The design of novel nanomaterials that can be used as multifunctional platforms allowing the combination of therapies is gaining increased interest. Moreover, if this nanomaterial is intended for a targeted drug delivery, the use of several guidance methods to increase guidance efficiency is also crucial. Magnetic nanoparticles (MNPs) allow this combination of therapies and guidance strategies. In fact, MNPs can be used simultaneously as drug nanocarriers and magnetic hyperthermia agents and, moreover, they can be guided toward the target by an external magnetic field and by their functionalization with a specific probe. However, it is difficult to find a system based on MNPs that exhibits optimal conditions as a drug nanocarrier and as a magnetic hyperthermia agent. In this work, a novel nanoformulation is proposed to be used as a multifunctional platform that also allows dual complementary guidance. This nanoformulation is based on mixtures of inorganic magnetic nanoparticles (M) that have been shown to be optimal hyperthermia agents, and biomimetic magnetic nanoparticles (BM), that have been shown to be highly efficient drug nanocarriers. The presence of the magnetosome protein MamC at the surface of BM confers novel surface properties that allow for the efficient and stable functionalization of these nanoparticles without the need of further coating, with the release of the relevant molecule being pH-dependent, improved by magnetic hyperthermia. The BM are functionalized with Doxorubicin (DOXO) as a model drug and with an antibody that allows for dual guidance based on a magnetic field and on an antibody. The present study represents a proof of concept to optimize the nanoformulation composition in order to provide the best performance in terms of the magnetic hyperthermia agent and drug nanocarrier.

For decades now, conventional sinusoidal signals have been exclusively used in magnetic hyperthermia as the only alternating magnetic field waveform to excite magnetic nanoparticles. However, there are no theoretical nor experimental reasons that prevent the use of different waveforms. The only justifiable motive behind using the sinusoidal signal is its availability and the facility to produce it. Following the development of a configurable alternating magnetic field generator, we aim to study the effect of various waveforms on the heat production effectiveness of magnetic nanoparticles, seeking to prove that signals with more significant slope values, such as the trapezoidal and almost-square signals, allow the nanoparticles to reach higher efficiency in heat generation. Furthermore, we seek to point out that the nanoparticle power dissipation is dependent on the waveform’s slope and not only the frequency, magnetic field intensity and the nanoparticle size. The experimental results showed a remarkably higher heat production performance of the nanoparticles when exposed to trapezoidal and almost-square signals than conventional sinusoidal signals. We conclude that the nanoparticles respond better to the trapezoidal and almost-square signals. On the other hand, the experimental results were used to calculate the normalized power dissipation value and prove its dependency on the slope. However, adjustments are necessary to the coil before proceeding with in vitro and in vivo studies to handle the magnetic fields required.

Assemblies of magnetic nanoparticles show a great potential for application in biomedicine, particularly, magnetic hyperthermia. However, to achieve desired therapeutic effect in magnetic hyperthermia, the assembly of nanoparticles should have a sufficiently high specific absorption rate (SAR) in alternating magnetic field of moderate amplitude and frequency. Using the Landau–Lifshitz stochastic equation, it is shown that dilute assemblies of iron oxide nanoparticles of optimal diameters are capable of providing SAR of the order of 400–600[Formula: see text]W/g in alternating magnetic field with the amplitude [Formula: see text][Formula: see text]Oe in the frequency range f = 300–500[Formula: see text]kHz. Unfortunately, in dense clusters of magnetic nanoparticles, which are often formed in a biological medium, there is a sharp decrease in SAR due to the influence of strong magneto-dipole interaction of closest nanoparticles. To overcome this difficulty, it is suggested covering the nanoparticles with nonmagnetic shells of sufficient thickness or using non-single-domain nanoparticles being in magnetization curling states.

AbstractA well-established method for treating cancerous tumors is magnetic hyperthermia, which uses localized heat generated by the relaxation mechanism of magnetic nanoparticles (MNPs) in a high-frequency alternating magnetic field. In this work, we investigate the heating efficiency of cylindrical NiFe MNPs, fabricated by template-assisted pulsed electrodeposition combined with differential chemical etching. The cylindrical geometry of the MNP enables the formation of the triple vortex state, which increases the heat generation efficiency by four times. Using time-dependent calorimetric measurements, the specific absorption rate (SAR) of the MNPs was determined and compared with the numerical calculations from micromagnetic simulations and vibrating sample magnetometer measurements. The magnetization reversal of high aspect ratios MNPs showed higher remanent magnetization and low-field susceptibility leading to higher hysteresis losses, which was reflected in higher experimental and theoretical SAR values. The SAR dependence on magnetic field strength exhibited small SAR values at low magnetic fields and saturates at high magnetic fields, which is correlated to the coercive field of the MNPs and a characteristic feature of ferromagnetic MNPs. The optimization of cylindrical NiFe MNPs will play a pivotal role in producing high heating performance and biocompatible magnetic hyperthermia agents.

The activation of MNPs for hyperthermia therapy via an external alternating magnetic field is an interesting method in targeted cancer therapy. This mini-review explains new developments and implications of magnetic nanofluids mediated magnetic hyperthermia for their potential use in future clinical settings. The external alternating magnetic field generates heat in the tumor area to eliminate cancer cells. Depending on the tumor type and targeted area, several kinds of MNPs with different coating agents of various morphology and surface charge have been developed. The tunable physiochemical characteristics of MNPs enhance their heating capability. In addition, heating efficiency is strongly associated with the amount of the applied magnetic field and frequency. The great efforts have offered promising preclinical trials of magnetic hyperthermia via MNPs as a smart nanoagent. MNPs are very appropriate to be considered as a heating source in MHT and prospective research in this field will lead to tackle the problems from chemotherapy and introduce promising therapeutic techniques and nanodrug formulations for remotely controlled drug release and anticancer effects. This mini-review aims to pinpoint synthesis and structural analysis of various magnetic nanoparticles examined for magnetic hyperthermia therapy and controlled drug release in cancer treatment.

The cancer therapy with the lowest possible toxicity is today an issue that raises major difficulties in treating malignant tumors because chemo- and radiotherapy currently used in this field have a high degree of toxicity and in many cases are ineffective. Therefore, alternative solutions are rapidly being sought in cancer therapy, in order to increase efficacy and a reduce or even eliminate toxicity to the body. One of the alternative methods that researchers believe may be the method of the future in cancer therapy is superparamagnetic hyperthermia (SPMHT), because it can be effective in completely destroying tumors while maintaining low toxicity or even without toxicity on the healthy tissues. Superparamagnetic hyperthermia uses the natural thermal effect in the destruction of cancer cells, obtained as a result of the phenomenon of superparamagnetic relaxation of the magnetic nanoparticles (SPMNPs) introduced into the tumor; SPMNPs can heat the cancer cells to 42–43 °C under the action of an external alternating magnetic field with frequency in the range of hundreds of kHz. However, the effectiveness of this alternative method depends very much on finding the optimal conditions in which this method must be applied during the treatment of cancer. In addition to the type of magnetic nanoparticles and the biocompatibility with the biological tissue or nanoparticles biofunctionalization that must be appropriate for the intended purpose a key parameter is the size of the nanoparticles. Also, establishing the appropriate parameters for the external alternating magnetic field (AMF), respectively the amplitude and frequency of the magnetic field are very important in the efficiency and effectiveness of the magnetic hyperthermia method. This paper presents a 3D computational study on specific loss power (Ps) and heating temperature (ΔT) which allows establishing the optimal conditions that lead to efficient heating of Fe3O4 nanoparticles, which were found to be the most suitable for use in superparamagnetic hyperthermia (SPMHT), as a non-invasive and alternative technique to chemo- and radiotherapy. The size (diameter) of the nanoparticles (D), the amplitude of the magnetic field (H) and the frequency (f) of AMF were established in order to obtain maximum efficiency in SPMHT and rapid heating of magnetic nanoparticles at the required temperature of 42–43 °C for irreversible destruction of tumors, without affecting healthy tissues. Also, an analysis on the amplitude of the AMF is presented, and how its amplitude influences the power loss and, implicitly, the heating temperature, observables necessary in SPMHT for the efficient destruction of tumor cells. Following our 3D study, we found for Fe3O4 nanoparticles the optimal diameter of ~16 nm, the optimal range for the amplitude of the magnetic field of 10–25 kA/m and the optimal frequency within the biologically permissible limit in the range of 200–500 kHz. Under the optimal conditions determined for the nanoparticle diameter of 16.3 nm, the magnetic field of 15 kA/m and the frequency of 334 kHz, the magnetite nanoparticles can be quickly heated to obtain the maximum hyperthermic effect on the tumor cells: in only 4.1–4.3 s the temperature reaches 42–43 °C, required in magnetic hyperthermia, with major benefits in practical application in vitro and in vivo, and later in clinical trials.

The activation of magnetic nanoparticles in hyperthermia treatment by an external alternating magnetic field is a promising technique for targeted cancer therapy. The external alternating magnetic field generates heat in the tumor area, which is utilized to kill cancerous cells. Depending on the tumor type and site to be targeted, various types of magnetic nanoparticles, with variable coating materials of different shape and surface charge, have been developed. The tunable physical and chemical properties of magnetic nanoparticles enhance their heating efficiency. Moreover, heating efficiency is directly related with the product values of the applied magnetic field and frequency. Protein corona formation is another important parameter affecting the heating efficiency of MNPs in magnetic hyperthermia. This review provides the basics of magnetic hyperthermia, mechanisms of heat losses, thermal doses for hyperthermia therapy, and strategies to improve heating efficiency. The purpose of this review is to build a bridge between the synthesis/coating of magnetic nanoparticles and their practical application in magnetic hyperthermia.
