Phagocytic activity of PMN's in five healthy and five burned patients were measured in vitro. Addition of 1 μg per millilitre of amphotericin B to the assay produced a marked inhibitory effect of the phagocytic activity of PMN against C. albicans.

The inhibitory effect of human immunodeficiency virus (HIV) proteinase inhibitors amprenavir and saquinavir and antifungal agents terbinafine, ketoconazole, amphotericin B and ciclopiroxolamine on aspartyl proteinases (Saps) secreted by Candida albicans was tested in an in vitro spectophotometric assay. As expected, both HIV proteinase inhibitors showed a significant inhibitory effect on Sap activity, which was comparable to that of the classical aspartyl proteinase inhibitor pepstatin A (P < 0.001). Antifungal drugs such as ketoconazole, terbinafine and amphotericin B had no, or only minor, inhibitory effects on proteolytic activity. In contrast, a significant reduction in Sap activity could be demonstrated during treatment with the antifungal agent ciclopiroxolamine (P < 0.001). These results point to a multiple effect of this antimycotic agent and might explain the reduced adherence of C. albicans to human epithelial cells at subinhibitory doses.

ABSTRACT The in vitro activity of a new water-soluble polyene, SPA-S-843, was evaluated against 116 strains of Candida,Cryptococcus, and Saccharomyces spp. and compared with that of amphotericin B. SPA-S-843 demonstrated better inhibitory activity against all of the yeasts examined and better fungicidal activity against Candida albicans, Candida glabrata, Candida krusei, and Candida tropicalis than did amphotericin B.

ABSTRACT Several salivary proteins exhibit fungicidal activity against the opportunistic oral pathogen Candida albicans when they are tested as pure proteins in vitro. However, salivary secretions that are examined by the same assays either lack or exhibit very low candidacidal activity. Since ionic strength is known to have an inhibitory effect on the fungicidal activities of some proteins, parotid secretion was subjected to dialysis with membranes having molecular weight cutoffs (MWCOs) of 500, 1,000, 10,000, and 25,000. Dialysis with membranes with MWCOs of ≥1,000 promoted fungicidal activity of parotid secretion, and this activity was dose dependent. The addition of sodium chloride to dialyzed, fungicidal parotid secretion abolished this activity, indicating that the fungicidal component was salt sensitive. Similar results were obtained with submandibular and sublingual secretions. Polyacrylamide gel electrophoresis under native and denaturing conditions was used to analyze the composition of the dialysate. Unexpectedly, proteins with MWs much lower than the nominal MWCOs of the membranes were not lost during dialysis. Among the retained proteins, the two fractions with MWs of approximately 17,000 and 4,000 exhibited fungicidal activity. These results are consistent with the presence of lysozyme and histatins, respectively, which may represent the major candidacidal capacity of dialyzed parotid secretion.

The present study is concerned with the development and characterization of bioadhesive carageenan gel encapsulating Amphotericin B and Lactobacillus acidophillus, prepared by graft co-polymeriztion against Candidal vaginitis. Intravaginal gel systems based on bioadhesive polymer (carrageenan) were characterized with respect to swelling index, bioadhesive strength, percent encapsulation and in vitro drug release antimicrobial studies. A marked increase in swelling index of gel encapsulating Lactobacillus was found to be 1.9±0.35. The percent encapsulation of drug was found to be 98.63%±.0.2% and that of Lactobacillus was 91.81 ±0.01. The viability was observed for interval of 6 hrs on trypton soya agar and showed that viability was highly conserved till 4 hrs. The antimicrobial study of gels encapsulating Amphotericin B and Lactobacillus showed that carageenan gel can inhibit Candida albicans upto a maximum extent. Bioadhesivity study also conducted for gels that showed a bioadhesivity of 84.66% ±.0.5% with drug, 88.66% ±.02% with Lactobacillus. In-vitro drug release showed a sustained type release of drug from the polymer i.e. there was initial burst of Amphotericin B up to 5 hours, after which there was a sustained release upto 10 days. Thus it has been concluded from the present study that bioadhesive gels encapsulating Amphotericin B can act as promising drug carriers along with Lactobacillus against candidal vaginitis.

This study was designed to analyze the interaction of 21 antifungal combinations consisting of seven major antifungal agents against 11 echinocandin- susceptible and six-resistant C. glabrata isolates. The combinations were divided into five major groups and were evaluated by checkerboard, disc diffusion, and time-killing assays. Synergy based on the fractional inhibitory concentration index of ≤0.50 was observed in 17.65–29.41% of the cases for caspofungin combinations with azoles or amphotericin B. Amphotericin B combination with azoles induced synergistic interaction in a range of 11.76–29.41%. Azole combinations and 5-flucytosine combinations with azoles or amphotericin B did not show synergistic interactions. None of the 21 combinations showed antagonistic interactions. Interestingly, 90% of the detected synergism was among the echinocandin-resistant isolates. Disk diffusion assays showed that the inhibition zones produced by antifungal combinations were equal to or greater than those produced by single drugs. The time-killing assay showed the synergistic action of caspofungin combination with fluconazole, voriconazole, and posaconazole, and the amphotericin B-5-flucytosine combination. Furthermore, for the first time, this assay confirmed the fungicidal activity of caspofungin-voriconazole and amphotericin B-5-flucytosine combinations. The combination interactions ranged from synergism to indifference and, most importantly, no antagonism was reported and most of the synergistic action was among echinocandin-resistant isolates.

Shallot (Allium ascalonicum L.) has been known as traditional medicine. Shallot which has same genus with garlic(Allium sativum L.) contains allicin that is also found in garlic and has been suspected has fungicidal activity toCandida albicans. It is supported by several researches. Therefore, shallot is suspected has antifungal activity too.The aim of this research was to know antifungal activity of shallot’s water extortion againsts Candida albicans invitro. This was a laboratory experimental research which used completely randomized design, with diffusion method.Shallot’s water extortion was devided into three concentrations, there were 50%, 100% and 200%. Ketoconazole 2%was positive control and aquadest was negative control. The result of this research based on analysis of varians(Anova), there was significant difference between several treatments and was confirmed with Duncan New MultipleRange Test (DNMRT) p<0,05, there was significant difference between 100% shallot’s water extortion with othertreatments, but there was no significant difference between 50% shallot’s water extortion with 200% shallot’s. Theconclusion was shallot’s water extortion had antifungal activity againsts Candida albicans with the best concentration100%, but it was lower than ketoconazole 2%.

Abstrak Candida albicansb (C. albicans) adalah salah satu mikroorganisme penyebab masalah kesehatan reproduksi wanita, yaitu keputihan (fluor albus). Penggunaan larutan povidone iodine dan bahan alam seperti ekstrak daun sirih menjadi pilihan masyarakat sebagai pembersih alat kewanitaan. Tujuan penelitian ini adalah untuk membandingkandaya hambat larutan antiseptik povidone iodine dan ekstrak daun sirih terhadap jamur C. albicans secara in vitro. Penelitian dilakukan terhadap lima isolat jamur C. albicans dengan larutan kontrol akuades.Perlakuan terdiri dari povidone iodine, ekstrak daun sirih dengan konsentrasi 5%, 10%, dan 20%.Hasil penelitian menunjukkan bahwa povidone iodine memiliki daya hambat terhadap C. albicans. Ekstrak daun sirih dengan konsentrasi 5% dan 10% tidak memiliki daya hambat terhadap C. albicans, namun ekstrak daun sirih konsentrasi 20% memiliki daya hambat terhadap C. albicans. Analisis statistik dengan uji ANOVA yang dilanjutkan dengan uji Post-hoc menunjukkan perbedaan bermakna antara daya hambat larutan povidone iodine dan ekstrak daun sirih 20% terhadap kontrol(p < 0.05).Larutan povidone iodine memiliki daya hambat dua kali lebih besar terhadap pertumbuhan C. albicans dibandingkan ekstrak daun sirih 20%. Dari penelitian ini dapat disimpulkan bahwa larutan povidone iodine dan ekstrak daun sirih 20% dapat menghambat pertumbuhan jamur C. albicans secara in vitro. Kata kunci: povidone iodine, ekstrak daun sirih, Candida albicansAbstract Candida albicans (C. albicans) is one of the frequent causes of  reproductive health problems in women, namely vaginal discharge (fluor albus). The antiseptic solution, povidone iodine, is still an option to overcome vaginal discharge. The use of natural materials such as betel (Piper betle L.) leaves extract also become a popular choice as adouche for women. The objective of this study was to compare the inhibitory activity of povidone iodine solution and betel leaf extract against the growth of C. albicans in vitro. We used five different isolates of C. albicans with distilled water as control. Each isolate was treated with povidone iodine solution, betel leaf extract at concentration of 5%, 10%,and 20%. The results showed that povidone iodine had inhibitory effect on C. albicans. Betel leaf extract at concentration of 5% and 10% did not have inhibitory effectwhile betel leaf extract at concentration of 20% hadinhibitory effect on C. albicans. Analysis by ANOVA followed by Post-hoc tests showed a significant difference in inhibitory activity of povidone iodine and betel leaf extract at 20% concentration compared to control (p < 0,05). Povidone iodine solution showed twice as much as inhibitory effect on C. albicans compared to betel leaf extract (20% concentration). It is concluded that povidone iodine solution and betel leaf extract at 20% concentration can inhibit the growth of C. albicans in vitro.Keywords: povidone iodine, betel leaf extract, Candida albicans
