Electron microscopy of susceptible and resistant near-isogenic (sr6/Sr6) lines of wheat infected by Puccinia graminis tritici. I. The host–pathogen interface in the compatible (sr6/P6) interaction

Membrane associations in the interfacial zone between host cytoplasm and haustoria of wheat stem rust are reported. The invaginated host plasmalemma (IPL) adhered tightly to the haustorial neck wall above an indistinct zone at the base of the neck. There was extensive proliferation of host endoplasmic reticulum (ER) near haustoria during the formative growth stages of the haustoria. Most of the proliferation occurred along the neck from the neck ring region to the area at the base of the haustorial body where the extrahaustorial sheath became evident. Closely spaced arrays of ER occurred along the neck and radiated into the host cytoplasm. The ER was closely associated with the IPL in the vicinity of the neck wall, and at points of contact or near contact, the ER was ribosome free and stained more densely. There was luminal continuity between ER cisternae and the extrahaustorial sheath matrix. In the host cytoplasm adjacent to haustoria, ER-derived membranous complexes were found. These complexes consisted of a lattice arrangement of large tubules and small tubules, the latter containing a thread of denser staining material at their centers. The large tubules were bounded by lightly stained membranes, whereas the membranes of the small tubules stained more densely. These complexes were interconnected by rough ER to form networks in host cytoplasm. The complexes formed part of the interface between host and pathogen through connections with the invaginated host plasmalemma.

Seven races have been described in the Ug99 race group of Puccinia graminis f. sp. tritici (2). Ug99-related races previously recorded in South Africa are TTKSF, TTKSP, and PTKST (4). In December 2010, severe stem rust infection of the winter wheat cv. Matlabas was observed for the first time in South Africa. Race analysis using the 20 North American (NA) stem rust differential lines and letter code system classified the race as TTKSF. In comparative infection studies in a greenhouse, cv. Matlabas seedlings were susceptible (infection type [IT] 4) to isolate UVPgt61/1 (TTKSF+) collected from Afrikaskop in the eastern Free State, whereas the cultivar was resistant (IT 1 to 2) to stem rust isolates 2013 (TTKSF), UVPgt55 (TTKSF), UVPgt59 (TTKSP), and UVPgt60 (PTKST). Isolate 2013 represents the original collection of race TTKSF in South Africa (1). In addition to the NA differentials, no variation in the IT range of seedlings of lines with Sr7a, 8b, 12, 13, 14, 16, 18, 19, 22, 25, 26, 27, 28, 29, 32, 33, 34, 35, 39, 41, 42, 43, 44, Em, R, Tt2, and Satu was observed between UVPgt61/1 and UVPgt55. With the exception of cv. Matlabas, ITs of 106 South African cultivars likewise did not differentiate UVPgt61/1 and UVPgt55. Seedling IT studies were conducted at least twice. Microsatellite analysis (4) showed that all single pustule isolates established from the original Matlabas isolate formed part of the Ug99 group. When characterized with selected single nucleotide polymorphisms (SNPs), all single pustule isolates shared an identical genotype that differed from UVPgt55 (TTKSF), a foreign introduction into South Africa (1,3). SNP genotype analysis suggests that UVPgt61/1 is genetically dissimilar to UVPgt55, as is Zim1009, another TTKSF+ isolate that was collected from Birchenough in Zimbabwe. Studies are underway to determine the identity of the defeated Sr gene in Matlabas and the cultivar has been added to the South African stem rust differential set. TTKSF+ is the eighth race detected in the Ug99 group. Since no other cultivars or advanced lines were found to carry the Matlabas gene, it is unlikely that race TTKSF+ will threaten wheat production in South Africa. The fine structure of uredospores and germ tubes of Puccinia graminis f. sp. trilici is described from electron micrographs of cells fixed in KMnO4. In terms of their principal cytoplasmic structures (nuclei, mitochondria, endoplasmic reticulum, and protoplasmic and vacuolar membranes) these cells bear a general resemblance to other fungi and higher plants but differences between resting cells and germ tubes were noted in the size of oil bodies and the structure and extent of the endoplasmic reticulum.

The interaction between avirulent wheat stem rust and wheat mesophyll cells containing the temperature-sensitive Sr6 gene for stem rust resistance was studied by electron microscopy. Mesophyll cells that were invaded at 26 °C (conditioning compatibility) did not develop any signs of incompatibility after they were transferred to 19 °C, at which temperature incompatibility is normally expressed. In host tissue that appeared to be invaded after the change from 26 to 19 °C, the early ultrastructural symptoms of incompatibility were a more electron-dense and often perforated invaginated host plasmalemma, disruptions of the noninvaginated host plasmalemma, vacuolation of the cytoplasm, and accumulations of electron-dense material along the membranes of the vacuoles. At later stages in the development of incompatible interactions, the electron-dense accumulations along the vacuole membranes increased in size and occurred along chloroplast and mitochondrial membranes. Eventually, the entire protoplasts were electron dense and collapsed. In haustoria and haustorial mother cells, incompatibility was usually expressed by a uniform increase in electron density of the cytoplasm. In the Sr6/P6 interaction at 19 °C, host cell necrosis was not always accompanied by fungal necrosis or vice versa. In Sr5/P5 interactions, which were examined for comparison, the intracellular symptoms of incompatibility were similar to those of the Sr6/P6 interactions.

Electron microscopy shows that the nuclei in immature uredospores of Puccinia graminis tritici possess prominent nucleoli with a mean diameter of 1.7 ± 0.4 μ. The nucleoli in mature uredospores are much smaller (0.5 μ). This reduction in size of the nucleoli may indicate that ribosomal RNA (ribonucleic acid) formation is repressed as uredospores mature.

Early in spermiogenesis the manchette is rapidly assembled in a distal direction from the nuclear-ring-densities. The association of vesicles of smooth endoplasmic reticulum (SER) and the manchette microtubules (MTS) has been reported. In the mouse, osmophilic densities at the distal ends of the manchette are the organizing centers (MTOCS), and are associated with the SER. Rapid MT assembly and the lack of rough ER suggests that there is an existing pool of MT protein. Colcemid potentiates the reaction of vinblastine with tubulin and was used in this investigation to detect this protein.

There have been many reports which establish a relationship between the pineal and sexual structures, including gonadal hypertrophy after pinealectomy, and gonadal atrophy after injection of pineal homogenates or of melatonin. In order to further delineate this relationship the pineals from 5 groups of female rats were studied by electron microscopy:ControlsPregnant ratsAfter 4 weekly injections of 0.1 mg. estradiol benzoate.After 8 daily injections of 150 mcgm. melatonin (pineal hormone).After 8 daily injections of 3 mg. serotonin (melatonin precursor).No ultrastructural differences were evident between the control, and the pregnancy and melatonin groups. However, the estradiol injected animals exhibited a marked increase in the amount and size of rough endoplasmic reticulum within the pineal cells.

In order to study the retention of copper in cirrhotic liver, rats were made cirrhotic by carbon tetrachloride inhalation twice weekly for three months and fed 0.2% copper acetate ad libidum in drinking water for one month. The liver tissue was fixed in osmium, sectioned approximately 2000 Å thick, and stained with lead citrate. The section was examined in a scanning electron microscope (JEOLCO JSM-2) in the transmission electron mode.Figure 1 shows a typical area that includes a red blood cell in a sinusoid, a disse, and a portion of the cytoplasm of a hepatocyte which contains several mitochondria, peribiliary dense bodies, glycogen granules, and endoplasmic reticulum.

Thirty-three mature male Holtzman rats were randomly placed in 3 treatment groups: Controls (C); Ethanolics (E); and Wine drinkers (W). The animals were fed synthetic diets (Lieber type) with ethanol or wine substituted isocalorically for carbohydrates in the diet of E and W groups, respectively. W received a volume of wine which provided the same gram quantity of alcohol consumed by E. The animals were sacrificed by decapitation after 6 weeks and the livers processed for quantitative triglycerides (T3), proteins, malic enzyme activity (MEA), light microscopy (LM) and electron microscopy (EM). Morphometric analysis of randomly selected LM and EM micrographs was performed to determine organellar changes in centrilobular (CV) and periportal (PV) regions of the liver. This analysis (Table 1) showed that hepatocytes from E were larger than those in C and W groups. Smooth endoplasmic reticulum decreased in E and increased in W compared to C values.

Potassium permanganate has been successfully employed to study membranous structures such as endoplasmic reticulum, Golgi, plastids, plasma membrane and myelin sheath. Since KMnO4 is a strong oxidizing agent, deposition of manganese or its oxides account for some of the observed contrast in the lipoprotein membranes, but a good deal of it is due to the removal of background proteins either by dehydration agents or by volatalization under the electron beam. Tissues fixed with KMnO4 exhibit somewhat granular structure because of the deposition of large clusters of stain molecules. The gross arrangement of membranes can also be modified. Since the aim of a good fixation technique is to preserve satisfactorily the cell as a whole and not the best preservation of only a small part of it, a combination of a mixture of glutaraldehyde and acrolein to obtain general preservation and KMnO4 to enhance contrast was employed to fix plant embryos, green algae and fungi.
