Potential Applications

Pathologic posterior neovascularization of eye is a major cause of irreversible vision loss and limitations of therapeutics to be successfully delivered to back of the eye has been a main obstacle for its effective treatment. Current pharmacological treatment using anti-VEGF agents being delivered intravitreally are effective but complicated due to anatomical and physiological barriers, as well as administration of high and frequent doses. With expanding horizons of nanotechnology, it can be possible to formulate promising nanoscale delivery system to improve penetration and sustained the release of therapeutic in posterior segment of the eye. Taking into consideration advances in the field of nanoscale delivery systems, this special report focuses on emerging strategies and their applications for treatment of posterior ocular neovascularization.

A commercial controlled delayed cooling or preconditioning treatment was developed to extend peach (Prunus persica) market life of the most popular California peach cultivars. A 24 to 48 h cooling delay at 68 °F (20.0 °C) was the most effective treatment for extending market life of internal breakdown susceptible peaches without causing fruit deterioration. This treatment increased minimum market life by up to 2 weeks in the cultivars tested. Weight loss and softening occurred during the controlled delayed cooling treatments, but did not reduce fruit quality. Detailed monitoring of these fruit quality changes during the delayed cooling period and proper use of fungicides is highly recommended for success in this new fruit delivery system. Rapid cooling after preconditioning is important to stop further fruit deterioration such as flesh softening, senescence, decay and weight loss. Controlled delayed cooling can also be used to pre-ripen susceptible and nonsusceptible peaches to deliver a ready-to-buy product to the consumer.

AbstractThe current standard of care for posterior segment eye diseases, such as age-related macular degeneration and diabetic macular edema, is frequent intravitreal injections or sustained-release drug implants. Drug implants have side effects due to the burst release of the drugs, and their release cannot be easily controlled after implantation. Present study attempts to develop a dosage-controllable drug delivery implant which consists of a nanoporous biodegradable PLGA capsule and light-activated liposomes. Controllable drug release from the implant was achieved by using pulsed near-infrared (NIR) laser both in vitro and in vivo. The in vitro drug release kinetics from two different initial dose implants, 1000 μg and 500 μg, was analyzed by fitting zero order and first order kinetics, as well as the Korsmeyer-Peppas and Higuchi models. The 1000 μg and 500 μg implants fit the first-order and zero-order kinetics model, respectively, the best. The multiple drug releases in the vitreous was determined by in vivo fluorimeter, which was consistent with the in vitro data. The dose released was also clinically relevant. Histology and optical and ultrasound imaging data showed no abnormality in the eyes received implant treatment suggesting that the drug delivery system was safe to the retina. This on-demand dose-controllable drug delivery system could be potentially used for long-term posterior eye disease treatment.
