Effects of dietary supplemental lysophospholipids and vitamin C on performance, antioxidant enzymes, lipid peroxidation, thyroid hormones and serum metabolites of broiler chickens reared under thermoneutral and high ambient temperature

The aim of this study was to evaluate the effects of high ambient temperature (HT) and orotic acid supplementation on the plasma and muscle metabolomic profiles in broiler chickens. Thirty-two 14-day-old broiler chickens were divided into four treatment groups that were fed diets with or without 0.7% orotic acid under thermoneutral (25 ± 1 °C) or cyclic HT (35 ± 1 °C for 8 h/day) conditions for 2 weeks. The chickens exposed to HT had higher plasma malondialdehyde concentrations, suggesting an increase in lipid peroxidation, which is alleviated by orotic acid supplementation. The HT environment also affected the serine, glutamine, and tyrosine plasma concentrations, while orotic acid supplementation affected the aspartic acid, glutamic acid, and tyrosine plasma concentrations. Untargeted gas chromatography–triple quadrupole mass spectrometry (GC-MS/MS)-based metabolomics analysis identified that the HT affected the plasma levels of metabolites involved in purine metabolism, ammonia recycling, pyrimidine metabolism, homocysteine degradation, glutamate metabolism, urea cycle, β-alanine metabolism, glycine and serine metabolism, and aspartate metabolism, while orotic acid supplementation affected metabolites involved in pyrimidine metabolism, β-alanine metabolism, the malate–aspartate shuttle, and aspartate metabolism. Our results suggest that cyclic HT affects various metabolic processes in broiler chickens, and that orotic acid supplementation ameliorates HT-induced increases in lipid peroxidation.

Cisplatin-induced nephrotoxicity is associated with an increase in lipid peroxidation and oxygen free radicals in rat kidneys. In this study, the effects of desferrioxamine were compared to vitamin C and E on cisplatin-induced lipid peroxidation and antioxidant enzyme activities in rat kidneys. Rats were divided into five groups, with 15 Wistar rats in each group. In the control group, rats received 1 mL/100 g isotonic saline solution intraperitoneally (i.p.). In Group II, 10 mg/kg cisplatin i.p. was injected to rats. Thirty minutes before the same dosage of cisplatin administration, 100 mg/kg i.p. vitamin C or E was given to rats in groups III and IV, respectively. Rats in Group V received 250 mg/kg desferrioxamine i.p., before the same dose of cisplatin administration. All rats were killed by cervical dislocation after 72 hours. The kidneys were immediately removed and washed in cold saline. Spectrophotometric method was used for all analyses. While catalase, glutathione reductase (GR), and super oxide dismutase (SOD) levels were found to be significantly decreased (P B < 0.001), malondialdehyde (MDA) (P < 0.05) and hydrogen peroxide (H2O2) (P < 0.001) levels were significantly increased in the cisplatin group when compared to the controls. MDA levels were decreased by desferrioxamine (P < 0.005) as well as vitamin C and E (P < 0.05 and P < 0.001, respectively). These three compounds induced a significant increase in SOD levels (P B < 0.05), but only in the vitamin C group, were SOD levels not significantly different than the levels of the controls (P > 0.05). In the desferrioxamine (P < 0.05), vitamin C and E groups (P < 0.001 for both), the cisplatin elevated H2O2 levels were decreased. None of these drugs had any effect on GR and catalase levels (P > 0.05). Desferrioxamine is useful to prevent cisplatin-induced lipid peroxidation, however, vitamin C and E are more effective on antioxidant enzymes than desferrioxamine.

A 30-day experiment involving 720 White Leghorn layer hens was carried out to evaluate the effects of vitamins C and E on leukocyte and heterophil/lymphocytes ratios of the layer hens during the natural summer conditions. The birds were divided into 4 groups, one control with basal diet and three experimental with diet supplemented with vitamin C, vitamin E, or combination of both vitamins. Each group was divided into 4 subgroups. Exposure of layer hens to 31 ± 3 °C and 33 ± 0 °C ambient temperature and 84.6% and 81.5% relative humidity during the experiment caused an increase in the value of temperature humidity index of 15.5 above the threshold value of 70 established for this species. Total white blood cells, lymphocytes, and eosinophil levels were significantly (p < 0.001) higher in all treated groups compared to control. Similarly, heterophil/lymphocyte ratios of 0.51, 0.52, and 0.50 for groups with diet supplemented with vitamin C, vitamin E and vitamins C + E, respectively, were significantly (p < 0.001) lower compared to 0.59 recorded in the control group. However, heterophil, basophil and monocyte values were significantly (p < 0.001) higher in the control group compared to all treated groups. The results of the present study showed that dietary supplementation of vitamins C and E, or their combination, reduces the harmful effect of high ambient temperature and humidity on the leukocyte indicators of exposed layer hens, thereby alleviating the stress imposed on their thermoregulatory mechanism and physiological status.
