In vitro micrografting of Sequoia sempervirens was used as a rejuvenation method: apices were collected from in vitro cultured shoots issued from a 500-year-old tree; the rootstocks originated from a 1-year-old tree. Only a few grafted apices survived. Once producing leafy shoots, they exhibited some juvenile morphological and physiological characteristics. Some scions were isolated and rooted in a new medium. Although the restoration of the orthotropy is very difficult, a few of the rooted plantlets were orthotropic and this characteristic was maintained after acclimatization in greenhouses. Therefore, the in vitro micrografting of Sequoia sempervirens can be considered as a rejuvenation method that must be improved. In vitro micrografting of apices of two mature genotypes of Hevea brasiliensis (Müll. Arg.), IRCA 18 and PB 235, on 3-wk-old seedlings grown in vitro permitted successfull in vitro micro-cutting of these two genotypes. Microcutting of micrografted material was impossible from nodal explants or shoot tips of scions developed in vitro on rootstocks. Such explants were incapable of caulogenesis activity after their isolation. This problem was resolved using mixed explants, each consisting of a part of the rootstock in contact with the culture medium, and of the clonal scion from which axillary shoots are developed regularly along the subcultures. Budding and shoot elongation abilities along the subcultures have been compared between mixed explants from micrografts, nodal explants from juvenile material and nodal explants from non-micrografted mature genotypes. The results show a very positive effect of micrografting on the in vitro caulogenesis ability in both genotypes. Moreover, shoots produced by mixed explants from micrografts exhibit the same rooting ability as shoots produced by explants collected on juvenile material. <p>ABSTRAK</p><p>Studi in vitro tentang karakteristik fungi isolat Pleurotus spp. telahdilaksanakan di Bogor dari bulan Juli sampai Agustus 2004. Penelitianmenggunakan rancangan faktorial dalam rancangan acak lengkap danbertujuan untuk mempelajari pengaruh media, temperatur inkubasi dan pHmedia terhadap 6 isolat Pleurotus sp. Karakter lain yang juga dipelajariadalah kemampuan untuk mengoksidasi asam tanat dan asam galat dalammedia agar. Hasil penelitian menunjukkan bahwa Pleurotus isolat sp.6dan sp.8 tumbuh baik pada media MPA, isolat Pleurotus sp.1, 3 dan 4pada media MEA dan isolate sp.2 pada media PDA. Kecuali isolat sp.8,isolat lainnya tidak dapat tumbuh pada temperatur 10 dan 35 o C. Pertum-buhan isolat sp.8 terbaik dibandingkan isolat lainnya pada semuatemperatur. Diameter koloni isolat lainnya hanya mencapai 0,2 – 2,33 cm.Pertumbuhan isolat sp.8 juga terbaik pada semua pH media diikuti isolatsp.6 kemudian isolat sp.4. Semua isolat menunjukkan reaksi oksidasipositif pada agar asam tanat dan asam galat yang ditunjukkan oleh warnacoklat pada media yang melingkari koloni.</p><p>Kata kunci : Pleurotus spp, media, temperature, pH, oksidasi, asam tanat,asam galat</p><p>ABSTRACT</p><p>Physiological Characteristics of Pleurotus spp. IsolatesPhysiological characteristics of some Pleurotus sp. isolates werestudied in vitro, from July until August 2004 in Bogor. Experiments tostudy the effect of kind of media, temperature of incubation room, and pHof medium on six isolates of Pleurotus sp. were arranged in factorialrandomized complete design and replicated three times with colony in apetri dish as experimental units. Another physiological character studiedwas the ability to oxidize tannic and gallic acids in agar medium. Resultsshowed that isolates Pleurotus sp.6 and -8 grew better in MPA medium,Pleurotus sp.1, -3, and -4 in MEA, and Pleurotus sp.2 in PDA. ExceptPleurotus sp.8, other isolates could not grow in incubation roomtemperature of 10 and 35 o  C. The growth of Pleurotus sp.8 was the bestamong the isolates in all temperature levels. Other isolates grew poorly in20 and 29o C with diameter range was 0.2 – 2.33 cm. The growth ofPleurotus sp.8 was also the best in all pH medium levels, followed byPleurotus sp.6, and then Pleurotus sp.4. The in vitro glucose consumption and lactic acid production by erythrocytes from 20 cattle of a Beef Synthetic (SY) breed group and 25 cattle of a double-muscled (DM) breed group were determined. There were three age groups and two sexes within each breed group. Animals within the DM breed group were categorized as either phenotypically normal- to moderate-muscled (DM carriers) or extreme-muscled (extreme DM) based on the phenotypic expression of the double-muscling trait and on their breeding history. Both DM phenotypes had higher (P < 0.01) erythrocyte glucose consumption and lactic acid production than normal-muscled, noncarrier cattle of the SY breed group. Extreme DM cattle were not different (P > 0.05) in either their erythrocyte glucose consumption or lactic acid production compared with DM carriers. No difference (P > 0.05) due to breed or phenotype was observed in the molar ratio of lactic acid produced to glucose consumed by erythrocytes. These results suggest that carriers of the Double Muscled Syndrome, regardless of phenotypic expression of the double-muscling trait, have a higher rate of erythrocyte glycolysis than normal cattle. Purpose. Еxperimental substantiation of the effectiveness of biocompatible biodegradable hydrogels based on hyaluronic acid and chitosan succinate as a carrier of ranibizumab in antiglaucoma operations. Material and methods. Hydrogel drainage (HD) was obtained immediately before surgery. A solution of ranibizumab (0.23 ml) was mixed with a solution of hyaluronic acid dialdehyde (0.5 ml), then a solution of chitosan succinate (0.5 ml) was added. Experimental studies were performed in 12 (12 eyes) healthy rabbits. The first group consisted of 6 eyes – 0.187 ml of ranibizumab per 1 ml of gel. In the control group, HD was used intraoperatively without the addition of ranibizumab (6 eyes). Morphological studies were performed on 7th, 21st, and 42nd days. Results. In experimental studies in vitro and in vivo, it was proved that ranibizumab, administered as a part of 0.1 ml of hydrogel drainage in the antiglaucoma surgery area is released within 3 weeks and suppresses vascularization, scarring of the operating area, and preserves the intrascleral cavity. The optimal concentration of ranibizumab was selected-0.02 ml in 0.1 ml of gel. Conclusion. The safety and effectiveness of the use of hydrogel drainage with ranibizumab based on hyaluronic acid dialdehyde and chitosan succinate in anti-glaucoma operations has been proven. Nodal explants of 12 accessions from four species of yam (Dioscorea spp.) were cultured for six weeks on MS to evaluate the influence of IAA, Kn, NAA and BAP on the production of leaves and microtubers. Four Dioscorea polystachya Turcz., three each of D. bulbifera L. and D. sansibarensis Pax. and two D. japonica Thunb. accessions were used. Five and 10 mg/l of Kn along with IAA and sucrose, and 0.2 and 0.5 mg/l of NAA, sucrose and with or without BAP were used in four treatments. The accessions Yam 23 and Yam 25 of D. sansibarensis failed to initiate any leaf under four treatments. The remaining accessions produced 0.11 to 1.76 leaves per explant. The medium containing IAA with higher concentration of Kn (10 mg/l) and 3% sucrose was found to be best for in vitro production of leaf (0.71/explant) and the most productive species was D. japonica (1.36), followed by D. polystachya (1.19/explant). At the same culture period, Yam 16 of D. bulbifera failed to initiate any microtuber at IAA with Kn, and NAA with or without BAP. The remaining accessions produced 0.09 to 1.15 microtubers per explant. Lower concentration of Kn (5 mg/l) with IAA and sucrose was favourable for producing microtubers (0.61/explant on an average), the best species being D. sansibarensis (1.27) followed by D. japonica (0.59/ explant). Shoot tip explants from in vitro germinated seedlings of Solanum capsicoides All. inoculated on MS containing 2 mg/l BA produced maximum shoot induction response (26 shoots per explant). Rooting of the microshoots (19.4 roots per explant) was obtained better in half strength of MS supplemented with NAA (0.5 mg/l). A new endomycetaceous fungus, Botryoascus cladosporoides, is described. This species differs from the previously described Botryoascus synnaedendrus in conidial structures and physiological characteristics and occurs in a very different habitat. Experiments were carried out to transform laboratory mice and domestic chickens by use of sperm incubated with bacterial plasmid DNA. Following demonstration of "uptake" of such DNA by sperm of both species, attempts were made to replicate a previously published procedure (Lavitrano et al. 1989, Cell 57: 717–723) for producing transgenic mice through in vitro fertilization (IVF). Also, female mice and hens were inseminated (AI) with sperm which had been incubated in a DNA solution. Such incubation did not influence the fertility or hatchability of the hens' eggs. However, no transformed progeny were detected among 45 mice produced by IVF or among 69 mice and 470 chickens produced by AI. Key words: Sperm-mediated DNA transfer, mice, chickens

Basal steroid production in vitro by comparable follicles from cyclic and PMSG/hCG-treated gilts was similar. Follicular responses to LH in vitro varied, however, confirming earlier evidence for functional differences in the pattern of follicular development in immature gilts in response to exogenous gonadotropins compared with naturally cyclic gilts.
