Ocimum basilicum L. (sweet basil) is an economically and ethnobotanically important aromatic, medicinal, ornamental and culinary herb, with a very wide gene pool, that is sensitive to cold and prone to several plant pathogens that can demolish harvest and lessen yield. In this research, the effects of BAP (6-Benzylaminopurine) and TDZ (Thidiazuron) on different genotypes for in vitro cloning were determined, in order to provide a detailed protocol guide concerning Ocimum basilicum L. propagation. The results from the O. basilicum seed propagations revealed that the best condition for the secondary shoot growth is with 5.0 mg/l TDZ or 1.5 mg/l BAP on all types of explants except the root, the secondary root growth can be obtained on all types explant with any BAP concentration and all cytokinins can induce callus on all types of explants. On the whole, it shows that multiple secondary shoot induction and regeneration in Ocimum basilicum L. is regulated by appropriate cytokinin concentration.

An efficient regeneration/transformation system was developed for `Beurre Bosc' pear. Young leaves were harvested from in vitro shoots proliferated on a medium containing MS basal salts and 5 BAP, 0.5 μM IBA, and 0.6M3. Shoot regeneration was optimized using a modification of the medium of Chevreau and Leblay (1993). Explants were cultured on shoot induction medium contained 10 μM TDZ and 1 μM IBA for 4 weeks in the dark, and then transfered to a similar, but auxinless, regeneration medium until shoots developed, usually after an additional 4 to 8 weeks. Leaf tissues were transformed by co-cultivation for 3 days with Agrobacterium tumefaciens EHA101 carrying a pGA482 plasmid containing NPTII, GUS, and rolC genes, followed by cultivation on SIM containing 300 mg/L timentin. Putative transgenic plants were selected on shoot induction medium containing 80mg/L kanamycin, and multiplied on shoot proliferation medium. Four clones were confirmed as transgenic using the GUS histochemical assay and Southern blots for the NPTII and rolC genes. Plants of each clone have been rooted and successfully transfered to the greenhouse for further analysis of gene expression.

An efficient protocol for plant regeneration through multiple shoots induction from shoot tips of Polygonum hydropiper (L.) was established. The highest percentage (96.6) of multiple shoot induction and number of shoots (9.0) per culture were found on MS supplemented with 2.0 mg/l Kn. The induced shoots were excised and inoculated on to MS contains different concentrations of IBA or NAA for rooting. The highest percentage (90.0) of root induction and the highest number of roots per shoot (12.0) was found on MS having 1.0 mg/l IBA. Shoot tip explants from in vitro germinated seedlings of Solanum capsicoides All. inoculated on MS containing 2 mg/l BA produced maximum shoot induction response (26 shoots per explant). Rooting of the microshoots (19.4 roots per explant) was obtained better in half strength of MS supplemented with NAA (0.5 mg/l). An experiment was conducted on in vitro regeneration and multiplication of Dendrobium bensoniae. Different concentrations of BA and IBA alone or combination of both hormones were used as treatment for regeneration.  It was revealed that shoot regeneration from node was the best at 2.0 mg/l BA supplemented to MS medium. It gave better responses than all other concentrations and combinations of BA and BA+IBA, used in the present study. The highest number of shoots and leaves were found when 1.0 mg/l BA with 1.5 mg/l IBA was supplemented into MS medium.  For rooting, 0.5 mg/l BA with 1.0 mg/l IBA was found to be the most effective. The well-rooted plantlets were successfully acclimatized under 70-80% humidity and planted in pots and transferred to the shade house for establishment. Around 85% of plantlets survived in the field. From the present result, it may be recommended that MS medium supplemented with 2.0 mg/l BA may be used for rapid shoot induction and regeneration of D. bensoniae.The Agriculturists 2016; 14(2) 24-31

An efficient protocol was developed for shoot proliferation and plant regeneration of Phlogacanthus thyrsiflorus Nees. (Acanthaceae) - a rare medicinal shrub of Bangladesh, through in vitro culture using shoot tip and nodal explants. Best shoot induction was observed on MS with 1.0 mg/l BAP + 0.5 mg/l NAA, in which 84.2% of nodal explants responded to produce maximum number (12.4 ± 0.66) of shoots per culture. In vitro raised shoots rooted on half-strength MS with 0.5 mg/l IBA + 0.5 mg/l NAA. For acclimation and transplantation, the plantlets in the rooting culture tubes were kept in normal room temperature for 7 days before transplanting in pots where plantlets were reared for three weeks. This study investigated in vitro production of diploid (AS2) and tetraploid (AS4) cytotypes of snowdrop anemone. The effect of plant growth regulators (PGRs) on in vitro shoot multiplication and rooting was investigated. The effect of activated charcoal (AC) on root induction was also studied. Ploidy level affected growth characteristics during multiplication and rooting. Shoot induction in AS4 was higher on medium supplemented with cytokinin (3.2–3.6), while the AS2 clone formed the most shoots on PGR-free medium (3.6). The highest rooting percentage was achieved on PGR-free medium in both genotypes (AS2 clone, 100% and AS4 clone, 93.3%). The addition of AC to the PGR media largely increased root induction and root length. Rooted plantlets were successfully acclimatised in the greenhouse with 100% survival. Thus, the described micropropagation protocol represents a rapid and effective in vitro propagation method for utilisation in horticulture and conservation programmes of snowdrop anemone.

After inoculation of young leaves of date palm offshoot required about six months to complete the morphogenesis process. Fourteen weeks were required for embryogenic callus formation under continuous dark condition and nine weeks for shoot initiation (under 16/8 h light/dark). The highest number of explants (80%) produced callus in modified MS containing 5 mg/l 2,4-D + 2 mg/l 2ip. Sixty per cent of explants produced callus in the modified medium containing 5 mg/l 2,4-D + 5 mg/l NAA. while only 50 per cent of the explants formed callus in the same medium when supplemented with only 5 mg/l 2,4-D. The induced calli were transferred to modified MS for shoot proliferation. A combination of two cytokines showed better performance than single ones in shoot induction. The highest percentage (70) of shoot developed in modified MS containing 2 mg/l BAP + 1 mg/l Kn. Forty per cent shoot induction was found in the same medium supplemented with 2 mg/l of BAP. Thirty per cent shoot formed in MS containing 1 mg/l of Kn. The shoots were subcultured at three- four week intervals throughout culture duration. D. O.
