Plant genotype, juvenility and mechanisms of inhibition of rooting Persoonia virgata R. Br. Cuttings

Summary. Persoonia virgata is an endemic shrub of Australia that is being bush-harvested for its foliage and sold on the domestic and export flower markets. The limiting factor to the cultivation of many desirable Persoonia species has been the extreme difficulty in developing a reliable propagation system. This study investigated the effects of cutting type, maturity of the stock plant, and genotype, on the rooting of P. virgata cuttings. It was found that juvenile cuttings rooted significantly better than mature cuttings. Some juvenile seedling genotypes yielded no rooted cuttings, whereas others produced up to 90% rooting. No anatomical barrier to root formation was discovered. High performance liquid chromatography showed differences in the abundance of unidentified compounds in cuttings, related to season and to genotypic differences in rooting abilities. A possible seasonal rooting inhibitor was present in some genotypes. The potential for selection of genotypes with improved rooting ability has been demonstrated.

Summary We have evaluated a new commercially available enzyme-linked immunorsorbant assay for neopterin :or its suitability in the context of screening of voluntary blood donors. The assay was performed on 1040 consecutive blood donors, and compared with radioimmunoassay and. in a fraction of 142 donors . . : Iso with high performance liquid chromatography. On repetitive assays of all donations showing a concentration exceeding 8.0 nmol/L in the initial assay. three of the radioimmunoassay results were identified as gross outliers. No such gross outliers were detected for the enzyme-linked immunosorbant assay. RegarJing the reproducibility of results exceeding a cut-off limit of \0 nmol/L neopterin. the enzyme-linked ;mmunosorbant assay was better than the radioimmunoassay. Moreover. the enzyme-linked immunosorbant assay was slightly superior to radioimmunoassay when both tests were compared with high performance liquid chromatography (based on linear regression analysis. evaluation of frequencies of concentrations bant assay was slightly superior to radioimmunoassay when both tests were compared with high performance liquid chromatography (based on linear regression analysis. evaluation of frequencies of concentrations rations. Its slight superiority compared to the conventional radioimmunoassay likely results from the higher degree of automatization employed.

An analysis of the yield of eleutherosides B and E inAcanthopanax divaricatusandA. koreanumwas performed using high performance liquid chromatography to evaluate production by different cultivation methods. InA. divaricatusandA. koreanum, the total content of eleutherosides B and E was 2.466–7.360 mg/g varying by plant section, 3.886–11.506 mg/g by pinching site, 3.655–10.083 mg/g by planting time, and 3.652–10.108 mg/g by fertilizer ratio. Thus the total content of eleutherosides B and E inA. divaricatusandA. koreanumdiffered depending on cultivation methods. These results present useful information for high eleutheroside content applications inA. divaricatusandA. koreanum. This information can affect selection of plant section and cultivation methods for nutraceutical, pharmaceutical, and cosmeceutical material development.
