Effects of porcine reproductive and respiratory syndrome virus infection on the performance of pregnant gilts and growing pigs

This study examined the effects of porcine reproductive and respiratory syndrome virus (PRRSV) infection on (1) pregnant and (2) growing Landrace and crossbred (Large White × Pietrain) pigs. First, recently pregnant gilts were spilt into a control and a challenged group, which was inoculated with the PRRSV, and phenotypic effects were recorded. In the Landrace breed, infected gilts had a significantly reduced number of fetuses (8.9 versus 11.8), but there were insufficient data to make the same comparison in crossbred gilts. The Landrace had a smaller suppression of weight gain during pregnancy than the crossbred line [56% (0.66 to 0.29 kg/day) versus 85% (0.45 to 0.07 kg/day) reduction], suggesting greater tolerance of the PRRSV infection. Second, impacts on growing pigs were examined with the same deliberate challenge methodology. Some pigs appeared not to become infected from the initial inoculation, but were possibly subsequently infected by cohorts. However, there were indications of Landrace line resistance in terms of an increased time to seroconvert, with weight gain patterns also suggesting Landrace tolerance. In summary, this study demonstrated that breeds differ consistently in phenotypic impacts of PRRSV infection.

This study was carried out to determine the effect of object combination as environmental enrichment on behaviour of growing pigs. Thirty-six (36) crossbreed (Large White X Landrace) growing pigs with average weight of 34-36Kg were allotted randomly into four treatments, replicated three times in a completely randomized design. The four treatments were; Control (without enrichment), PRope (2 pieces of polyester rope), treatment SWood (2 pieces of soft wood) and treatment PRope+SWood (1 piece of soft wood and 1 piece of polyester rope). Behavioural observations were monitored by CCTV and recordings were made for 6 hours/day and 3days/week. At the end of the experiment, data collected includes average feed intake and average weight gain while feed conversion ratio was calculated. Likewise, behavioural observations; enrichment use, general activities, pen manipulation and pen-mate manipulation were observed. Behavioural data were analyzed using repeated measures analysis of variance (ANOVA) while performance data were analysed using one-way ANOVA. Performance data were significantly influenced (P<0.05) by enrichments for all the parameters measured. PRope+SWood had the highest value for the final weight (50.63kg), average weight gain (15.95kg) and average feed intake (56.61kg) while PRope had the highest FCR 4.18). Penmate manipulation (10.48%), pen manipulation (19.02%) and general activities (14.62%) were lower and significantly better for PRope+SWood compared to PRope and SWood and the highest interaction with enrichment objects was observed in the PRope+SWood (55.88%). It can be concluded that the use of combination of enrichment objects enhanced the performance and behaviour of pigs.

Sixty (60) grower-finisher pigs (Landrace x Large white) were used to investigate the nutritional value of beniseed hull (BSH) as substitute for maize on performance, carcass, and organs characteristics of the experimental animals. Five experimental diets were formulated by incorporating BSH into basal diet to replace maize at 0, 25, 50, 75 and 100%; designated as T1 (control), T2, T3, T4 and T5, respectively. The experimental pigs were individually weighed and assigned to five dietary treatments of three replicates of four animals each in a completely randomized design. The animals were fed for 2 weeks pre-experimental period and thereafter for ten weeks during which data were collected. Feed cost per kg live weight gain (? 668.67) of the animals fed 0% BSH significantly (P<0.05) reduced to ? 620.06 (25% BSH), ? 527.24 (50% BSH), ? 509.74 (75% BSH) and ? 456.73 (100% BSH). The bled weight, scald weight, eviscerated weight, dressing percentage, bacon, fore hock, hind hock and tail were significantly (P<0.05) affected by dietary treatments. The biggest liver weight (2.27g/kg) obtained in the animals fed the control diet (0% BSH) reduced significantly (P<0.05) to 2.20 (25% BSH), 1.91 (50% BSH), 1.76 (75% BSH) and 2.00 (100% BSH). Variations obtained in the kidney weight (0.40 g/kg) of the experimental animals fed with 25% BSH-based diets were similar (P>0.05) to 0.38 g/kg in the control (0% BSH) with both significantly (P<0.05) reduced to 0.36 (50% BSH), 0.36 (75% BSH) and 0.32 (100% BSH), respectively. Beniseed hull (BSH) poses no health challenge to the pigs used in this study. There were reductions in the cost/kg and cost of feed per kilogram live-weight gain with increasing levels of BSH in the diets, the lowest being at 100% level of substitution; beniseed hull could economically and safely replace maize in pig's diets up to 100% level.

In gilts and sows, the more severe clinical manifestation of porcine reproductive and respiratory syndrome virus (PRRSV) occurs in late gestation and can result in up to a 40% abortion incidence. Despite the known genetic component in resilience to PRRSV, there is scarce information regarding the abortive outcome of this disease. We tested the relationship between eight molecular markers (six from published studies and two identified in the present study in the HDAC6 gene) and the probability of abortion during a PRRSV outbreak, using data from two commercial Landrace x Large White sow farms with an incidence of abortion of 35% and 17%. From the markers tested, USP18_-1533G>A did not segregate in these populations, and CD163_c.3534C>T and HDAC6_g.2360C>T did not affect the abortion rate. In contrast, the minor allele of two markers in SSC4 (WUR1000125 in GBP1 and rs340943904 in GBP5), which lower viremia in growing pigs, and the major alleles of CD163_rs1107556229 and HDAC6_rs325981825 were associated with a lower probability of abortion during PRRSV outbreaks. The more striking result was for the MX1 gene, where the odds ratio of aborting versus not aborting was nine times lower in the sows homozygous for a 275-bp insertion than in the other genotypes. Interactions between markers were not relevant. All together, we bring here the first evidence that mutations in the host genome can predispose or protect from complete reproductive failure in sows infected with PRRSV.

Pig production is one of the fastest growing livestock sectors. Development of this sector is hampered by rapidly increasing costs of fishmeal (FM), which is a common protein source in animal feeds. Here, we explored the potential of substituting FM with black soldier fly larval meal (BSFLM) on growth and blood parameters of pigs as well as economic aspects. At weaning, 40 hybrid pigs, i.e., crossbreeds of purebred Large White and Landrace were randomly assigned to five iso-nitrogenous and iso-energetic dietary treatments: Control (0% BSFLM and 100% FM (T0)), and FM replaced at 25% (T25), 50% (T50), 75% (T75) and 100% (T100) with BSFLM. Average daily feed intake (ADFI), average daily gain (ADG), body weight gain (BWG) and feed conversion ratio (FCR) were calculated for the whole trial. Hematological and serum biochemical parameters, the cost–benefit ratio (CBR) and return on investment (RoI) were evaluated. No significant effect of diet type was observed on feed intake and daily weight gain. Red or white blood cell indices did not differ among diets. Pigs fed T25, T75 and T100, had lower platelet counts compared to T0 and T50. Dietary inclusion of BSFLM did not affect blood total cholesterol, triglycerides, low-density lipoprotein and high-density lipoprotein. CBR and RoI were similar for the various diets. In conclusion, BSFLM is a suitable and cost-effective alternative to fishmeal in feed for growing pigs.

Abstract Immunoglobulin G antibody response, measured as sample-to-positive (S/P) ratio, to Porcine Reproductive and Respiratory Syndrome virus (PRRSV) has been proposed as an indicator trait for improved reproductive performance in PRRSV-infected purebred sows and PRRSV-vaccinated crossbred gilts. In this study, we investigated the genetic correlations (rg) of S/P ratio following a PRRSV outbreak and PRRSV-vaccination with performance in non-exposed and PRRSV-exposed sows. PRRSV outbreak phase was defined based on previously described methodologies after the detection of typical clinical signs of PRRSV infection. 541 Landrace sows had S/P ratio measured at ~54 days after the beginning of the PRRSV outbreak (S/Poutbreak), and 906 Landrace x Large White naïve F1 gilts had S/P ratio measured at ~50 days after vaccination with a commercial modified live PRRSV vaccine (S/PVx). 711 and 428 Landrace sows had reproductive performance recorded before and during the PRRSV outbreak, respectively. 811 vaccinated F1 animals had farrowing performance for up to 3 parities. All animals were genotyped for ~28K SNPs. The estimate of rg of S/Poutbreakwith S/PVx was high (rg±SE = 0.72±0.18). Estimates of rg of S/Poutbreak with reproductive performance in F1 sows were low to moderate, ranging from 0.05±0.23 (number stillborn) to 0.30±0.20 (total number born). Estimates of rg of S/PVxwith reproductive performance in non-infected purebred sows were moderate and favorable with number born alive (0.50±0.23), but low (0 to -0.11±0.23) with litter mortality traits. Estimates of rg of S/PVx were moderate and negative (-0.47±0.18) with the number of mummies in PRRSV-infected purebred sows and low with other traits (-0.29±0.18 for total number born to 0.05±0.18 for number stillborn). These results indicate that selection for antibody response following a PRRSV outbreak collected in purebred sows and to PRRSV vaccination collected in commercial crossbred gilts may increase litter size of non-infected and PRRSV-exposed purebred and commercial crossbred sows.

In July 2019, a vaccine-derived recombinant Porcine reproductive and respiratory syndrome virus 1 strain (PRRSV-1) (Horsens strain) infected more than 40 Danish sow herds, resulting in severe losses. In the present study, the pathogenicity of the recombinant Horsens strain was assessed and compared to a reference PRRSV-1 strain using a well-characterized experimental model in young SPF pigs. Furthermore, the efficacies of three different PRRSV-1 MLV vaccines to protect pigs against challenge with the recombinant strain were assessed. Following challenge, the unvaccinated pigs challenged with the Horsens strain had significant increased viral load in serum compared to all other groups. No macroscopic changes were observed at necropsy, but tissue from the lungs and tonsils from almost all pigs were PRRSV-positive. The viral load in serum was lower in all vaccinated groups compared to the unvaccinated group challenged with the Horsens strain, and only small differences were seen among the vaccinated groups. The findings in the present study, combined with two other recent reports, indicate that this recombinant “Horsens” strain indeed is capable of inducing infection in growing pigs as well as in pregnant sows that is comparable to or even exceeding those induced by typical PRRSV-1, subtype 1 strains. However, absence of notable clinical signs and lack of significant macroscopic changes indicate that this strain is less virulent than previously characterized highly virulent PRRSV-1 strains.

Abstract Porcine reproductive and respiratory syndrome virus (PRRSV) is an economically-important disease and ingestion of soy isoflavones (ISF) may benefit PRRSV-infected pigs due to demonstrated anti-inflammatory and anti-viral properties. The objective of this study was to quantify long-term effects of ISF consumption on fecal microbiome characteristics under disease challenge. In total, 96 weaned barrows were group-housed in a BSL-2 containment facility and allotted to 1 of 3 experimental treatments that were maintained throughout the wean-to-finish study: non-infected pigs receiving an ISF-devoid control diet (NC, n=24), and infected pigs receiving either the control diet (PC, n=36) or that supplemented with total ISF in excess of 1,600 mg/kg (ISF, n=36) (Table 1). Following a 7-day adaptation, pigs were inoculated intranasally with either a sham-control (PBS) or live PRRSV (1×105 TCID50/mL, strain NADC20). Fecal samples were collected from 48 individual pigs at pre-infection (-2 days post-inoculation, DPI), peak-infection (10 DPI), and post-infection (144 DPI) time-points and extracted DNA was used for 16S bacterial rRNA sequencing. Differences in bacterial communities among diet groups were evaluated using UniFrac distance matrices (weighted and unweighted) in QIIME. All other data were analyzed by one-way ANOVA performed on transformed data using R. Across all time-points, only minimal differences were observed due to ISF alone. At 10 DPI, PRRSV infection reduced Prevotella 9 genera abundance from approximately 20% to less than 10%, but the specific function of this variety in pigs is unclear. The most notable finding was decreased relative abundance of Actinobacteria at 144 DPI between non-infected and infected treatments (P &lt; 0.05), which is consistent with various dysbioses observed in other disease models. Our findings indicate that differences present were mainly due to PRRSV infection and not strongly influenced by ISF ingestion, which implies previously observed performance benefits conferred by dietary ISF are not likely due to changes in microbiome composition.
