Effect of mulberry silage supplementation during late fattening stage of Hanwoo (Bos taurus coreanae) steer on antioxidative enzyme activity within the longissimus muscle

This study was conducted to determine the possible application of mulberry silage as a functional feed in the feeding management of Hanwoo, Korean native cattle, for high quality beef production. Twenty Hanwoo steers in the last stage of fattening were randomly assigned to two groups with (MSS) or without (Control) supplemental mulberry silage. The Control group was fed on a standard total mixed commercial ration, while the MSS group was fed on a total mixed commercial ration formulated with mulberry silage at the rate of 10%, fresh matter basis, for 30 days. The mulberry silage contained active ingredients such as 1-deoxynojirimycin, γ-aminobutyric acid, and phenol compounds. In the present study, the oxidative stress biomarkers such as glutathione, superoxide dismutase, glutathione peroxidase, catalase, and glutathione-S-transferase were improved in the MSS group compared with the Control group. Electron spin resonance assays showed that 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity was significantly elevated by 75.5% in the MSS group (P < 0.001). Moreover, the MSS group exhibited stronger scavenging activity against hydroxyl radicals, superoxide radicals, and alkyl radicals, and the radical scavenging activity occurred in a dose-dependent manner. Findings from the present study suggest that mulberry silage may be useful as a functional feed by improving the antioxidant activity against various reactive radicals for beef cattle.

A preparation of crude polysaccharides (TPS) was isolated from Camellia sinensis by precipitation and ultrafiltration. TPS1, TPS2, and TPS3 had molecular weights of 240, 21.4, and 2.46 kDa, respectively. The radical scavenging activities of TPS were evaluated by DPPH free radical, hydroxyl radical and superoxide radical scavenging. These results revealed that TPS exhibited strong radical scavenging activity in a concentration-dependent manner. TPS3 with lowest molecular weight showed a higher radical scavenging activity.

Background: Cataract is a major cause of visual impairment and blindness around the world. This study evaluated the in vitro antioxidant and anti-cataract activities of Cnidoscolus aconitifolius leaves extract and fractions. Antioxidant activities were evaluated by 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2-azinobis (3-ethylbenzothiozoline-6-sulfonic acid) (ABTS), total reducing power, and hydrogen peroxide scavenging assays. Anti-cataract potential was evaluated in vitro using goat lenses divided into eight groups of different treatments and incubated in artificial aqueous humor at 37 °C for 72 hours. Glucose-induced opacity in the lenses was observed and biochemical indices quantified (catalase, Malondialdehyde (MDA) and total protein in the lens homogenate). Results: The crude extract and its fractions possess substantial antioxidant activities. The aqueous fraction exhibited the best DPPH radical scavenging activity (IC50 value 78.599 µg/ml); while the dichloromethane fraction exhibited the highest ABTS radical scavenging activity with IC50 66.68 µg/ml. The anti-cataract evaluation of crude and fractions at 250 μg/ml showed a significant increase (p<0.05) in the total protein and catalase activity compared to the cataract control group. The malonaldehyde level decreased significantly (p<0.05) in all the treated groups.

Light-emitting diodes (LEDs) were used for the spring greenhouse cultivation of eggplant (Solanum melongena L.) ‘Milar F1’ and tomato (S. lycopersicon L.) ‘Benito F1’ transplants. Seedlings were grown under natural light conditions with the supplemental LED light. A 16-h photoperiod provided plants with a DLI of 12.6 (eggplant) and 9.6 (tomato) mol m2/day. Four supplemental light spectra were tested: L1 (90% red + 10% blue); L2 (80% red + 20% blue); L3 (43% red + 42% blue+15% green) and L4 (56% red + 26% blue + 15% green + 3% UV-A). The PPFD in each LED light treatment was 150 ± 20 µmol/m2·s. Compared to the control plants (without LED lighting), the eggplant transplants had about a 25% larger leaf area and a higher level of total phenol content as well as a higher antiradical scavenging activity under the L1 spectrum. The favourable spectrum for the tomato transplants consisted of red to blue in a ratio of 1 : 1 mixed with a green light (L3) – the leaves were characterised by a higher content of dry matter, soluble sugars, photosynthetic pigments and total phenols; also the radical scavenging activity increased in comparison to the control group. It was shown that the supplemental irradiation of transplants was economically acceptable.

Anthocyanins from various vegetables and fruits have antioxidant activities, however, the bioactivities of coloured potato anthocyanins are not well studied. We examined the antioxidant capacities of pigmented fractions from purple potato flakes in vitro, and the antioxidant potentials of purple potato flakes in vivo. 1,1-Diphenyl-2-picrylhydrazyl radical scavenging activity of the pigmented fraction from Hokkai no. 92 (H92) potato flakes was higher than that from Kitamurasaki (KM) potato flakes. Extracts equivalent to 600 μg pigmented fractions from KM and H92 potato flakes inhibited linoleic acid oxidation in the order trolox>H92 ≥ KM>control. Rats were fed 25 % KM or H92 potato flake diets for 4 weeks. The major anthocyanin was identified as petanin. Control rats were fed a diet with cornstarch instead of potato flakes for 4 weeks. The serum antioxidant potential level in the H92 group was significantly higher than that in the control group. The degree of hepatic lipid peroxidation in the H92 group was significantly lower than that in the control group. Hepatic Cu/Zn-superoxide dismutase (SOD), Mn-SOD and glutathione peroxidase (GSH-Px) mRNA levels in the H92 group were significantly higher than those in the control group. Similar significant differences in Cu/Zn-SOD and Mn-SOD mRNA levels between the KM and control groups were found. The present results suggest that purple potato flakes have antioxidant functions with regard to radical scavenging activity and inhibition of linoleic acid oxidation, and that they improve the antioxidant potentials in rats by enhancing hepatic Mn-SOD, Cu/Zn-SOD and GSH-Px mRNA expression.

The methanol extract of the bark of Machilus odoratissima was subjected to investigate its antioxidant and antibacterial properties. The phytochemical screening demonstrated the presence of different types of compound like terpenoids, tannins, deoxy sugar, saponins and phenolic compounds. The methanol extract of the plant was tested for antioxidant activity using scavenging activity of DPPH(1,1-diphenyl-2-picrylhydrazil) radical method and antibacterial activity against Staphylococcus aureus, Enterococcus faecalis, Escherichia coli & Pseudomonas aeruginosa bacteria using cup plate method. The extract exhibited high free radical scavenging activity. IC50 was found to be 3.37μg/ml. Objective: Bauhinia acuminate L (Fabaceae) is a species of flowering shrub native to tropical south eastern Asia. The flowerof B. acuminataare used against various skin diseases,worms, tumours and diabetes. The Indian vaiydas recommended as a remedy to treat severe biliousness.The qualitative and quantitative estimation of phytochemical constituents in various extracts and the antioxidant potential of flower of Bauhinia acuminata were evaluated.Methods: All the analysis was done based on standard protocols.Results: The phytochemical screening reveals the presence of various primary and secondary metabolites like sugar, carbohydrate, amino acid, fat, quinone, steroids, phenol, saponin, alkaloids, and acid content. In quantitative analysis the important secondary metabolites such as alkaloids and phenolic compounds were estimated in all the extracts.The ethanol and chloroformextract produced significant antioxidant properties in a dose-dependent manner. DPPH free radical scavenging assay of ethanol and chloroform extracts of flower exhibited IC 50 values of 24.44±1.201μg/ml and 196.68±0.456μg/ml respectively. At 1000 μg/ml concentration both extracts shows maximum radical scavenging activity (98.97% and 85.67 %).Conclusion: The findings of this study indicate that this plant is medicinal with prominent antioxidant property.

Sygygium cumini, commonly known as Malabar plum, Java plum, black plum, jamun or jambolan, is an evergreen tropical tree in the flowering plant family Myrtaceae. The present study the antioxidant effects of microparticles of S. cumini plant extract were evaluated using in vitro, 2, 2-diphenylpicrylhydrazine photometric assay, total antioxidant activity and super oxide free radical scavenging activity. The microparticles showed different levels of radicals scavenging activity in a dose-dependent manner over the range of 50–150 µg/mL concentration, indicating the high antioxidative capacity of the extract. In all the experimental screening models microparticles of aqueous extract show the maximum inhibition of free radicals comparison of other experimental model at different concentrations. These findings suggest that the seed of Syginium cumini has potent antioxidant activity which may be responsible for some of its reported pharmacological activities and can be used as antioxidant supplement.

The research has been performed on roots of <em>Vitis vinifera</em>, cv. Himrod, obtained from seedlings grown under chill stress conditions (+10<sup>o</sup>C in the day and +7<sup>o</sup>C at night), under optimum conditions (+25<sup>o</sup>C in the day and +18<sup>o</sup>C at night) and from seedling which underwent a recover period after the chill stress treatment. The purpose of the study has been to determine quantitative and qualitative changes in phenolic compounds as well as to demonstrate changes in antiradical properties of extracts from grapevine roots, which appeared as a result of chill stress and during recovery under the optimum conditions following the stress. Phenolic compounds from grapevine roots were extracted using 80% acetone. The total content of phenolics was determined by colorimetry. The content of tannins was tested by precipitation with bovine serum albumin. The reducing power as well as DPPH• free radical and ABTS+• cation radical scavenging activity of the extracts were also tested. In order to identify phenolic compounds present in the extracts the RP-HPLC technique was employed. The tested material was found to contain tannins and three identified phenolic acids: ferulic, caffeic and p-coumaric ones. The latter occurred in the highest concentrations (from 4.46 to 6.28 µg/g fresh matter). Ferulic acid appeared in smaller amounts (from 1.68 to 2.65 µg/g fresh matter), followed by caffeic acid (from 0.87 to 1.55 µg/g fresh matter). Significantly less total phenolic compounds occurred in roots of seedlings subjected to chill stress. However, the total content of these compounds increased significantly in roots of plants which underwent recovery after chill stress. Concentration of tannins was determined by two methods. The content of condensed tannins was depressed in roots as a result of low temperature stress, whereas the content of condensed and hydrolysing tannins (determined via the BSA method) rose under chill stress conditions. A significant increase in tannins in root extracts (determined with both methods) was found during the recovery process after the stress. The three identified phenolic acids appeared in grapevine roots as ester-bound compounds. It has been demonstrated that the content of phenolic acids significantly fell as a result of low temperatures, but increased during recovery after chill stress. The weakest ability to scavenge DPPH• and ABTS+• free radicals as well as the reducing power were shown by the extract obtained from grapevine roots from the seedlings subjected to chill stress. Both free radical scavenging activity and reducing power were observed to increase considerably during recovery after stress. This seems to prove that during the recovery process following chill stress the synthesis of antioxidative compounds in grapevine roots is much more intensive.

Introduction: Oxidative stress is involved in many diseases, including hypertension, kidney failure, and heart disease. This study aimed to evaluate the effect of hydroalcoholic Cichorium intybus extract on blood pressure in rats. Antioxidant activity, phenolic and flavonoid contents of the plant extract were also evaluated. Methods: In this study, 32 male Wistar rats weighing 250-300 g were divided into four groups of eight each. Animals in the control group were administered with normal saline and in the C. intybus groups with extract at 25, 50, and 100 mg/kg for two weeks. Then, the homodynamic parameters were examined by the Power lab. The phenolic and flavonoid contents were also evaluated by a spectrophotometer and the rate of free radical scavenging activity was measured by the diphenyl-1-picyryl-hydrazyl (DPPH) free radical method. Results: The free radical scavenging activity of C. intybus extract was obtained 47.85% of DPPH, and flavonoid and phenolic contents were 8.21 and 27.19 mg/g of dry extract, respectively. Meanwhile, median (MAP), systolic (SAP) and diastolic arterial pressure (DAP) significantly decreased in the 50 mg/kg extract-treated group compared to the control and 200 mg/kg extract-treated groups. Conclusion: Ethanol extract of C. intybus plays a protective role against hypertension, which, in part, might be due to antioxidant compounds of the plant against free radicals.

Pteridophytes have been widely used in several systems of medicine. Several reports have increasingly assessed their bioactive effects, but for Sphaerostephanos unitus (L.) Holttum, only its antibacterial potential has been assessed. In this sense, the present study was carried out to reveal the phytochemical profile and to determine the toxicity, antioxidant, antidiabetic, and anti-inflammatory potential of S. unitus. Brine shrimp lethality, 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, phosphomolybdenum assay, superoxide radical scavenging activity, 2,2-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) assay (ABTS), and in vitro α-amylase inhibitory and membrane stabilization assays were applied. S. unitus extract toxicity showed variable mortality percentages, with LC50 values ranging from 4 to 30 mg/mL. DPPH radical scavenging effects of S. unitus extracts were as follows: methanol > acetone > petroleum ether > chloroform. S. unitus acetone extract displayed the strongest phosphomolybdenum reduction (10 ± 2 mg Ascorbic Acid Equivalent/g). The studied extracts also revealed efficient, superoxide scavenging effects in a dose-dependent manner. In S. unitus, the highest ABTS radical scavenging rate was observed in the chloroform extract (3000 ± 40 µmol/g). The S. unitus anti-inflammatory effect was as follows: petroleum ether > chloroform > methanol > acetone. In S. unitus extract, the highest percentage of α-amylase activity (80%) was observed for the petroleum ether extract (25 µg/mL). Faced with these findings, further studies should be performed to isolate and identify the S. unitus compounds responsible for their antioxidant, antidiabetic and anti-inflammatory effects.
