This study examined the effects of porcine reproductive and respiratory syndrome virus (PRRSV) infection on (1) pregnant and (2) growing Landrace and crossbred (Large White × Pietrain) pigs. First, recently pregnant gilts were spilt into a control and a challenged group, which was inoculated with the PRRSV, and phenotypic effects were recorded. In the Landrace breed, infected gilts had a significantly reduced number of fetuses (8.9 versus 11.8), but there were insufficient data to make the same comparison in crossbred gilts. The Landrace had a smaller suppression of weight gain during pregnancy than the crossbred line [56% (0.66 to 0.29 kg/day) versus 85% (0.45 to 0.07 kg/day) reduction], suggesting greater tolerance of the PRRSV infection. Second, impacts on growing pigs were examined with the same deliberate challenge methodology. Some pigs appeared not to become infected from the initial inoculation, but were possibly subsequently infected by cohorts. However, there were indications of Landrace line resistance in terms of an increased time to seroconvert, with weight gain patterns also suggesting Landrace tolerance. In summary, this study demonstrated that breeds differ consistently in phenotypic impacts of PRRSV infection.

In July 2019, a vaccine-derived recombinant Porcine reproductive and respiratory syndrome virus 1 strain (PRRSV-1) (Horsens strain) infected more than 40 Danish sow herds, resulting in severe losses. In the present study, the pathogenicity of the recombinant Horsens strain was assessed and compared to a reference PRRSV-1 strain using a well-characterized experimental model in young SPF pigs. Furthermore, the efficacies of three different PRRSV-1 MLV vaccines to protect pigs against challenge with the recombinant strain were assessed. Following challenge, the unvaccinated pigs challenged with the Horsens strain had significant increased viral load in serum compared to all other groups. No macroscopic changes were observed at necropsy, but tissue from the lungs and tonsils from almost all pigs were PRRSV-positive. The viral load in serum was lower in all vaccinated groups compared to the unvaccinated group challenged with the Horsens strain, and only small differences were seen among the vaccinated groups. The findings in the present study, combined with two other recent reports, indicate that this recombinant “Horsens” strain indeed is capable of inducing infection in growing pigs as well as in pregnant sows that is comparable to or even exceeding those induced by typical PRRSV-1, subtype 1 strains. However, absence of notable clinical signs and lack of significant macroscopic changes indicate that this strain is less virulent than previously characterized highly virulent PRRSV-1 strains.

Abstract Porcine reproductive and respiratory syndrome virus (PRRSV) is an economically-important disease and ingestion of soy isoflavones (ISF) may benefit PRRSV-infected pigs due to demonstrated anti-inflammatory and anti-viral properties. The objective of this study was to quantify long-term effects of ISF consumption on fecal microbiome characteristics under disease challenge. In total, 96 weaned barrows were group-housed in a BSL-2 containment facility and allotted to 1 of 3 experimental treatments that were maintained throughout the wean-to-finish study: non-infected pigs receiving an ISF-devoid control diet (NC, n=24), and infected pigs receiving either the control diet (PC, n=36) or that supplemented with total ISF in excess of 1,600 mg/kg (ISF, n=36) (Table 1). Following a 7-day adaptation, pigs were inoculated intranasally with either a sham-control (PBS) or live PRRSV (1×105 TCID50/mL, strain NADC20). Fecal samples were collected from 48 individual pigs at pre-infection (-2 days post-inoculation, DPI), peak-infection (10 DPI), and post-infection (144 DPI) time-points and extracted DNA was used for 16S bacterial rRNA sequencing. Differences in bacterial communities among diet groups were evaluated using UniFrac distance matrices (weighted and unweighted) in QIIME. All other data were analyzed by one-way ANOVA performed on transformed data using R. Across all time-points, only minimal differences were observed due to ISF alone. At 10 DPI, PRRSV infection reduced Prevotella 9 genera abundance from approximately 20% to less than 10%, but the specific function of this variety in pigs is unclear. The most notable finding was decreased relative abundance of Actinobacteria at 144 DPI between non-infected and infected treatments (P < 0.05), which is consistent with various dysbioses observed in other disease models. Our findings indicate that differences present were mainly due to PRRSV infection and not strongly influenced by ISF ingestion, which implies previously observed performance benefits conferred by dietary ISF are not likely due to changes in microbiome composition.

Heat stress (HS) diminishes animal production, reducing muscle growth and increasing adiposity, especially in swine. Excess heat creates a metabolic phenotype with limited lipid oxidation that relies on aerobic and anaerobic glycolysis as a predominant means of energy production, potentially reducing metabolic rate. To evaluate the effects of HS on substrate utilization and energy expenditure, crossbred barrows (15.2 ± 2.4 kg) were acclimatized for 5 days (22 °C), then treated with 5 days of TN (thermal neutral, 22 °C, n = 8) or HS (35 °C, n = 8). Pigs were fed ad libitum and monitored for respiratory rate (RR) and rectal temperature. Daily energy expenditure (DEE) and respiratory exchange ratio (RER, CO2:O2) were evaluated fasted in an enclosed chamber through indirect calorimetry. Muscle biopsies were obtained from the longissimus dorsi pre/post. HS increased temperature (39.2 ± 0.1 vs. 39.6 ± 0.1 °C, p < 0.01) and RER (0.91 ± 0.02 vs. 1.02 ± 0.02 VCO2:VO2, p < 0.01), but decreased DEE/BW (68.8 ± 1.7 vs. 49.7 ± 4.8 kcal/day/kg, p < 0.01) relative to TN. Weight gain (p = 0.80) and feed intake (p = 0.84) did not differ between HS and TN groups. HS decreased muscle metabolic flexibility (~33%, p = 0.01), but increased leucine oxidation (~35%, p = 0.02) compared to baseline values. These data demonstrate that HS disrupts substrate regulation and energy expenditure in growing pigs.

The nematode Caenorhabditis elegans has been a versatile model for understanding the molecular responses to abiotic stress and pathogens. In particular, the response to heat stress and virus infection has been studied in detail. The Orsay virus (OrV) is a natural virus of C. elegans and infection leads to intracellular infection and proteostatic stress, which activates the intracellular pathogen response (IPR). IPR related gene expression is regulated by the genes pals-22 and pals-25, which also control thermotolerance and immunity against other natural pathogens. So far, we have a limited understanding of the molecular responses upon the combined exposure to heat stress and virus infection. We test the hypothesis that the response of C. elegans to OrV infection and heat stress are co-regulated and may affect each other. We conducted a combined heat-stress-virus infection assay and found that after applying heat stress, the susceptibility of C. elegans to OrV was decreased. This difference was found across different wild types of C. elegans. Transcriptome analysis revealed a list of potential candidate genes associated with heat stress and OrV infection. Subsequent mutant screens suggest that pals-22 provides a link between viral response and heat stress, leading to enhanced OrV tolerance of C. elegans after heat stress.

Porcine reproductive and respiratory syndrome virus (PRRSV) causes reproductive failure in sows and respiratory disease in piglets and growing pigs. The disease rapidly spreads in swine populations, making it a serious problem causing great financial losses to the swine industry. However, past mathematical models used to describe the spread of the disease have not yielded sufficient understanding of its spatial transmission. This work has been designed to investigate a mathematical model for the spread of PRRSV considering both time and spatial dimensions as well as the observed decline in infectiousness as time progresses. Moreover, our model incorporates into the dynamics the assumption that some members of the infected population may recover from the disease and become immune. Analytical solutions are derived by using the modified extended hyperbolic tangent method with the introduction of traveling wave coordinate. We also carry out a stability and phase analysis in order to obtain a clearer understanding of how PRRSV spreads spatially through time.

A controlled crossover experimental design was used to determine the effect of altered water sprinkling duration on heifers subjected to heat stress conditions. Heifers were subjected to 3 days of thermoneutral conditions followed by 3 days of hot conditions accompanied by water sprinkling between 1300 and 1500 h (HOT1–3). Then on the following 2 days (HOT4–5), environmental conditions remained similar, but 3 heifers were sprinkled between 1200 and 1600 h (WET) and 3 were not sprinkled (NONWET). This was followed by a 1-day period (HOT6) in which environmental conditions and sprinkling regimen were similar to HOT1–3. Rectal temperature (RT) was collected hourly, and respiration rate (RR) was monitored every 2 h on HOT Days 2, 4, 5, and 6. Dry matter intake and rate of eating were also determined. Sprinkling reduced RR and RT (P < 0.01) of all heifers during HOT1–3. During HOT4–5, WET heifers had lower (P < 0.05) RT than NONWET from 1300 to 700 h and lower RR from 1400 to 2000 h. Dry matter intake of NONWET heifers was reduced by 30.6% (P < 0.05) during HOT4–5 and by 51.2% on HOT6. On HOT4–5 the dry matter intakes of WET heifers were similar to intakes under thermoneutral conditions. During HOT6, RT was again reduced following sprinkling in all heifers. Comparison of RT and RR of NONWET and WET heifers on HOT1–3 v. HOT6 revealed that under similar environmental conditions, NONWET heifers had increased RT, partially due to carry-over from HOT4–5. However, NONWET heifers had 40% lower feed intake but tended to have lower RR on HOT6 v. HOT1–3. Only RR of WET heifers was greater on HOT6, possibly a result of switching from a 4-h back to a 2-h sprinkling period, while maintaining a 62% greater intake (5.80 v. 3.58 kg/day) than NONWET heifers during this time. Results suggest that inconsistent cooling regimens may increase the susceptibility of cattle to heat stress and elicit different physiological and metabolic responses.
