There is growing interest in the use of inulins as substrates for the selective growth of beneficial gut bacteria such as bifidobacteria and lactobacilli because recent studies have established that their prebiotic effect is linked to several health benefits. In the present study, the impact of a very-long-chain inulin (VLCI), derived from globe artichoke (Cynara scolymus), on the human intestinal microbiota compared with maltodextrin was determined. A double-blind, cross-over study was carried out in thirty-two healthy adults who were randomised into two groups and consumed 10 g/d of either VLCI or maltodextrin, for two 3-week study periods, separated by a 3-week washout period. Numbers of faecal bifidobacteria and lactobacilli were significantly higher upon VLCI ingestion compared with the placebo. Additionally, levels ofAtopobiumgroup significantly increased, whileBacteroides–Prevotellanumbers were significantly reduced. No significant changes in faecal SCFA concentrations were observed. There were no adverse gastrointestinal symptoms apart from a significant increase in mild and moderate bloating upon VLCI ingestion. These observations were also confirmed byin vitrogas production measurements. In conclusion, daily consumption of VLCI extracted from globe artichoke exerted a pronounced prebiotic effect on the human faecal microbiota composition and was well tolerated by all volunteers.

The aim of this work is to establish the most suitable proteolysis conditions to obtain bovine casein hydrolysates containing peptides with antioxidant and antihypertensive capacity. To this end, the proteolytic activity of Cynara scolymus L. flower extracts was characterized on whole bovine casein, evaluating the effect of several factors (pH, temperature, substrate concentration, enzyme concentration, and hydrolysis time). The optimal conditions to carry out the hydrolysis with the C. scolymus L. extract were as follows: pH 6.2, 50 °C, and 0.023 mg·mL−1 of extract-protein concentration. A Michaelis constant (Km) value of 5.66 mg·mL−1 and a maximum rate of reaction (Vmax) of 8.47 mUAbs∙min−1 were observed. The optimal hydrolysis time was 17 h. The casein hydrolysates obtained with these conditions contained peptides with antioxidant activity (1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging capacity: 30.89%; Trolox equivalent antioxidant capacity (TEAC) against 2,2’-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) free radical (ABTS●+): 4.43 mM Trolox equivalent·mg−1 peptide) and antihypertensive activity, showing 55.05% angiotensin-converting enzyme-I inhibition in vitro.

As indicações do uso de plantas medicinais para cura, prevenção e tratamento de doenças, desde a antiguidade, servem como importante fonte de informação para a busca de compostos com propriedades terapêuticas, a planta Cynara scolymus, conhecida como Alcachofra, apresenta-se como um vegetal de baixo valor calórico, rico em fibras e ferro, baixo teor de gordura, carboidratos, em sua maioria, formados pela inulina, é, indicado em dietas de diabéticos e pessoas com constipação intestinal, obesidade e também é recomendada como antianêmico. Diante dessas indicações terapêuticas este trabalho apresenta como objetivo a identificação do perfil fitoquímico no extrato hidroalccólico e análise toxicológica para avaliação de metemoglobina in vitro nos extratos hidroalcóolico e aquoso da alcachofra. A amostra para análise foi adquirida em um ervanário na Cidade de Belém, no estado do Pará, todas as análises foram feitas em triplicatas.Os resultados encontrados para análise fitoquímica qualitativa indicaram positivo para Alcaloides, Triterpenos, Taninos/Fenois e Saponinas, as demais identificações de metabolitos foram negativas; já  os resultados de  toxicidade de metamoglobina no extrato hidroalcolico toas as concentrações apresentaram porcentagens acima do permitido e no extrato aquoso, somente duas concentrações de 600 µg/mL  e 500 µg/mL, foram aceitáveis para o corpo humano. Conclui-se que a alcachofra apresenta atividade terapêutica, porém também apresentou um alto índice de porcentagem de toxicidade de metemoglobina.
