Ovicidal and toxic effects of crude leaf extracts of Clerodendrum inerme, Clerodendrum splendens, Clerodendrum multiflorum, Vitex negundo and Argemone mexicana were evaluated against the castor semilooper, Achaea janata L. (Noctuidae: Lepidoptera) using different bioassay methods. Among the extracts tested, the highest ovicidal activity was observed in C. splendens as well as A. mexicana with LD50 values of 7.65 and 9.14 mg-1, respectively. Leaf extracts of A. mexicana and C. inerme were toxic to both third and fourth instar larvae of A. janata through topical application. However, the extracts of A. mexicana and C. inerme found to be more toxic to third instar larvae with 70 and 73 per cent mortality, respectively. The lowest LD50 value of 5.33 mg-1 was recorded by A. mexicana extract and was followed by C. inerme extract with LD50 value of 7.26 mg-1. Present results indicated that A. mexicana and C. inerme plants have potential to use in IPM programme.

Serious malnutrition problems occur in developing countries where people’s diets are mainly based on staple crops. To alleviate this, high-production crops are being developed that are better adapted to climate change, enriched in micronutrients and vitamins, or resistant to pests. In some cases, new varieties have been developed with several of the characteristics mentioned above, such as biofortified and pest-resistant crops. The development of biofortified Bacillus thuringiensis (Bt) crops raises the question of whether vitamin enrichment of Bt crops can in any way favor those pests that are not very susceptible to the Bt toxin that feed on these crops, such as Helicoverpa armigera (Hübner) or Mythimna unipuncta (Haworth) (Lepidoptera: Noctuidae). In this study, the response to a Bt diet enriched with vitamins A (β-carotene) and C (ascorbic acid) was somewhat different between the two species. M. unipuncta was less sensitive to the toxin than H. armigera, although the ingestion of the Bt diet resulted in oxidative stress (longer larval development and lower pupal weight) which was not mitigated by the vitamins. However, the two vitamins reduced the mortality of H. armigera larvae fed on a Bt-enriched diet; in addition, ß-carotene reduced the activity of the antioxidant glutathione S-transferase (GST) of both species, suggesting it has an antioxidant role. The results obtained here indicate that biofortified Bt crops will not favor the development of H. armigera very much and will not affect M. unipuncta’s development at all, although the effect of the increase in vitamins may be very variable and should be studied for each specific phytophagous.

The present paper reports the occurrence of insects damaging Eucalyptus in nursery stage. Out of nine insects recorded, seven species observed as occasional defoliating species, viz. Achaea janata (Linnaeus) (Lepidoptera : Noctuidae), Catapsilia crocale Cramer (Lepidoptera : Pieridae), Dasychira grotei Moore (Lepidoptera : Lymantriidae), Eurema hecabe Linnaeus (Lepidoptera : Pieridae), Hyposidra talaca (Walker) (Lepidoptera : Geometridae), Lymantria sobrina Moore (Lepidoptera : Lymantriidae) and Spodoptera litura (Fabricius) (Lepidoptera : Noctuidae), one rare sap-sucking species, Chrysocoris purpureus Westwood (Heteroptera : Pentatomodae) and one gall making species, Leptocybe invasa Fisher & LaSalle (Hymenoptera : Eulophidae). Among the nine species collected, six defoliators and one sap sucker are new addition to the Eucalyptus entomology. Further, H. talaca is the first record of occurrence on Eucalyptus in India. On the basis of percentage infestation/damage caused by above insects, L. invasa, commonly known as blue gum chalcid wasp, was recorded only as a key insect pest in nursery stage of Eucalyptus. The results have been discussed in the light of the present observations.

AbstractMidgut of lepidopteran larvae is a multifunctional tissue, which performs roles in digestion, absorption, immunity; transmission of pathogens and interaction with ingested various molecules. The proteins localized at the inner apical brush border membrane are primarily digestive proteases but some of them like aminopeptidase N, alkaline phosphatase, cadherins, ABC transporter C2 etc. interact with Crystal (Cry) toxins produced by Bacillus thuringiensis (Bt). In the present study aminopeptidase N (APN) was characterized as Cry toxin interacting protein in larval midgut of castor semilooper, Achaea janata. Transcriptomic and proteomic analyses revealed the presence of multiple isoforms of APNs (APN1, 2, 4, 6 and 9) which have less than 40% sequence similarity but show the presence of characteristic “GAMENEG” and zinc-binding motifs. Feeding of sublethal dose of Cry toxin caused differential expression of various APN isoform. Further, 6th generation Cry toxin exposed larvae showed reduced expression of APN2. This report suggests that A. janata larvae exploit altered expression of APNs to overcome the deleterious effects of Cry toxicity, which might facilitate toxin tolerance in long run.

Efficacy of the combined treatment of a neem oil formulation and endosulfan on feeding and midgut enzyme activities of Helicoverpa armigera larvae was studied. The antifeedant activity was recorded at 24 h after treatment and the activities of midgut digestive (total serine protease and trypsin) and detoxifying (esterase and glutathione-S-transferase) enzymes were estimated at 72 h after treatment. The antifeedant activity in endosulfan + neem oil formulation (endosulfan 0.01% and neem oil formulation 1% at 1:1 ratio) was 85.34%, significantly greater than in individual treatments. Midgut digestive enzymes and EST activities were significantly reduced and the GST activity significantly increased in the combined treatment of endosulfan + neem oil formulation, thus showing increased effect of the combined treatment of the two pesticides. These results suggest that neem oil can be used in combination with endosulfan to reduce its quantity.
