The even-numbered C2–C22saturated and the C18-unsaturated fatty acid esters of beta- and gamma-sitosterol, stiginasterol, ergosterol, stigmastanol, and gamma-sitostanol were used separately and in model mixtures for the evaluation of existing and the development of new paper chromatographic methods for the separation and identification of these compounds. Satisfactory separations of the esters were obtained by the ascending reversed phase chromatography on cellulose or glass fiber paper using one of the following systems depending upon the number of carbon atoms in the fatty acid moiety of the ester: C2–C12, aqueous acetic acid/chloroform/paraffin oil; C8–C18, aqueous propionic acid/paraffin oil; C8–C22, acidified aqueous methanol/chloroform/paraffin oil or aqueous butyric acid/paraffin oil. The rate of migration of the esters was inversely proportional to the molecular weight and was increased by the introduction of unsaturation.The increased movement due to unsaturation has been correlated with the number of ethylenic hydrogens, and the possibility that polarity of these atoms might account for this phenomenon is discussed. The reversed phase paper partition chromatography of sterols as their esters is suggested as a potential tool for evaluation of polarity of the various attributes of the sterol nucleus in terms of the chain length of the ester fatty acid. The rule of independent contributions of the constituents of a molecule to its distribution isotherm was observed not to hold in this fatty ester series.

Depending on conditions in the refining process, soybean oil deodorizer distillate (SODD) contains 42-51% free fatty acids (FFA) and 16-25% triacylglycerol (TAG). Bioactive compounds such as tocopherols, free phytosterols, fatty acid steryl esters (FASE) and squalene also make up a significant portion of SODD. The efficient removal of FFA and TAG is a crucial step in the isolation and purification of bioactive compounds from SODD. In this work, a modified soxhlet extraction technique was developed and its optimal operation conditions were determined for the efficient removal of FFA and TAG from SODD. Starting with SODD that contains 3.53% FASE and 1.99% squalene, it was possible to obtain a fraction enriched with FASE (22.0%, recovery 91.3%) and squalene (8.63%, recovery 100%) by this modified soxhlet extraction under the following operation conditions: SODD/(silica gel) = 1/3 (w/w), extraction temperature = -6°C, number of extraction/h = 8.7. FFA remained in this FASE and squalene enriched fraction is 38.7% and there was no detectable TAG. The percentage removal of FFA and TAG for this FASE and squalene enriched fraction are 84.29% and 100%, respectively. The advantages of modified soxhlet extraction over open-column chromatography are that less solvent usage, larger sample size per batch and shorter operation time.Keywords: Fatty acid steryl ester; free fatty acid; modified soxhlet extraction; soybean oil deodorizer distillate; squalene; triacylglycerol AbstrakDalam proses pemurnian minyak kedelai, produk samping dari proses penghilangan bau (soybean oil deodorizer distillate, SODD) mengandung 42-52% free fatty acids (FFA) dan 18-28% triacylglycerols (TAG). Komponen bioaktif seperti tocopherols, free phytosterols, fatty acid steryl esters (FASE) dan squalene juga mempunyai kontribusi yang besar dalam komposisi SODD. Efisiensi penghilangan FFA dan TAG adalah langkah yang sangat penting dalam pemisahan dan pemurnian komponen bioaktif dari SODD. Dalam penelitian ini, modifikasi soxhlet ekstraksi telah ditemukan dan optimisasi kondisi operasi ditentukan berdasarkan efisiensi penghilangan FFA dan TAG dari SODD. Fraksi lemak non polar (nonpolar lipid fraction, NPLF) dengan kandungan FASE (22.0%, recovery 91.3%) dan squalene (8.63%, recovery 100%) telah diperoleh dengan menggunakan modifikasi soxhlet ektraksi dengan kondisi operasi:  SODD/(silica gel) = 1/3 (w/w), dan suhu ekstraksi = -6°C. FFA dan TAG yang tersisa di NPLF secara berurutan adalah 38.7% dan 0%. Persentase penghilangan FFA dan TAG di NPLF secara berurutan adalah 84.29% and 100%. Keuntungan menggunakan modifikasi soxhlet ekstraksi dibandingkan dengan silica gel column chromatography adalah sedikitnya pelarut yang digunakan, besarnya sampel per unit batch yang digunakan, dan pendeknya waktu operasi.Kata kunci: Free fatty acid, fatty acid steryl ester, modifikasi soxhlet ekstraksi, soybean oil deodorizer distillate, squalene, triacylglycerols

California-grown `Hass' avocado fruit were stored at 5C, in air or a controlled atmosphere (CA) of 2% oxygen and 5% carbon dioxide. Fruit were evaluated at 0, 2, 4, 6, 8, 10, and 12 weeks, both immediately upon removal from storage and after ripening at 20C. Severe chilling injury (flesh browning) developed in the airstored fruit after 6 weeks, while only moderate symptoms were observed in CA-stored avocado fruit after 12 weeks. Lipid peroxidation breakdown products increased during storage and ripening in both air and CA treatments. Sterols, steryl esters, steryl glycosides, glycolipids, and phospholipids were analyzed. Quantity of acylated steryl glycoside in ripe fruit changed from 34 nmoles initially, to 51 or 27 nmoles after 6 weeks at 5C in air or CA, respectively. Glycolipid fatty acid unsaturation in air-stored fruit decreased with the development of chilling injury. Fatty acid unsaturation in phospholipids (phosphatidylinositol, phosphatidylcholine, phosphatidylglycerol, and phosphatidylethanolamine) of air-stored avocados decreased with the development of chilling injury. CA storage delayed the development of chilling injury and the loss of fatty acid unsaturation.

Background/Aim: Sweet almond (Prunus amygdalus dulcis) oil is one of the most famous cold-pressed seed oils. However, other species of Prunus can provide oils with healthy properties. We analyzed the fatty acid (FA) composition, as well as the antioxidant, the in vitro anti-inflammatory properties, and the antibiofilm activity of five commercial vegetable cold-pressed seed oils of apricot, peach, plum, cherry, and black cherry.Methods: Gas Chromatography-Mass Spectrometry was performed for the analysis of FAs The antioxidant property of the oils was carried using different tests [2, 2-diphenyl-1-picrylhydrazyl (DPPH assay)], Ferric Reducing Antioxidant Power (FRAP), and the 2, 20 -azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS·+). The denaturation assay performed on bovine serum albumin (BSA) was used to evaluate the in vitro anti-inflammatory activity. The anti-biofilm activity was assessed using five pathogenic strains, namely, Acinetobacter baumannii, Escherichia coli, Listeria monocytogenes, Pseudomonas aeruginosa, and Staphylococcus aureus, through the crystal violet test and the 3-(4,5-dimethylthiazol-2-yl)−2,5-diphenyltetrazolium bromide (MTT), used to evaluate the metabolism of the microbial cells present within the biofilm.Results: Oleic acid and linoleic acids were the most abundant FAs. Black cherry seed oil exhibited the best antioxidant activity, but in general, the amount of oil needed to inhibit the activity of 1 ml of DPPH assay at 50% did not exceed 10 μg. The extract concentration for the 50% inhibition of the denaturation of the protein (IC50) did not exceed 4.4 μg. Linoleic and stearic acids affected the antioxidant activity of the oils; oleic acid, linolenic, and palmitoleic acids exhibited beneficial effects in preserving the BSA denaturation, as shown by the correlation data. The oils were able to inhibit the biofilm formation of the pathogens (up to 71.40% of inhibition) as well as act against their mature biofilm, although with different strengths, with values up to 61.54%. Concurrently, they also acted on the pathogen metabolism.Conclusion: The oils represent a valuable source of some healthy FAs. They showed potential antioxidant and anti-inflammatory in vitro activity, in addition, their potential effect on the biofilm can offer important ideas for research and reflection on their use as functional foods and/or ingredients.
