The new complex [Rh 2 (phen) 2 (CH 3 CN) 6 ](BF 4 ) 4 ( 1 ) was synthesized and characterized in solution and its crystal structure was determined. Irradiation of 1 with visible light ( λ irr >590 nm) in water results in the release of two equatorial CH 3 CN ligands, CH 3 CN eq , as well as in the formation of mononuclear radical Rh(II) fragments stemming from the homolytic photocleavage of the metal–metal bond. The photoproducts, identified by electrospray ionization mass spectrometry, include [Rh(phen)(CH 3 CN)(OH)] + and [Rh(phen)(CH 3 CN)(H 2 O) 3 (BF 4 )] + . The quantum yield for the photochemical transformation of 1 in H 2 O exceeds unity ( Φ 550 nm =1.38) indicative of dark reactions following the initial photoprocess. DNA photocleavage was observed for 1 ( λ irr >590 nm), whereas the complex is unreactive in the dark. This feature makes 1 a promising photodynamic therapy agent that does not operate via the production of singlet oxygen, 1 O 2 .

AbstractPhotodynamic therapy (PDT) is a potential option for treatment of cancer since it can be performed non- invasive for superficial cancers or minimal-invasive with low traumatization. But PDT is intrinsically inefficient due to the complex accumulation of the photosensitizing drug inside the tumor and the processes of heme syntheses to create the needed cell killing components. To optimize the outcome of PDT and increase acceptance as viable option it is necessary to predict the optimal time for the start of the treatment based on measurable precursors. A former cell study proposed a new filter fluorometer in a complex and sensitive setup. In this work we now designed and tested a simplified system that can be used in combination with standard endoscopic imaging systems. This system will be used as base to prove viability of this approach for a future clinical study.

With 4′-amino-N,N-diethylaniline and aniline as starting materials, methylene violet 3RAX 1 and its derivatives 2–5 were synthesised. The five compounds were characterised by IR, UV-vis, and 1H NMR spectroscopy and mass spectrometry. The binding mode between the synthesised compounds and DNA were investigated. The results show that both compounds 1 and 5 bind to DNA by an intercalative mode, while compounds 2–4 interact with DNA through a mixed binding mode involving groove binding and electrostatic interactions. The photocleavage ability of the five compounds to DNA were calculated to be 38, 40, 30, 20, and 13 %, respectively, when their concentration was adjusted to 400 μM. The singlet oxygen production of compounds measured by the 1,3-diphenylisobenzofuran method was consistent with the trend of DNA photocleavage ability. The DNA studies suggest that the binding mode between methylene violet 3RAX and DNA, the ability of methylene violet 3RAX to generate singlet oxygen, and the DNA photocleavage activity could be adjusted through modification of the amino group on methylene violet 3RAX.

We present a new family of porphyrin-functionalized coordination star polymers prepared through combination of coordination-driven self-assembly and post-assembly polymerization. Their self-assembly behaviour in water and potential for photodynamic therapy were demonstrated.
