292 EFFECT OF A SINGLE INJECTION OF FSH DISSOLVED IN POLYETHYLENE GLYCOL ON SUPEROVULATION RESPONSE IN KOREAN NATIVE COWS

Superovulation in bovine is usually achieved after several FSH injection in a time-consuming protocol, and the half-life of FSH is very short. This study was carried out to assess the efficiency of embryo production after induction of superovulation with a single injection of FSH (Folltropin-V, Vetrepharm, Canada) dissolved in polyethylene glycol (PEG, 8000 MW, Sigma-Aldrich Corp., St. Louis, MO, USA) in Korean Native cows. Eighty-eight cows were randomly assigned to one of four groups. In Group 1 (control), cows were intramuscularly treated with twice-daily administration of 50 mg FSH for 4 days. In Group 2 and 3, cows received a single subcutaneous injection of 400 mg and 200 mg FSH dissolved in 30% PEG, respectively. In Group 4, cows received a single subcutaneous injection of 200 mg FSH dissolved in 30% PEG given seven days after CIDR insertion. On days 10 to 12 after observed estrus, superovulation was started. Luteolysis was induced by administration of 25 mg of PGF2α (Pharmacia, Puurs, Belgium) IM 2 days after the start of the FSH injection. Donors were timed-inseminated with frozen–thawed semen at 48 and 60 h after the administration of PGF2α. Embryos were collected by non-surgical methods on 7 days after the second AI. The numbers of corpus lutea (CL), total ova, transferable embryos, and progesterone concentration were analyzed by General Linear Models Procedure with the Statistical Analysis System (SAS; SAS Institute, Inc., Cary, NC, USA) and means were compared with the Duncan’s multiple range test. Data on the pregnancy rate was analyzed by chi-square test. P values <0.05 were considered to be significantly different. The number of CL in Group 2 was significantly (P < 0.05) higher compared to Groups 1, 3 and 4 (18.5 ± 7.5 v. 11.2 ± 5.1, 13.1 ± 5.1 and 13.9 ± 6.4, respectively). However, the number of total ova (7.9 ± 6.0 to 10.4 ± 7.2) and transferable embryos (3.7 ± 3.3 to 4.7 ± 3.9) did not differ among treatment groups. In addition, blood progesterone concentrations at embryo recovery did not differ among all groups (10.0 ± 3.5 ng mL–1 to 15.7 ± 3.2 ng mL–1). No difference was observed among groups in pregnancy rate after transferring the recovered embryos (36.0 to 50.0%). In conclusion, our results show that a single injection of FSH dissolved in polyethylene glycol is an efficient and simple procedure for inducing superovulation in Korean native cows.

ABSTRACT The aim of this study was to evaluate the effectiveness of a GnRH synthetic analog, as an estrous inducer in female dogs when administered during the anestrous phase, and to evaluate the pregnancy rate achieved through natural copulation. For this purpose, ten female dogs of different breeds were used. The subjects received buserelin by intramuscular injections at a dose of 2,1mcg when female dogs weighed up to 10kg (Group 1) and of 4,2mcg when the dogs weighed above 10kg (Group 2). Of the ten subjects, only three presented estrus after a single injection of buserelin: two dogs from Group 1 and one dog from Group 2 on average 7±1.29 days. The remaining seven dogs were given a second dose of buserelin, equal to the first administration. Of these, three belonged to Group 1 and four to Group 2. Four of these dogs exhibited clinical signs of estrus within, on average 9±7.3 days from the second injection. The seven female dogs that did enter estrus were fertilized successfully through natural copulation. The administration of buserelin was effective in inducing estrus in female dogs during the anestrous phase, with a maximum of two administrations.

This study aimed to evaluate the efficacy of different hormonal treatments protocols (PGF2α ,GnRH, estradiol and progesterone) hormones on reproductive performance of postpartum anoestrus native dairy River buffaloes (Bubalus bubalis), endemic south of Baghdad under field conditions . Present study was conducted on 128 animals that had postpartum anoestrus (PPA) for a period between 4 to 8 months. The animals were subjected to two experiments according to the type of anoestrus. In the first experiment 94 animals (73.5%) with persistent corpus luteum on their ovaries without any signs of estrous (sub-oestrus) were classified into two sub-groups.Sub-group1 (n=47) treated with PGF2α hormone alone and sub-group 2 (n= 47) were treated by two injections. The first injection was PGF2α. while the second injection GnRH+ PGF2α was injected after 9 days.In second experiment 34 buffalo cows without any structure on their ovaries (True anestrous) were classified into two sub-groups according to design of the treatment. Sub-group 1(n=14) was treated with estradiol as single injection. Sub-Group 2(n=20) received estradiol + progesterone .The results indicated that the pregnancy rate in sub- groups1 and 2 of the first experiment were 85.1% and 89% respectively , which was not significantly differ from each other (P < 0.05). While in the second experiment, the pregnancy rate for the first and second sub- groups were 71% and 75%, respectively. This study concluded that the prevailing situation of anestrous in postpartum buffaloes endemic south of Baghdad is anestrous with corpus luteum (Sub-oestrus) , 94 out of 128 (73.5%) , and the most efficient treatment protocol of these case are PGF2α + GnRH hormones ( pregnancy rate= 89%) . While estradiol + progesterone treatment are efficient in the treatment of animals suffering from true anestrous (pregnancy rate 75%).

SUMMARY Uterine luminal fluid accumulation (ULFA) was induced in ovariectomized rats by twice daily treatment for 3 days with 0·5 μg oestradiol-17β. At the end of this treatment, rats were given a single subcutaneous injection of 0·2 ml 0·9% saline containing 0, 0·1, 1, 10, or 100 guinea-pig units (GPU) of relaxin, and ULFA was determined 15–17 h thereafter. It was reduced in animals receiving ≥ 1 GPU relaxin. This reduction in ULFA commenced 12 h after treatment of rats with 10 GPU relaxin, and was completed by 24 h. Ligation of the cervical end of the uterus prevented the loss of fluid in response to a single injection of relaxin, indicating the cervix as the route of fluid loss. Uterine luminal fluid accumulation was reduced to a greater extent by treatment of rats with 1 mg progesterone than by 10 GPU relaxin. Given concomitantly with oestrogen, relaxin had no effect on ULFA in horns which were ligated at the cervical end, indicating that relaxin did not inhibit luminal fluid formation. It is concluded from these results that the effects of progesterone on ULFA are not mediated by relaxin. The results, however, do not allow rejection of the hypothesis that the effect of prolactin on ULFA is mediated by relaxin.

The aims of this study were to compare the embryo recovery rate in Angora goats based on application timing; at the beginning (September – October; Group 1) and end (December; Group 2) of the breeding season and to evaluate the viability and survivability of fresh or vitrified-thawed embryos when transferred. For this purpose, nine Angora goats were used as donors and thirthy Angora goats were used as recipients. Donor goats were synchronized and superovulated with traditional protocol and were mated with fertile bucks. At the 156th hour of the mating, embryos were collected surgically and evaluated under a stereo microscope. In group 1, 103 embryos and in group 2, 63 embryos were collected from nine goats. Fresh or vitrified-thawed embryos were transferred surgically to synchronized recipients. In Group 1 fresh/thawed embryos were transferred to 15/15 goats and in group 2, fresh/thawed embryos were transferred to 8/8 goats, respectively. Each recipient received 1 or 2 embryos ipsilateral to the ovary containing one or more corpora lutea. On day 30 of the transfer, goats were examined by transrectal ultrasonography, pregnancy rates of fresh/thawed embryos were 66.6%/26.6% for group 1 and 62.5%/62.5% for group 2. On day 100 of the transfer, goats were examined again by ultrasonography, and pregnancy rates were 46.6%/0% for group 1 and 37.5%/0% for group 2, respectively. After about 50 days, goats were kidded. In group 1, 3 twins and 4 single kids were born; in group 2, 2 twins were born. The total number of collected embryos and pregnancy rates among the groups were analysed using SPSS® (version 14.01, Chicago, IL, USA) and for all comparisons, differences were considered with a minimum of 5% significance level. After statistical analyses, the numbers of collected embryos at the beginning and at the end of the breeding season were compared. There was no difference in freezable/transferable embryo quality. As a result, embryos could be collected after superovulation protocols in Angora goats both at beginning and end of the breeding season, however there might be a decrease in numbers of collected embryos and the reasons for this might not be only the seasonal factors but also the environment, care, nutrition and previous superovulation protocols. The pregnancy rate following transfer of fresh embryos was satisfactory but not all does confirmed pregnant kidded; hence, reducing the number of recipients kidding. The pregnancy rate following transfer of vitrified-thawed embryos was generally low and unsatisfactory. Further research is warrented in improving the cryopreservation techniques and thus the embryo survival rate of Angora goat embryos. This study was financed with the University of Ankara Grant.

Abstract STUDY QUESTION Does an individualised luteal phase support (iLPS), according to serum progesterone (P4) level the day prior to euploid frozen embryo transfer (FET), improve pregnancy outcomes when started on the day previous to embryo transfer? SUMMARY ANSWER Patients with low serum P4 the day prior to euploid FET can benefit from the addition of daily subcutaneous P4 injections (Psc), when started the day prior to FET, and achieve similar reproductive outcomes compared to those with initial adequate P4 levels. WHAT IS KNOWN ALREADY The ratio between FET/IVF has spectacularly increased in the last years mainly thanks to the pursuit of an ovarian hyperstimulation syndrome free clinic and the development of preimplantation genetic testing (PGT). There is currently a big concern regarding the endometrial preparation for FET, especially in relation to serum P4 levels around the time of embryo transfer. Several studies have described impaired pregnancy outcomes in those patients with low P4 levels around the time of FET, considering 10 ng/ml as one of the most accepted reference values. To date, no prospective study has been designed to compare the reproductive outcomes between patients with adequate P4 the day previous to euploid FET and those with low, but restored P4 levels on the transfer day after iLPS through daily Psc started on the day previous to FET. STUDY DESIGN, SIZE, DURATION A prospective observational study was conducted at a university-affiliated fertility centre between November 2018 and January 2020 in patients undergoing PGT for aneuploidies (PGT-A) IVF cycles and a subsequent FET under hormone replacement treatment (HRT). A total of 574 cycles (453 patients) were analysed: 348 cycles (leading to 342 euploid FET) with adequate P4 on the day previous to FET, and 226 cycles (leading to 220 euploid FET) under iLPS after low P4 on the previous day to FET, but restored P4 levels on the transfer day. PARTICIPANTS/MATERIALS, SETTING, METHODS Overall we included 574 HRT FET cycles (453 patients). Standard HRT was used for endometrial preparation. P4 levels were measured the day previous to euploid FET. P4 &gt; 10.6 ng/ml was considered as adequate and euploid FET was performed on the following day (FET Group 1). P4 &lt; 10.6 ng/ml was considered as low, iLPS was added in the form of daily Psc injections, and a new P4 analysis was performed on the following day. FET was only performed on the same day when a restored P4 &gt; 10.6 ng/ml was achieved (98.2% of cases) (FET Group 2). MAIN RESULTS AND THE ROLE OF CHANCE Patient’s demographics and cycle parameters were comparable between both euploid FET groups (FET Group 1 and FET Group 2) in terms of age, weight, oestradiol and P4 levels and number of embryos transferred. No statistically significant differences were found in terms of clinical pregnancy rate (56.4% vs 59.1%: rate difference (RD) −2.7%, 95% CI [−11.4; 6.0]), ongoing pregnancy rate (49.4% vs 53.6%: RD −4.2%, 95% CI [−13.1; 4.7]) or live birth rate (49.1% vs 52.3%: RD −3.2%, 95% CI [−12; 5.7]). No significant differences were also found according to miscarriage rate (12.4% vs 9.2%: RD 3.2%, 95% CI [−4.3; 10.7]). LIMITATIONS, REASONS FOR CAUTION Only iLPS through daily Psc was evaluated. The time for Psc injection was not stated and no serum P4 determinations were performed once the pregnancy was achieved. WIDER IMPLICATIONS OF THE FINDINGS Our study provides information regarding an ‘opportunity window’ for improved ongoing pregnancy rates and miscarriage rates through a daily Psc injection in cases of inadequate P4 levels the day previous to FET (P4 &lt; 10.6 ng/ml) and restored values the day of FET (P4 &gt; 10.6 ng/ml). Only euploid FET under HRT were considered, avoiding one of the main reasons of miscarriage and implantation failure and overcoming confounding factors such as female age, embryo quality or ovarian stimulation protocols. The potential of laparoscopic ovum pick-up (LOPU) followed by in vitro embryo production (IVEP) as a tool for accelerated genetic programs in ruminants is reviewed in this article. In sheep and goats, the LOPU-IVEP platform offers the possibility of producing more offspring from elite females, as the procedure is minimally invasive and can be repeated more times and more frequently in the same animals compared with conventional surgical embryo recovery. On average, ~10 and ~14 viable oocytes are recovered by LOPU from sheep and goats, respectively, which results in 3–5 transferable embryos and >50% pregnancy rate after transfer. LOPU-IVEP has also been applied to prepubertal ruminants of 2–6 months of age, including bovine and buffalo calves. In dairy cattle, the technology has gained momentum in the past few years stemming from the development of genetic marker selection that has allowed predicting the production phenotype of dairy females from shortly after birth. In Holstein calves, we obtained an average of ~22 viable oocytes and ~20% transferable blastocyst rate, followed by >50% pregnancy rate after transfer, declaring the platform ready for commercial application. The present and future of this technology are discussed with a focus on improvements and research needed.

SUMMARY Ovariectomized rats treated with 2·5 or 5·0 mg. progesterone daily received a single subcutaneous injection of 0·2 μg. oestradiol on the third day of the progesterone treatment. The deciduomal response to trauma by endometrial scratching was used to determine the degree of uterine sensitivity at various times after oestradiol. Uterine sensitivity was partially and then completely lost 36 and 48 hr. after oestradiol administration. The inhibition of uterine sensitivity persisted until 9 and 11 days after oestradiol when the animals received 2·5 and 5·0 mg. progesterone daily. Uterine sensitivity was completely inhibited on day 11 with doses of oestradiol from 0·2 to 0·05 μg. Withdrawal of progesterone treatment for 48 or 72 hr., but not for 24 hr., after oestradiol treatment restored uterine sensitivity. These results show that the oestradiol-induced inhibition of uterine sensitivity in the progestational endometrium is maintained by continuous progesterone treatment and that the duration of this effect is dependent on the dose of progesterone given.
