Effect of aqueous extract of Ajuga iva supplementation on plasma lipid profile and tissue antioxidant status in rats fed a high-cholesterol diet

Aim: A high cholesterol diet (HCD) is known to cause metabolic dysregulation, oxidative stress, cardiovascular diseases and atherogenesis. Zingerone is a pharmacologically active component of dry ginger. Zingerone has been shown to have a wide range of pharmacological properties, including scavenging free radicals, high antioxidant activity, suppressing lipid peroxidation and anti-inflammatory. This study aimed to investigate the effects of Zingerone on HCD-induced atherosclerosis in rats. Methods: Animals were divided into four categories (n = 6). Group I: normal control, Group II: zingerone control (20 mg/kg b.wt.), group III: HCD-induced atherosclerosis, Group IV: HCD + zingerone, respectively, for 8 weeks. Results: The HCD-fed rats resulted in a significant increase in an atherosclerotic lesion, lipid peroxidation, lipid profile, high-density lipoprotein concentration, cardiac markers, body weight, reduced antioxidant status, and displayed atherosclerosis. These findings were conventional by up-regulated expression of lipid regulatory genes like sterol-regulatory-element-binding protein-c (SREBP-c), fatty acid synthase (FAS), acetyl-CoA carboxylase (ACC), acetyl-CoA synthetase (ACS), liver X receptor–alpha (LXR-α), and down-regulated expression of acetyl-CoA oxidase (ACO), peroxisome proliferator-activated receptor-alpha (PPAR-α) and carnitine palmitoyl transferase-1 (CPT-1) in HCD-fed rats. These significant changes were observed in the zingerone-treated rats for the last 4 weeks. Conclusion: These findings suggest that zingerone reduced atherosclerosis by modulated the atherosclerotic lesion, lipid profile, antioxidant status and lipid regulatory gene expression in HCD-fed rats.

1. The effects of feeding isolated saponins on plasma lipid concentrations and on concentrations of biliary and faecal bile acids and neutral sterols were studied in the rat.2. The animals were given one of four diets, i.e. a standard low-cholesterol synthetic diet, the diet+10 g saponins/kg, the diet+10 g cholesterol/kg, the diet+10 g cholesterol+10 g saponins/kg.3. Saponins partially reversed the hypercholesterolaemia caused by the high-cholesterol diet and increased both the rate of bile acid secretion and the faecal excretion of bile acids and neutral sterols. The proportionate contribution of the primary bile acids (particularly chenodeoxycholic) to faecal excretion was also increased by saponins.4. The results are discussed in relation to the hypothesis that saponins act by inducing the adsorption of bile acids by dietary fibre.

Purpose This study aims to investigate the possible effect of Citrus latifolia (CL) extract on biomarkers of oxidative stress, including lipid peroxidation products in rats fed a high cholesterol diet Design/methodology/approach Hypercholesterolemia was induced by feeding normocholesterolemic rats 1 per cent cholesterol-enriched diet for 15 days. An experimental group (n = 20) was divided into two groups (n = 10) and fed the same diet with or without CL lyophilized aqueous extract (1 per cent) for four weeks. At day 28, ten rats from each group were killed. Findings Treatment with CL lyophilized aqueous extract compared with the untreated group had decreased plasma total cholesterol (TC) (−36 per cent), triacylglycerols (−48 per cent), isoprostanes values (−74 per cent) and reduced thiobarbituric acid reactive substances in erythrocytes (−21 per cent). However, the supplementation of CL peels in the hypercholesterolemic diet enhanced superoxide dismutase (+69 per cent), glutathione reductase (+30 per cent) and catalase activities (+34 per cent). Originality/value In hypercholesterolemic rats, administering CL extract ameliorates dyslipidemia and attenuates lipid peroxidation in tissues. These results suggest that CL could be beneficial in the primary treatment of hypercholesterolemia and oxidative damage caused by a high-cholesterol diet.
