic classes such as polar lipids, sterols, free fatty acids, triacylglycerols, monoankyldiacylglycerols, hydrocarbons and waxes. In studied soft corals, there are the appearance of all fatty acids from C12 to C24. There are differences in composition and content of lipids and fatty acids in samples collected in different locations: Samples collected at Nha Trang bay contain more polyunsaturated fatty acids than the other samples collected in Ha Long, Thanh Hoa, Quang Binh. Another two samples, collected in Hon Vang (Thanh Hoa) have similar composition and content of lipid classes, the composition and content of fatty acids in total lipids. The obtained results also show the significant impact of habitat and time of acquisition on the studied subject.

The present study investigated the lipid content and FA profiles of total lipids, phospholipids (PL) and triacylglycerols (TAG) of Mastacembelus mastacembelus from the Atatürk Dam Lake (Turkey). The results showed that the total lipid content of the dorsal muscle varied seasonally from 0.50% to 3.59%. Wide ranges of saturated fatty acids (SFAs) (36.21 to 50.52%) and polyunsaturated fatty acids (PUFAs) (25.22 to 42.02%) were found in the PL fraction. However, higher monounsaturated fatty acids (MUFAs) (37.63 to 45.07%) and SFA (34.71 to 38.82%) were found in TAG (triacylglycerol). The ratios of ω-3 to ω-6 PUFA ranged from 0.65 to 1.32 and 1.07 to 3.48 in PL (phospholipid) and TAG fraction, respectively. The results also showed that the major components were palmitic acid (C16:0), stearic acid (C18:0), oleic acid (C18:1ω-9), palmitoleic acid (C16:1ω-7), docosahexaenoic acid (DHA, C22:6ω-3), arachidonic acid (AA, C20:4ω-6) and docosapentaenoic acid (C22:5ω-3) in total lipid, C16:0, C18:1ω-9, AA and DHA in PL, C16:0, C16:1ω-7, C18:1ω-9, linoleic acid (LA, C18:2ω-6), and DHA in TAG extracted from the muscle of M. mastacembelus in all seasons.

In this study, buffalo spermatozoa were in-vitro capacitated and acrosome reacted using modified Tyrode’s bicarbonate-buffered medium (sp-TALP media). Lipids were extracted from freshly ejaculated, in-vitro capacitated and acrosome reacted buffalo spermatozoa. The total lipid content of freshly ejaculated spermatozoa (171.5±2.90 μg/108 cells), capacitated spermatozoa (128.2±3.29 μg/108 cells) and acrosome reacted spermatozoa (115.7±2.04 μg/108 cells) were found significantly (P<0.05) different. During the process of capacitation and acrosome reaction (AR) there was substantial loss of total lipids. The total Lipid content of freshly ejaculated, capacitated and acrosome reacted spermatozoa were fractionated into three major classes namely, Phospholipids, Neutral lipids and Glycolipids. Among the classes of lipids; there was no change in concentration of glycolipids. Phospholipids in fresh, capacitated and acrosome reacted spermatozoa were 84.27±1.96, 58.40±2.40 and 50.51±1.27 μg/108 cells respectively. In case of Neutral lipids, the concentration were 33.63±2.62, 22.55±1.86 and 11.61±0.87 μg/108 cells in fresh, capacitated and acrosome reacted spermatozoa respectively. The results generated by this study will be one step further to understand the lipid changes in buffalo spermatozoa during capacitation, acrosome reaction and fertilization process.

Objective: Lipids are secreted into milk as bilayer-coated structures: milk lipid globules (MLGs). Adipophilin (ADRP) and perilipin 3 (TIP47) are associated with MLGs in human breast milk; however, the role of these proteins in milk lipid secretion is not fully understood. The aim of the study was to investigate levels of ADRP, TIP47 and total lipid content in human breast milk, their mutual correlations and dynamics during lactation. Research Methods &amp; Procedures: Milk samples from 22 healthy lactating women (Caucasian, Central European) were collected at five time points during lactation (1&ndash;3, 12&ndash;14, 29&ndash;30, 88&ndash;90 and 178&ndash;180 days postpartum). Mass spectrometry-based method was used for quantification of ADRP and TIP47 in the samples. The gravimetric method was used to determine milk total lipid content. Results: We observed distinctive trends in ADRP, TIP47 levels and lipid content in human breast milk during the first 6 months of lactation. We also found a significant association between lipid content and ADRP, lipid content and TIP47, and ADRP and TIP47 concentrations in breast milk at all sampling points. Moreover, we derived an equation for estimating the mean lipid content of milk based on ADRP concentration in human breast milk. Conclusions: A mass spectrometry-based method was developed for quantifying ADRP and TIP47 in human breast milk. Strong mutual correlations were found between ADRP, TIP47 and total lipid content in human breast milk.
